BS-dqPCR streamlines miRNA quantification by dispensing with ligation and reverse transcription procedures, thereby minimizing both time and expense when contrasted with traditional miRNA PCR methods. MiRNA samples, when subjected to standard PCR thermocycling conditions, produced digital signals that were lower than expected, while blanks showed higher than expected signals. Thus, a new asymmetric thermocycling regime was developed, ensuring the preferential amplification of the miRNA target while minimizing any non-specific amplification reactions. The BS-dqPCR analytical response was then examined for its performance over different miRNA concentration levels.
