Dynamical processes induced by the external time-dependent fields can provide valuable insight into the characteristic energy scales of a given physical system. We investigate them here in a nanoscopic heterostructure, consisting of the double quantum dot coupled in series to the superconducting and the metallic reservoirs, analyzing its response to (i) abrupt bias voltage applied across the junction, (ii) sudden change of the energy levels, and imposed by (iii) their periodic driving. We explore subgap properties of this setup which are strictly related to the in-gap quasiparticles and discuss their signatures manifested in the time-dependent charge currents. The characteristic multi-mode oscillations, their beating patters and photon-assisted harmonics reveal a rich spectrum of dynamical features that might be important for designing the superconducting qubits.Beat-to-beat R-R intervals (RRI) and pulse arrival time (PAT) provide pivotal information to evaluate cardiac autonomic functions for predicting arrhythmias and cardiovascular morbidity. However, their relationship has not been clearly understood. In this study, we simultaneously recorded electrocardiograms and photoplethysmograms on 34 subjects in the natural state, and on 55 subjects under the cold stimulation. The RRI and the PAT were calculated and then analyzed using Pearson correlation coefficient. The results showed that the RRI and the PAT were strongly correlated (r = 0.562) and the RRI series were 2.18 ± 0.40 beats advanced to the PAT series. After smoothing, the RRI and the PAT were more correlated in the low frequency than in the high frequency. Furthermore, when involving RRI with the phase effect, the proposed PAT based model showed better performance for blood pressure estimation. We think these results are helpful to understand the underlying regulatory mechanisms of the two cardiovascular factors, and would provide useful suggestions for non-invasive cuffless blood pressure estimation.We theoretically investigate a new stabilization mechanism of a skyrmion crystal (SkX) in centrosymmetric itinerant magnets with magnetic anisotropy. MEK activation By considering a trigonal crystal system without the horizontal mirror plane, we derive an effective spin model with an anisotropic Ruderman-Kittel-Kasuya-Yosida (RKKY) interaction for a multi-band periodic Anderson model. We find that the anisotropic RKKY interaction gives rise to two distinct SkXs with different skyrmion numbers of one and two depending on a magnetic field. We also clarify that a phase arising from the multiple-Q spin density waves becomes a control parameter for a field-induced topological phase transition between the SkXs. The mechanism will be useful not only for understanding the SkXs, such as that in Gd[Formula see text]PdSi[Formula see text], but also for exploring further skyrmion-hosting materials in trigonal itinerant magnets.Acid ceramidase (AC) is a lysosomal hydrolase encoded by the ASAH1 gene, which cleaves ceramides into sphingosine and fatty acid. AC is expressed at high levels in most human melanoma cell lines and may confer resistance against chemotherapeutic agents. One such agent, doxorubicin, was shown to increase ceramide levels in melanoma cells. Ceramides contribute to the regulation of autophagy and apoptosis. Here we investigated the impact of AC ablation via CRISPR-Cas9 gene editing on the response of A375 melanoma cells to doxorubicin. We found that doxorubicin activates the autophagic response in wild-type A375 cells, which effectively resist apoptotic cell death. In striking contrast, doxorubicin fails to stimulate autophagy in A375 AC-null cells, which rapidly undergo apoptosis when exposed to the drug. The present work highlights changes that affect melanoma cells during incubation with doxorubicin, in A375 melanoma cells lacking AC. We found that the remarkable reduction in recovery rate after doxorubicin treatment is strictly associated with the impairment of autophagy, that forces the AC-inhibited cells into apoptotic path.In eukaryotes the entry into mitosis is initiated by activation of cyclin-dependent kinases (CDKs), which in turn activate a large number of protein kinases to induce all mitotic processes. The general view is that kinases are active in mitosis and phosphatases turn them off in interphase. Kinases activate each other by cross- and self-phosphorylation, while phosphatases remove these phosphate groups to inactivate kinases. Crucial exceptions to this general rule are the interphase kinase Wee1 and the mitotic phosphatase Cdc25. Together they directly control CDK in an opposite way of the general rule of mitotic phosphorylation and interphase dephosphorylation. Here we investigate why this opposite system emerged and got fixed in almost all eukaryotes. Our results show that this reversed action of a kinase-phosphatase pair, Wee1 and Cdc25, on CDK is particularly suited to establish a stable G2 phase and to add checkpoints to the cell cycle. We show that all these regulators appeared together in LECA (Last Eukaryote Common Ancestor) and co-evolved in eukaryotes, suggesting that this twist in kinase-phosphatase regulation was a crucial step happening at the emergence of eukaryotes.In this study, the minimal residual disease (MRD) levels in patients with multiple myeloma (MM) were assessed by comparing the new 8-color single-tube multiparameter flow cytometry method (DuraClone), which reduces the cost of antibodies and labor burden of laboratories, with the EuroFlow next-generation flow (NGF) method. A total of 96 samples derived from 69 patients with MM were assessed to determine the total cell acquisition number (tCAN), percentages of total and normal plasma cells (PCs), and MRD levels using two methods. We found that the tCAN was significantly higher with EuroFlow-NGF than with DuraClone (median 8.6 × 106 vs. 5.7 × 106; p  less then  0.0001). In addition, a significant correlation in the MRD levels between the two methods was noted (r = 0.92, p  less then  0.0001). However, in the qualitative analysis, 5.2% (5/96) of the samples showed discrepancies in the MRD levels. In conclusion, the DuraClone is a good option to evaluate MRD in multiple myeloma but it should be used with caution.
