Alkaline stress resulted in an immediate influx of Ca2+ from the extracellular medium. This increase was exacerbated in a mutant lacking the vacuolar Ca2+ transporter VCX1, thus confirming its role in Ca2+ homeostasis. Using mutants in components of a principal Ca2+ channel (MID1, CCH1), the alkaline-dependent Ca2+ spike was greatly reduced, thus highlighting the crucial role of this channel complex in Ca2+ uptake and homeostasis. Exposure to the antiarrhythmic drug amiodarone, known to perturb Ca2+ trafficking, resulted in increased cytoplasmic Ca2+ within seconds that was abrogated by the chelation of Ca2+ in the external medium. Ca2+ import was also dependent on the Cch1/Mid1 Ca2+ channel in amiodarone-exposed cells. In conclusion, the aequorin Ca2+ sensing reporter developed here is an adequate tool with which Ca2+ homeostasis can be investigated in C. albicans.With age, difficulties with masticating and swallowing means food consistency and structure must be modified, such as puréeing food. However, elderly consumers have reported that puréed food's taste lacks appeal. This study shows how puréeing food changes the recognition and dynamics of flavors and new strategies to improve them. Further, to measure the identification and dynamics of flavor, a new sensory method was investigated that combined Free Choice and Temporal Order of Sensation (Free-TOS). Three dishes (macaroni, pizza, and potato salad), their purées, and three pasta purées with added flavors (cheese and dry-cured ham; added directly or as an oil in water emulsion or using two types of emulsions (oil in water and water in oil) were assessed by three groups of 60 consumers using Free-TOS. Results showed that in the purée the frequency of mentioned sensations decreased compared with the dish, as it was more difficult to identify flavors. Adding flavors in powder form only allowed a cheese/ham flavor identification, but in the purées with emulsions, it was possible to identify the dry-cured ham flavor. Therefore, this study showed that the Free-TOS method does not need a predetermined attribute list and registers the actual identified flavors and their order of appearance.Self-healing, an intrinsic healing capacity, helps individuals' bodies and minds to regain wholeness and is significant in the pursuit of one's own healthy ageing and independence. This study was intended to develop and preliminarily test the reliability and validity of the self-healing assessment scale (SHAS) for community-dwelling older adults, and was conducted in three phases. Phase 1 The definitions of self-healing were synthesized from our knowledge of the literature regarding the ontology of self-healing and panels of 25 experts. The initial version of the 12-item questionnaire was developed by the in-depth interviews of focus groups and panels, and the content was validated by six experts. Phase 2 A cross-sectional survey, including a total of 500 community-dwelling older adults with a mean age of 71.76, was then conducted for the preliminary reliability and validity test. The content validity indices were satisfied. Twelve items were retained, and three factors were identified, namely, physical and mental state, socioeconomic and environmental status, and independent lifestyle, which explained 65.8% of the variance under explorative approval. Phase 3 the standardized factor above 60 obtained by confirmatory factorial analysis indicated good convergent validity. The relationship between self-healing and health-related quality of life was confirmed via concurrent validity testing. The SHAS can facilitate the evaluation of factors associated with community-dwelling older adults' self-healing capacity.
