Literature data indicate that feed intake is sensitive to the dietary Lys content particularly in fast-growing birds. From a conceptual and a practical viewpoint, an interaction between genotype (i.e., fast-growing vs. slow-growing birds) and dietary Lys content is of interest, but it needs confirmation owing to a dearth of studies addressing this issue. A study was conducted with 266 Cobb 500 birds and 266 Thai native crossbreed birds serving as models for fast-growing broilers (FGB) and slow-growing broilers (SGB), respectively. Within genotype, chicks were randomly allocated to diets containing either a high (H-LYS = 1.36%), medium (1.17%), or low Lys (1.01%) content. Growth performance and the accretion of protein and selected amino acids were determined in birds from 1 to 21 d of age. Treatments were arranged in a factorial design with 6 replications/treatment. Low Lys vs. H-LYS caused a 42.1% lower feed intake in FGB (P less then 0.001), but not in SGB (P = 0.596). The feed conversion ratio (FCR (g feed/g BW gain)) was lowest in FGB (P less then 0.001) and increased with decreasing dietary Lys contents (P less then 0.001). The Lys induced increase in FCR, however, was more pronounced in SGB (P = 0.025). The absolute protein gain (g/bird) was influenced by the Lys content of feed and decreased by ∼54% and ∼23% in FGB and SGB, respectively (P less then 0.001). The efficiency (% of intake) of protein accretion was found to be greater in FGB (P ≤ 0.001) and decreased with decreasing dietary Lys (P ≤ 0.001). The efficiency of Lys accretion was found to be negatively affected by the dietary Lys content in FGB (P less then 0.001) but not SGB (Pgenotype × dietary Lys = 0.008). It can be concluded that a dietary Lys content of 1.01% does not safeguard both growth performance and body protein accretion efficiency in both FGB and SGB. The suboptimal growth performance in FGB, but not SGB, is partially counteracted by a Lys-induced reduction in feed intake.Three isotopic tracers ([2,3,3-2H3]-L-serine, [2H11]-L-betaine, and [1-13C]-L-methionine) were administered by amnion injection into 18-day-old chick embryos to investigate the kinetics of methionine metabolism. The embryos utilized were from eggs collected from 34-week-old Cobb 500 broiler breeders that were fed either a control diet containing folic acid (1.25 mg/kg diet) and pyridoxine HCl (5 mg/kg diet) or diets devoid of supplemental pyridoxine or folic acid. Intermediate metabolites of methionine metabolism and polyamines were analyzed in 18-day-old chick embryos. There were no differences in hepatic [2H2] methionine or [2H3] cysteine enrichments or in physiological concentrations of sulfur amino acids for chick embryos from breeders fed the control diet and embryos from breeders fed diets containing no pyridoxine or folic acid. Supplementation of B6 or folic acid did not affect the production of methionine and cysteine in chick embryos. However, breeders fed the control diet with both folic acid and pyration when pyridoxine levels are provided in deficient concentrations to the breeder hen. In addition, this research demonstrates that folic acid deficient embryos conserve methionine, rather than catabolize it to cysteine.In birds, sperm storage tubules (SST) located in the utero-vaginal junction are thought to be a site of sperm selection; however, the exact mechanism of sperm selection is poorly understood. Here, we investigated sperm entry into the SST and subsequent fertilization success under a competitive situation created by artificial insemination of a sperm mixture obtained from 2 males. SNX-5422 We employed 2 quail strains, a wild-type and a dominant black (DB) type, as this allows easy assessment of paternity by feather coloration. We found paternity of embryos was biased toward DB males when a sperm mix with similar sperm numbers from the 2 males strains was artificially inseminated into females. Our novel sperm staining method with 2 different fluorescent dyes showed that the DB-biased fertilization was because of the better ability of DB sperm to enter the SST. Moreover, we found that DB sperm had a longer flagellum and midpiece. These characteristics probably allow sperm to swim faster in a high viscosity medium, which may be a similar environment to the lumen of the female reproductive tract. Our results indicated that sperm competition occurs to win a place in the SST and that filling the SST with their own spermatozoa is a critical step to achieve better fertilization success for the male Japanese quail.The aim of this study was to evaluate the effects of genotype, sex, dietary protein level, and their interactions on select carcass characteristics and meat quality of fast- (Ross 308), medium- (Hubbard JA757) and slow-growing (ISA Dual) chickens (n = 2,520). The diet of the low-protein group of chickens had 6% lower CP than the commercial diet fed to the control group. When the chickens reached an average live weight of 2 kg, 10 males and 10 females of each genotype and the diet were selected for slaughter and breast meat-quality analysis. The dressing out and breast percentages were lower in the JA757 (-2.0 and -5.9%, respectively) and ISA Dual chickens (-9.9 and -14.3%, respectively) than those in the Ross 308 chickens. The ISA Dual chickens had higher abdominal fat percentage, higher DM and protein contents and lower ether extract content and shear force value in breast meat than the other genotypes. Significant interaction effects of genotype, sex, and diet were found on the color of breast skin. Among the various combinations of genotype, sex, and diet group, Ross 308 females fed the low-protein diet had the highest redness and yellowness of breast skin, highest pH45 value, and largest fibers, whereas ISA Dual females had the lowest color parameters and pH45 value, and ISA Dual males had the smallest muscle fibers. The low-protein diet was associated with decreased abdominal fat percentage and changes in meat quality parameters, including increased darkness, meat color intensity, drip loss, and muscle fiber area, in all genotypes. The results indicated greater differences in meat quality owing to genotype than to sex or dietary protein level.
