High temperature is an abiotic stress factor that threatens plant growth and development. Autophagy in response to heat stress involves the selective removal of heat-induced protein complexes. Previously, we showed that a crucial autophagy protein from apple, MdATG18a, has a positive effect on drought tolerance. In the present study, we treated transgenic apple (Malus domestica) plants overexpressing MdATG18a with high temperature and found that autophagy protected them from heat stress. Overexpression of MdATG18a in apple enhanced antioxidase activity and contributed to the production of increased beneficial antioxidants under heat stress. Transgenic apple plants exhibited higher photosynthetic capacity, as shown by the rate of CO2 assimilation, the maximum photochemical efficiency of photosystem II (PSII), the effective quantum yield, and the electron transport rates in photosystems I and II (PSI and PSII, respectively). We also detected elevated autophagic activity and reduced damage to chloroplasts in transgenic plants compared to WT plants. In addition, the transcriptional activities of several HSP genes were increased in transgenic apple plants. In summary, we propose that autophagy plays a critical role in basal thermotolerance in apple, primarily through a combination of enhanced antioxidant activity and reduced chloroplast damage. © The Author(s) 2020.Background Well defined constitutional parameters support the physical fatigue resistance in handball to maintain the performance level for the majority of actions. Ideal constitutional conditions are necessary to achieve these physiological advantages in handball. But limited knowledge exists about the upper body posture or the postural control in correlation to the Body Mass Index (BMI), playing years, playing position and throwing arm in professional male handball. Methods Ninety-one male handball players participate (24.1 ± 5.9 years; playing experience 16.6 ± 5.7 years). A three-dimensional back scanner and a pressure measuring plate were used. Results Correlations between BMI and upper body posture and postural control were not significant. Same counts for the comparison between the left and right throwing arm according to upper body posture and postural control (p ≥ 0.05). Correlations between the years of playing can be found at pelvis height (p ≤ 0.04) and for the length of the Center of Pressure (CoP) (p ≤ 0.01). Wing players are 6.5-8.5 cm smaller. The playing position is independently of BMI, age or upper body posture (p ≥ 0.05). Backcourt players have a higher load of the left and a lower load of the right foot compared to wing players (p ≤ 0.001). Left-right comparison (p ≤ 0.001/ 0.01) can be seen in pivot player (covered area), backcourt player (weight distribution left/right [rear] foot), wing player (weight and force distribution left/right foot, covered area). PTC596 Conclusion Goalkeeper, Backcourt and pivot players are taller and heavier than wing players. These physiological demands are not detectable in the upper body posture and slightly in postural control. Wing players have the most asymmetric load distribution and the longest length of CoP. Fitness center employees are ideally placed to observe these forms of unhealthy behavior, and are responsible for ensuring that clients do not put themselves at undue risk; however, to date, no study has addressed both eating disorders and excessive exercise. Therefore, the aim of our study is to determine whether these employees report incidences of these issues, and if they believe they can differentiate between them. Methods One-hundred-and-forty fitness centers in the German-speaking regions of Switzerland were contacted. Given that both disorders can potentially lead to severe health consequences, a detailed description of symptoms, management techniques and resources should be a feature in all Swiss fitness center guidelines. © The Author(s). 2020.Objectives Primary open-angle glaucoma (POAG) is a neurodegenerative disorder leading to a gradual vision loss caused by progressive damage to the optic nerve. Immunological processes are proposed to be involved in POAG pathogenesis. Altered serological autoantibody levels have been frequently reported, but complete analyses of the natural autoantibodies with respect to disease-related alterations are scarce. Here, we provide an explorative analysis of pathways and biological processes that may involve naturally immunogenic proteins and highlight POAG-specific alterations. Methods Mass spectrometry-based antibody-mediated identification of autoantigens (MS-AMIDA) was carried out in healthy and glaucomatous trabecular meshwork (TM) cell lines, using antibody pools purified from serum samples of 30 POAG patients and 30 non-glaucomatous subjects. Selected antigens were validated by protein microarray (n = 120). Bioinformatic assessment of identified autoantigens, including Gene Ontology (GO) enrichment analysis and protein-protein interaction networks, was applied.
