Our findings demonstrate that HIV-1 infection hijacks PLK1 to prevent cell death induced by viral cytopathic effects, and that PLK1 is a promising target for chemical "killing" of HIV-1 reservoir cells.Cytosine base editors (CBEs) enable efficient cytidine-to-thymidine (C-to-T) substitutions at targeted loci without double-stranded breaks. However, current CBEs edit all Cs within their activity windows, generating undesired bystander mutations. In the most challenging circumstance, when a bystander C is adjacent to the targeted C, existing base editors fail to discriminate them and edit both Cs. To improve the precision of CBE, we identified and engineered the human APOBEC3G (A3G) deaminase; when fused to the Cas9 nickase, the resulting A3G-BEs exhibit selective editing of the second C in the 5'-CC-3' motif in human cells. Our A3G-BEs could install a single disease-associated C-to-T substitution with high precision. The percentage of perfectly modified alleles is more than 6000-fold for disease correction and more than 600-fold for disease modeling compared with BE4max. On the basis of the two-cell embryo injection method and RNA sequencing analysis, our A3G-BEs showed minimum genome- and transcriptome-wide off-target effects, achieving high targeting fidelity.Mouse embryonic stem cells cultured with MEK (mitogen-activated protein kinase kinase) and GSK3 (glycogen synthase kinase 3) inhibitors (2i) more closely resemble the inner cell mass of preimplantation blastocysts than those cultured with SL [serum/leukemia inhibitory factor (LIF)]. This selectively strengthens pluripotency loci and renders them addicted to transcription initiation for productive gene body elongation in detriment to Pol2 pause release. By contrast, cell cycle genes are not bound by β-catenin, and proliferation/self-renewal remains tightly controlled by Pol2 pause release under 2i conditions. Our findings explain how pluripotency is reinforced in the ground state and also provide a general model for transcriptional resilience/adaptation upon network perturbation in other contexts.Forecasting fields of oceanic phenomena has long been dependent on physical equation-based numerical models. The challenge is that many natural processes need to be considered for understanding complicated phenomena. In contrast, rules of the processes are already embedded in the time-series observation itself. Thus, inspired by largely available satellite remote sensing data and the advance of deep learning technology, we developed a purely satellite data-driven deep learning model for forecasting the sea surface temperature evolution associated with a typical phenomenon a tropical instability wave. During the testing period of 9 years (2010-2019), our model accurately and efficiently forecasts the sea surface temperature field. This study demonstrates the strong potential of the satellite data-driven deep learning model as an alternative to traditional numerical models for forecasting oceanic phenomena.Precipitation extremes will likely intensify under climate change. However, much uncertainty surrounds intensification of high-magnitude events that are often inadequately resolved by global climate models. In this analysis, we develop a framework involving targeted dynamical downscaling of historical and future extreme precipitation events produced by a large ensemble of a global climate model. This framework is applied to extreme "atmospheric river" storms in California. We find a substantial (10 to 40%) increase in total accumulated precipitation, with the largest relative increases in valleys and mountain lee-side areas. We also report even higher and more spatially uniform increases in hourly maximum precipitation intensity, which exceed Clausius-Clapeyron expectations. Up to 85% of this increase arises from thermodynamically driven increases in water vapor, with a smaller contribution by increased zonal wind strength. These findings imply substantial challenges for water and flood management in California, given future increases in intense atmospheric river-induced precipitation extremes.Weyl semimetals are crystals in which electron bands cross at isolated points in momentum space. Associated with each crossing point (or Weyl node) is a topological invariant known as the Berry monopole charge. The circular photogalvanic effect (CPGE), whereby circular polarized light generates a helicity-dependent photocurrent, is a notable example of a macroscopic property that emerges directly from the topology of the Weyl semimetal band structure. Recently, it was predicted that the amplitude of the CPGE associated with optical transitions near a Weyl node is proportional to its monopole charge. In chiral Weyl systems, nodes of opposite charge are nondegenerate, opening a window of wavelengths where the CPGE resulting from uncompensated Berry charge can emerge. Here, we report measurements of CPGE in the chiral Weyl semimetal RhSi, revealing a CPGE response in an energy window that closes at 0.65 eV, in agreement with the predictions of density functional theory.
