A line-box transformation method is also proposed to fit the widely used box-annotations. Meanwhile, we explore the influence of deformable convolution base-offset on detection performance and propose a base-offset initialization strategy to further promote the gain brought by ALR. Notably, the proposed ALR pattern can be introduced into both anchor-based and anchor-free frameworks. We validate the effectiveness of ALR on the Caltech-USA and CityPersons datasets. Experimental results show that our approach outperforms the baseline significantly through simple modifications and achieves competitive accuracy with other methods without bells and whistles.The neuronal Hu/ELAV-like proteins HuB, HuC and HuD are a class of RNA-binding proteins that are crucial for proper development and maintenance of the nervous system. These proteins bind to AU-rich elements (AREs) in the untranslated regions (3'-UTRs) of target mRNAs regulating mRNA stability, transport and translation. In addition to these cytoplasmic functions, Hu proteins have been implicated in alternative splicing and alternative polyadenylation in the nucleus. The purpose of this study was to identify transcriptome-wide effects of HuD deletion on both of these nuclear events using RNA sequencing data obtained from the neocortex of Elavl4-/- (HuD KO) mice. HuD KO affected alternative splicing of 310 genes, including 17 validated HuD targets such as Cbx3, Cspp1, Snap25 and Gria2. In addition, deletion of HuD affected polyadenylation of 53 genes, with the majority of significantly altered mRNAs shifting towards usage of proximal polyadenylation signals (PAS), resulting in shorter 3'-UTRs. None of these genes overlapped with those showing alternative splicing events. Overall, HuD KO had a greater effect on alternative splicing than polyadenylation, with many of the affected genes implicated in several neuronal functions and neuropsychiatric disorders.The presence of both microplastics and per- and polyfluoroalkyl substances (PFAS) is ubiquitous in the environment. The ecological impacts associated with their presence are still poorly understood, however, these contaminants are extremely persistent. Although plastic in the environment can concentrate pollutants, factors such as the type of plastic and duration of environmental exposure as it relates to the degree of adsorption have received far less attention. To address these knowledge gaps, experiments were carried out that examined the interactions of PFAS and microplastics in the field and in a controlled environment. For field experiments, we measured the abundance of PFAS on different polymer types of microplastics that were deployed in a lake for 1 month and 3 months. Based on these results, a controlled experiment was conducted to assess the adsorption properties of microplastics in the absence of associated inorganic and organic matter. The adsorption of PFAS was much greater on the field-incubated plastic than what was observed in the laboratory with plastic and water alone, 24 to 259 times versus one-seventh to one-fourth times background levels. These results suggest that adsorption of PFAS by microplastics is greatly enhanced by the presence of inorganic and/or organic matter associated with these materials in the environment, and could present an environmental hazard for aquatic biota.Asthma and chronic obstructive pulmonary disease (COPD) are now recognized to be able to co-exist as asthma-COPD overlap (ACO). It is clinically relevant to evaluate whether patients with COPD concurrently have components of asthma in primary care. This is because (i) ACO is a relatively common condition among asthma (over 40 years of age) or COPD irrespective of its diagnosis criteria; (ii) patients with ACO can have higher frequency of exacerbation and more rapid decline in lung function than those with asthma or COPD; and (iii) asthmatic features such as eosinophilic airway inflammation are promising indicators for prediction of inhaled corticosteroid-responsiveness in COPD. In this study, we aimed to investigate the effects of Kinsenoside (KD), a natural active ingredient of Anoectochilus roxburghii, on estrogen-induced cholestatic liver injury in Sprague-Dawley rats and the underlying mechanism. The rats were randomly divided into six groups control group, model group, low-dose KD group (50 mg/kg body weight, KD-L), medium-dose KD group (100 mg/kg body weight, KD-M), high-dose KD group (200 mg/kg body weight, KD-H) and ursodeoxycholic acid group (40 mg/kg body weight, UDCA). 17α-Ethinylestradiol (EE) was used to establish an experimental animal model of estrogen-induced cholestasis (EIC). The results demonstrated that KD alleviated liver pathologic damage, serum biochemical status and inhibited hepatocellular microstructure disorder and bile duct hyperplasia in EE-induced cholestatic rats. Mechanically, KD alleviated EE-induced cholestatic liver injury by inhibiting inflammatory responses and regulating bile acid homeostasis.
