We identified BGCs coding for paenibactin, paenibacterin, anabaenopeptin NZ857, icosalide A/B, polymyxin, and bicornutinA1/A2 with 100% similarity. The BGCs with lower sequence similarity to paenibacterin, polymyxin B, colistin A/B, pellasoren, tridecaptin, pelgipeptin, and marthiapeptide were also identified. Furthermore, 13 putative NRPS BGCs, 3 NRPS-T1PKS hybrid clusters, a T1PKS, and a bacteriocin BGC were identified with very low similarity (≤ 25%) or no similarity with known antibiotics. Further experimental investigations may result in the discovery of novel antimicrobial drugs.
The evaluation of German postanesthesia questionnaires is often restricted to inpatient medical care. The anesthesiological questionnaire for patients (ANP) has previously been validated only after medical care of inpatients. Therefore, the aim of the present study was to evaluate the validity of the ANP in the context of outpatient surgery and anesthesia.
Data from 4547patients (mean age 18-89 years, sex m/f, 55%/45%), scheduled for ambulatory orthopedic and trauma surgery, were analyzed. All patients received astandard induction of anesthesia with propofol, sufentanil and 4 mg dexamethasone. THALSNS032 Maintenance of anesthesia was achieved with abalanced technique comprising volatile anesthesia (either sevoflurane or desflurane) and sufentanil. In addition, patients undergoing operations on the anterior cruciate ligament received afemoral nerve block with 12-15 ml 0.5% ropivacaine. Of the patients 13% remained in hospital overnight as aresult of complex operations (combined anterior and posterior cruciate ligamenal analyses delivered plausible results, which support the validity of the ANP. The ANP may also be used for assessment of postoperative discomfort and patient satisfaction in the setting of outpatient surgery. In view of the short time interval between the two measurement points, the ANP for outpatients could in future be reduced to a single postoperative questionnaire.Motor adaptation plays an important role in the acquisition of new motor skills. It has been reported that cognitive tasks can promote motor adaptation; however, which cognitive tasks effectively promote motor adaptation remains unknown. This study aimed to examine what factors of cognitive tasks contribute to promoting motor adaptation. Forty-two healthy young adults were randomly assigned to one of three groups incongruent Stroop task group (iSTG), congruent Stroop task group (cSTG), and control group (CG). All participants underwent 20 blocks of a mouse-tracking task on the 1st and 2nd days. Before the mouse tracking task on the 1st day, the iSTG and cSTG completed the incongruent and congruent Stroop tasks, respectively. Participants in the CG did not perform any cognitive tasks. On the 28th day, all participants underwent 3 blocks of the mouse tracking task to evaluate their retention of motor adaptation. As a result, on the 1st day, the mouse tracking task performance improved equally for both groups. However, on the 2nd and 28th days, the mouse tracking task performance in the iSTG showed greater improvements for all blocks compared to those in the CG. These results suggest that the incongruent Stroop task promotes motor adaptation, but the congruent Stroop task does not. In addition, it is suggested that factors, which are primarily involved in the incongruent Stroop task, might promote motor adaptation.Point-of-care (POC) technologies and testing programs hold great potential to significantly improve diagnosis and disease surveillance. POC tests have the intrinsic advantage of being able to be performed near the patient or treatment facility, owing to their portable character. With rapid results often in minutes, these diagnostic platforms have a high positive impact on disease management. POC tests are, in addition, advantageous in situations of a shortage of skilled personnel and restricted availability of laboratory-based analytics. While POC testing programs are widely considered in addressing health care challenges in low-income health systems, the ongoing pandemic of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infections could largely benefit from fast, efficient, accurate, and cost-effective point-of-care testing (POCT) devices for limiting COVID-19 spreading. The unrestrained availability of SARS-CoV-2 POC tests is indeed one of the adequate means of better managing the COVID-19 outbreak. A large number of novel and innovative solutions to address this medical need have emerged over the last months. Here, we critically elaborate the role of the surface ligands in the design of biosensors to cope with the current viral outbreak situation. Their notable effect on electrical and electrochemical sensors' design will be discussed in some given examples. Graphical abstract.A photoelectrochemical (PEC) immunosensing strategy based on the multiple quenching of polydopamine nanoparticles (PDA NPs) to Mn2+-doped CdS-modified TiO2 nanoparticles (TiO2/CdSMn) was designed for the highly sensitive detection of carcinoembryonic antigen (CEA). The uniform PDA NPs possessed good dispersibility, good biocompatibility, and abundant functional groups for biomolecule assembly. They also had unique photophysical properties, with light absorption spanning the visible to infrared light range. When the immune-recognition brought the PDA NPs close to the TiO2/CdSMn interface, the PDA NPs competed with TiO2/CdSMn to absorb light, consumed photoelectrons generated in the TiO2/CdSMn, and hindered the access of electron donors to photoactive materials. The contribution from these aspects thus led to a significant decrease in photocurrent. Benefiting from the multiple quenching mechanism, the PEC immunosensor showed high sensitivity for CEA detection. Under optimal conditions, a low detection limit of 0.02 pg/mL and a wide linear relationship from 0.1 pg/mL to 100 ng/mL were obtained. The immunoassay showed good reproducibility and stability, and good selectivity and high accuracy in serum sample analysis. In this regard, PEC immunosensors may have great application potential for screening tumor markers and the prevention and monitoring of serious diseases.
