It was generally found from the experiments that (i) the innovative joint is capable of achieving the design criterion of the 'strong jointweak member' with appropriate designs, and (ii) by decreasing the height factor and increasing the volume ratio of the steel meshes, the axial compressive strengths of the joints significantly increased, while the increase of the length factor is advantageous but limited to the resistances of the joint specimens. Because of the lack of existing design methods for the innovative joints, new design expressions were proposed to calculate the axial compression resistances of the innovative joints subjected to bearing loads, with the local compression effect, the confinement effect provided by the multilayers of steel meshes and the height effect of concrete considered. It was found that the proposed design methods were capable of providing accurate and safe resistance predictions for the innovative joints. In continuous casting processes, inevitable voids (damage) are generated inside the material. The subsequent forming process of hot flat rolling offers the potential of healing these defects by closing the voids and bonding the internal surfaces. In this paper, different forming conditions from hot flat rolling process were characterized with micromagnetic measurement techniques and the influence of the damage evolution on the fatigue behavior was investigated. To characterize the reduction of voids through hot flat rolling processes, nondestructive testing techniques are required. Therefore, micromagnetic measurements such as Barkhausen noise, incremental permeability, and harmonic analysis were carried out, correlated with the number of voids, and compared with each other. The influence of damage evolution of different forming conditions on the fatigue behavior was characterized based on instrumented constant amplitude and multiple amplitude (load increase) tests. A significant increase in fatigue strength due to the hot flat rolling process, which leads to a reduction in the number of voids, was observed. In addition, the fracture surfaces of the specimens were analyzed in the scanning electron microscope.Through this Special Issue, you will discover the potentiality of inorganic polymerization (sol-gel process) which is a unique and versatile way for the preparation of materials [...].Recently, skeletal muscle represents a complex and challenging tissue to be generated in vitro for tissue engineering purposes. Several attempts have been pursued to develop hydrogels with different formulations resembling in vitro the characteristics of skeletal muscle tissue in vivo. This review article describes how different types of cell-laden hydrogels recapitulate the multiple interactions occurring between extracellular matrix (ECM) and muscle cells. A special attention is focused on the biochemical cues that affect myocytes morphology, adhesion, proliferation, and phenotype maintenance, underlining the importance of topographical cues exerted on the hydrogels to guide cellular orientation and facilitate myogenic differentiation and maturation. Moreover, we highlight the crucial role of 3D printing and bioreactors as useful platforms to finely control spatial deposition of cells into ECM based hydrogels and provide the skeletal muscle native-like tissue microenvironment, respectively.Potato (Solanum tuberosum) is among the best producers of edible biomass in terms of yield per hectare and a variety of different regional cultivars are used as a staple commodity in many countries. However, this crop is attacked by several diseases, with the worst being the late blight disease caused by Phytophthora infestans. Stacking of resistance (R) genes from wild Solanum relatives are interesting prospects for the sustainable control of late blight. Therefore, we optimized methods for the efficient generation and screening of R-gene-containing transformants in tetraploid and diploid hybrid potato genotypes. Using these methods, a high transformation efficiency was achieved for the transformation of tetraploid and diploid potato lines with a triple resistance (3R) gene construct. Transformation efficiencies were improved by optimizing several factors affecting regeneration, including the quality of the starting plant material, and the composition of the plant growth regulators used during selective regeneration. A refreshment protocol was designed to alleviate in vitro related stress in stock plants, which significantly improved the growth vigor and resulted in a 4- to 10-fold increase in transformation efficiency. Furthermore, long-term exposure to exogenous Indole-3-butyric acid that is usually used for the initiation of roots in vitro, was found to cause aberrant morphological phenotypes in potato.Synthesis of four compounds belonging to mesoionic class, (E)-3-phenyl-5-(phenylamino)-2-styryl-1,3,4-thiadiazol-3-ium chloride derivatives (5a-d) and their biological evaluation against MT2 and C92 cell lines infected with human T-cell lymphotropic virus type-1 (HTLV-1), which causes adult T-cell leukemia/lymphoma (ATLL), and non-infected cell lines (Jurkat) are reported. The compounds were obtained by convergent synthesis under microwave irradiation and the cytotoxicity was evaluated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays. Results showed IC50 values of all compounds in the range of 1.51-7.70 M in HTLV-1-infected and non-infected cells. Daporinad molecular weight Furthermore, it was observed that 5b could induce necrosis after 24 h for Jurkat and MT2 cell lines. The experimental (fluorimetric method) and theoretical (molecular docking) results suggested that the mechanism of action for 5b could be related to its capacity to intercalate into DNA. Moreover, the preliminary pharmacokinetic profile of the studied compounds (5a-d) was obtained through human serum albumin (HSA) binding affinity using multiple spectroscopic techniques (circular dichroism, steady-state and time-resolved fluorescence), zeta potential and molecular docking calculations. The interaction HSA5a-d is spontaneous and moderate (Ka ~ 104 M-1) via a ground-state association, without significantly perturbing both the secondary and surface structures of the albumin in the subdomain IIA (site I), indicating feasible biodistribution in the human bloodstream.
