This is the first trial to evaluate an irinotecan-based regimen in combination with antiangiogenesis tyrosine kinase inhibitors in Ewing sarcoma (EWS). A 3+3 design with dose de-escalation was used to determine the most appropriate dose of irinotecan in each cohort. The next dose-expanding phase II portion employed a conventional two-stage study design model. The objective response rate was 62.5% for adults and 83.3% for children. Median overall survival was not matured. The objective of this study was to identify the main socioeconomic characteristics, personal experiences, resources, and competencies associated with sexism in a sample of adolescents from different European countries. Baseline data from the Lights4Violence project included 1555 students ages 12-17 from secondary schools in six European countries (Spain, Italy, Romania, United Kingdom, Portugal, and Poland). Linear regression models were carried out, stratified by sex for benevolent (BS) and hostile (HS) dimensions of the Ambivalent Sexism Inventory. The average age of the sample was 14.3 years (SD = 1.5), 59.3% were girls. Boys scored higher on the measure of sexism (mean BS = 29.7; HS = 29.1) than girls (BS = 27.5, HS = 23.0; p less then .001). Girls whose mothers had a university degree reported lower BS (β = -0.113; p = .023) (reference lower education). Girls who had experienced dating violence reported higher HS (β = .080; p = .010) than those who had never been in an intimate relationship. For both sexes, high aggressiveness was associated with high levels of HS, and high aggressiveness was related to high levels of BS in boys. High assertiveness was associated with high levels of BS in both sexes and with high levels of HS in boys. A high level of problem-solving ability was associated with lower HS in both sexes. The study reinforces the need to invest in school programs aimed at preventing dating violence and promoting positive youth development.Semaphorins, a large group of highly conserved proteins, consist of eight subfamilies that are widely expressed in vertebrates, invertebrates, and viruses and exist in membrane-bound or secreted forms. First described as axon guidance cues during neurogenesis, semaphorins also perform physiological functions in other organ systems, such as bone homeostasis, immune response, and tumor progression. Semaphorin 7A (SEMA7A), also known as CDw108, is an immune semaphorin that modulates diverse immunoinflammatory processes, including immune cell interactions, inflammatory infiltration, and cytokine production. In addition, SEMA7A regulates the proliferation, migration, invasion, lymph formation, and angiogenesis of multiple types of tumor cells, and these effects are mediated by the interaction of SEMA7A with two specific receptors, PLXNC1 and integrins. Thus, SEMA7A is intimately related to the pathogenesis of multiple autoimmune and inflammation-related diseases and tumors. This review focuses on the role of SEMA7A in the pathogenesis of autoimmune disorders, inflammatory diseases, and tumors, as well as the underlying mechanisms. The aim of this study was to elucidate the salt tolerance mechanism of two Italian ryegrass cultivars, 'Abundant' and 'Angus'. Under hydroponic conditions, two cultivars of Italian ryegrass with different salt tolerance were exposed to 0 and 300 mM NaCl solution for 1 week, respectively. The results showed that salt stress decreased relative growth rate and relative water content, especially in salt-sensitive 'Angus'. The salt-tolerant 'Abundant' cultivar alleviated reactive oxygen species (ROS) induced burst and cell damage. However, 'Angus' exhibited a greater activity of superoxide dismutase (SOD) and peroxidase (POD) than 'Abundant'. Additionally, 'Abundant' exhibited higher photosynthetic efficiency than 'Angus' under salt stress condition. Salt treatment significantly increased the Na/K, Na/Mg, and Na/Ca ratios in the leaves and roots of both cultivars, with a pronounced effect in salt-sensitive 'Angus'. The metabolite analysis of leaf polar extracts revealed 41 salt responsive metabolites in both cultivars, mainly consisting of amino acids, organic acids, fatty acids, and sugars. Following exposure to salt conditions, salt-sensitive 'Angus' had a higher level of metabolites and more uniquely upregulated metabolites were detected. Based on these findings, we conclude that the 'Abundant' cultivar emerged as a favorite in saline-alkali soil, while the 'Angus' cultivar is suitable for planting in normal soil. It appears that the high salt tolerance of 'Abundant' is partly to prevent the plant from ionic homeostasis disruption.
To describe the clinical and procedural outcomes of per-oral endoscopic myotomy (POEM) for achalasia in Australia.
Prospective observational study in three Australian tertiary referral centres, 5 May 2014 - 27 October 2019 (66 months).
Patients who had undergone POEM for achalasia.
Eckardt scores calculated prior to POEM and six months, one year, and two years after POEM. The primary outcome was clinical success, defined as an Eckardt score of 3 or less without a second intervention.
142 patients underwent POEM for achalasia; their mean age was 52 years (SD, 18 years), 83 were men (58%), and the median length of hospital stay two days (IQR, 1-3 days). Their mean Eckardt score before POEM was 8.0 (SD, 2.4) and 1.1 (SD, 1.6) six months after POEM; it did not change significantly between six months and two years after POEM (mean monthly increase, 0.014 points; 95% CI, -0.001 to 0.029).
