Here, we introduce an improved mariner-based transposon system with an enhanced stability during propagation and versatile applications in mutagenesis. We used a low copy number plasmid as a transposon delivery vehicle that affords a lower frequency of unintended recombination during vector construction and propagation to improve the construct integrity in E. coli We generated a variety of transposons allowing for gene disruption or artificial overexpression each in combination with one of four different antibiotic resistance markers. In addition, we provide transposons that will report gene/protein expression due to transcriptional or translational coupling. We believe that the TnFLX system will help enhance the flexibility of future transposon modification and application in Bacillus and other organisms.ImportanceThe stability of transposase-encoding vectors during cloning and propagation is crucial for the reliable application of transposons. We recently showed that the emerging macrolide resistance in the soil-borne equine and zoonotic pathogen Rhodococcus equi is conferred by the erm (46) gene carried on the 87-kb conjugative plasmid pRErm46. Here we investigated the conjugal transferability of pRErm46 to 14 representative bacteria likely encountered by R. equi in the environmental habitat. In vitro mating experiments demonstrated conjugation to different members of the genus Rhodococcus as well as to Nocardia and Arthrobacter spp. at frequencies ranging from ∼10-2 to 10-6 pRErm46 transfer was also observed in mating experiments in soil and horse manure, albeit at a low frequency and after prolonged incubation at 22-30°C (environmental temperatures), not 37°C. All transcojugants were able to transfer pRErm46 back to R. equi Conjugation could not be detected with Mycobacterium and Corynebacterium spp. or several membdata support the notion that environmental microbiota may act as reservoirs for the endemic maintenance of antimicrobial resistance in an antibiotic pressurized farm habitat. PWD inflicts huge economic losses on the swine industry each year, making development of alternative treatment and prevention measures for PWD essential. In this study, we constructed two multi-epitope fusion antigens (MEFAs) that carried, expressed, and displayed representative epitopes of F4, F5, F6, F18 and F41 ETEC fimbriae. These MEFAs used either the F4 major subunit FaeG or the F18 adhesive subunit FedF as a backbone. To assess the potential of these MEFAs as anti-fimbriae vaccine candidates that could help prevent PWD, we generated computational models of the MEFAs, constructed them, and then tested their immunogenicity by using them to immunize mice. Computaty, a commercialized live monovalent F4 oral vaccine and a bivalent F4/F18 oral vaccine have been demonstrated to effectively protect piglets in the F4+ and F18+ ETEC challenge models. However, they will not provide cross-protection against F5+, F6+, or F41+ ETEC-associated PWD cases due to lack of expressing these fimbriae antigens. Thus, a multivalent vaccine containing all ETEC five fimbriae would be more effective in preventing against ETEC-driven PWD. In this study, we designed two fimbriae-targeted MEFAs using the MEFA technology, further study demonstrated co-administrated these MEFAs in mice can induced protective antibodies against five fimbriae expressed by ETEC. These MEFAs could be used as an efficient PWD vaccine candidate, furthermore, MEFA-based structural technology provides an alternative and promising strategy for the development of vaccines against pathogens with heterogeneous virulence factors. The aim of this study was to determine levels of PA among a community of free-living Hispanic/Latino adults with T2D using a research accelerometer, a consumer device and a pictogram self-assessment questionnaire. RESEARCH DESIGN AND METHODS This was a cross-sectional, observational study. Participants (57 women and 31 men, body mass index (kg/m2) 32.2±7.9 and 29.9±4.5, waist circumference 97.1±30.1 and 93.7±33.0 cm and hemoglobin A1c 8.0±2.0 and 8.1%±1.8%, respectively) wore an ActiGraph (AG) on the hip and a Fitbit (FB) on the wrist for 1 week to estimate daily steps and energy expenditure (EE). Participants reported type and intensity of PA using English-language or Spanish-language pictograms and a 10-point Likert scale (1='not active' to 10='very, very active').
