Studies so far conducted on irritable bowel syndrome (IBS) have been focused mainly on the role of gut bacterial dysbiosis in modulating the intestinal permeability, inflammation, and motility, with consequences on the quality of life. mTOR inhibitor Limited evidences showed a potential involvement of gut fungal communities. Here, the gut bacterial and fungal microbiota of a cohort of IBS patients have been characterized and compared with that of healthy subjects (HS). The IBS microbial community structure differed significantly compared to HS. In particular, we observed an enrichment of bacterial taxa involved in gut inflammation, such as Enterobacteriaceae, Streptococcus, Fusobacteria, Gemella, and Rothia, as well as depletion of health-promoting bacterial genera, such as Roseburia and Faecalibacterium. Gut microbial profiles in IBS patients differed also in accordance with constipation. Sequence analysis of the gut mycobiota showed enrichment of Saccharomycetes in IBS. Culturomics analysis of fungal isolates from feces showed enrichment of Candida spp. displaying from IBS a clonal expansion and a distinct genotypic profiles and different phenotypical features when compared to HS of Candida albicans isolates. Alongside the well-characterized gut bacterial dysbiosis in IBS, this study shed light on a yet poorly explored fungal component of the intestinal ecosystem, the gut mycobiota. Our results showed a differential fungal community in IBS compared to HS, suggesting potential for new insights on the involvement of the gut mycobiota in IBS. KEY POINTS • Comparison of gut microbiota and mycobiota between IBS and healthy subjects • Investigation of cultivable fungi in IBS and healthy subjects • Candida albicans isolates result more virulent in IBS subjects compared to healthy subjects.A recent correspondence suggested that taking home ashes and launching digital Lenten ashes filters are non-traditional yet creative ways for the Catholic Church to perform the ritual practice in celebrating Ash Wednesday. These creative practices are deemed as necessary so that the Catholic community can celebrate the liturgical celebration while ensuring public health. With this, we propose in this paper practices that can be done during the holy week so that the spread of the coronavirus will be mitigated.
HDL in humans is composed of a heterogeneous group of particles varying in protein composition as well as biological effects.

We investigated the prospective associations between HDL subspecies containing and lacking apoC-III at baseline and insulin sensitivity at year 3.

A prospective cohort study of 864 healthy volunteers drawn from the RISC study, a multi-center European clinical investigation, whose recruitment initiated in 2002 with a follow-up of 3 years.

Insulin sensitivity was estimated from an oral glucose tolerance test (OGTT) at baseline and year 3, and by euglycemic-hyperinsulinemic clamp at baseline only. The apolipoprotein concentrations were measured at baseline by a sandwich ELISA-based method.

The two HDL subspecies demonstrated significantly opposite associations with insulin sensitivity at year 3 (p-heterogeneity=0.004). The highest quintile of HDL containing apoC-III was associated with a 1.2% reduction in insulin sensitivity (p-trend=0.02), while the highest quintile of HDL lacking apoC-III was associated with a 1.3% increase (p-trend=0.01), compared to the lowest quintile. No significant association was observed for total HDL, and VLDL and LDL containing apoC-III. ApoC-III contained in HDL was associated with a decrease in insulin sensitivity even more strongly than plasma total apoC-III.

Both HDL containing apoC-III and apoC-III in HDL adversely affect the beneficial properties of HDL on insulin response to glucose. Our results support the potential of HDL-associated apoC-III as a promising target for diabetes prevention and treatment.

The purpose of this theoretical study was to examine the validity of some commonly used requirements in terms of image contrast and contrast stability in rooms with fluctuating illuminance. By using the grayscale standard display function (Dicom part 14), contrast was calculated for several combinations of display minimum and maximum luminance as well as the possible range of illuminance fluctuations that will not exceed given contrast tolerance levels. The results show that some requirements are only valid in dark viewing rooms, which are also usually recommended or enforced. However, image contrast, contrast stability, ergonomics and fatigue would improve by using brighter displays in brighter rooms. With a better set of requirements, it would also be possible for displays in brighter rooms, like dentist departments and operating rooms, to conform to the requirements.
Previous studies examining unidirectional relationships between social isolation and physical functioning amongst older adults may be biased due to reverse causality. This study leveraged data from a US national sample of older adults and a novel estimation method to identify bidirectional associations between these two phenomena and their associated temporal dynamics.

The analyses were based on nine waves of panel data from a sample of adults aged 65+ years from the US National Health and Aging Trends Study (n=12,427 individuals) and a cross-lagged panel model. Social isolation was measured using the Social Isolation Index (SII) and physical functioning through the Short Physical Performance Battery (SPPB).

One SD increase in the SPPB was associated with a -0.013 SD decrease (95% CI -0.023, -0.004) in the SII a year later, whereas one SD increase in the SII was associated with a larger -0.058 SD decrease (95% CI -0.069, -0.046) in the SPPB a year later. The SII→SPPB effect lasted for ~5 years, whereas the SPPB→SII lasted for ~3 years.
