Microbial communities drive diverse processes that impact nearly everything on this planet, from global biogeochemical cycles to human health. Harnessing the power of these microorganisms could provide solutions to many of the challenges that face society. However, naturally occurring microbial communities are not optimized for anthropogenic use. An emerging area of research is focusing on engineering synthetic microbial communities to carry out predefined functions. Microbial community engineers are applying design principles like top-down and bottom-up approaches to create synthetic microbial communities having a myriad of real-life applications in health care, disease prevention, and environmental remediation. Multiple genetic engineering tools and delivery approaches can be used to 'knock-in' new gene functions into microbial communities. A systematic study of the microbial interactions, community assembling principles, and engineering tools are necessary for us to understand the microbial community and to better utilize them. Published by Portland Press Limited on behalf of the Biochemical Society.Emerald ash borer (EAB, Agrilus planipennis Fairmaire [Coleoptera Buprestidae]) is a wood boring beetle that is an invasive pest of ash trees (Fraxinus spp.) in North America. In 2014, it was reported that EAB had infested white fringetree (Chionanthus virginicus L. [Lamiales Oleaceae]) in Ohio and was since found to have infested this species across its invasive range. In 2018, we reexamined 166 white fringetrees in Illinois, Indiana, Ohio, and Pennsylvania that had been previously examined for EAB attack in 2015 to determine their fate. We assessed tree health and EAB infestation in each tree, assigned an infestation status of newly, continuously, not reinfested, or never infested, and compared the trees' current status to their 2015 status. This assessment was done to determine whether their health and infestation status had changed through the EAB invasion wave. We found that attack rates declined 26% of trees were infested in 2015 whereas only 13% were in 2018, likely coinciding with declining beetle populations in the area. Overall tree health improved for trees that were not reinfested by EAB after a record of attack in 2015, suggesting that they can survive and recover from EAB attack. Conversely, health declined for newly and continuously infested trees, indicating that they became stressed from EAB attack. Although the majority of the trees survived the invasion wave, several were removed from various sites due to EAB attack suggesting that white fringetree varies in its resistance and tolerance to attack. As beetle populations continue to expand geographically, infestation rates will likely increase and health of white fringetrees will decrease with the EAB attack wave, especially as EAB reaches denser populations of fringetrees. © The Author(s) 2020. For permissions, please e-mail [email protected] To study the correlation between adenomatous polyposis coli (APC) gene 3' untranslated region (UTR) single nucleotide polymorphisms (SNPs) and their interactions with environmental factors and the risk of colorectal cancer (CRC) in a Chinese Han population. METHODS Genotypes of APC gene 3'UTR rs1804197, rs41116, rs448475, and rs397768 loci in 340 Chinese Han patients with CRC and 340 healthy controls were analyzed. All patients with CRC were analyzed for progression-free survival (PFS) during a 3-year follow-up. GSK-3008348 order RESULTS The risk of CRC in subjects carrying the APC gene rs1804197 A allele was 2.95-times higher than for the C allele carriers. The interactions of the rs1804197 SNP with body mass index (BMI) and smoking were associated with the risk of CRC. The risk of CRC in the APC gene rs397768 G allele carriers was 1.68-times higher than in the A allele carriers. The interaction between the rs397768 locus SNP and gender was also associated with the risk of CRC. The 3-year PFS of patients with APC gene rs1804197 AA genotype, CA genotype, and CC genotype CRC decreased in this order, with significant difference. In addition, the 3-year PFS of rs397768 locus GG genotype, AG genotype, and AA genotype CRC patients decreased in this order, and the difference was significant. CONCLUSION The rs1804197 locus in the 3'UTR region of the APC gene and its interactions with BMI and smoking are associated with the risk of CRC in a Chinese Han population. In addition, the interaction between rs397768 locus SNP and gender is related to the risk of CRC. © 2020 The Author(s).BACKGROUND The C5 complement inhibitor eculizumab is first-line treatment in atypical hemolytic uremic syndrome (aHUS) going along with a highly increased risk of meningococcal infections. Serogroup B meningococci (MenB) are the most frequently encountered cause for meningococcal infections in Europe. Efficacy of the protein-based MenB-vaccine Bexsero in aHUS has not been determined and testing is only possible in patients off-treatment with eculizumab as a human complement source is required. METHODS Patients with aHUS were vaccinated with two doses of the protein-based MenB-vaccine Bexsero. Serum bactericidal antibody (SBA) titers against factor H binding protein (fHbp) of MenB were determined in 14 patients with aHUS off-treatment with eculizumab. RESULTS Only 50% of patients showed protective human serum bactericidal antibody (hSBA) titers (≥14) against MenB following two vaccinations. Bactericidal antibody titers were relatively low (≤18) in three of seven patients with protective titers. While 71% of patients were on immunosuppressive treatment for either thrombotic microangiopathy or renal transplantation at either first or second vaccination, all four patients not receiving any immunosuppressive treatment showed protective bactericidal antibody response. Time between second vaccination and titer measurement was not significantly different between patients with protective titers compared with those with non-protective titers, while time between first and second vaccination was significantly longer in patients with protective titers going along with a tendency for reduction in immunosuppressive treatment. CONCLUSIONS Efficacy of vaccination against MenB is insufficient in patients with aHUS. Response to vaccination seems to be hampered by immunosuppression. Therefore, implementation of adequate antibiotic prophylaxis seems pivotal. © 2020 The Author(s).
