PM2.5 collected using cyclonic separation led to a higher secretion of interleukins 6 and 8 (IL-6, IL-8) from airway epithelial cells, and IL-6, IL-1β, tumor necrosis factor-α (TNF-α) secretion, cluster of differentiation 86 (CD86), and dendritic and epithelial cells 205 (DEC205) expression on antigen-presenting cells, compared with the effects of filter-collected PM2.5. Furthermore, PM2.5 collected using cyclonic separation increased inflammatory cytokine levels and induced lung inflammation in vivo. selleck inhibitor These results suggest that crude PM2.5 collected using cyclonic separation causes stronger biological responses than filter-collected PM2.5. Previous studies have shown an association between prenatal exposure to particulate matter (PM) and adverse brain development. However, it is unclear whether gestational exposure to community-sampled residential PM has an impact on the developing brain.
We aimed to test whether in utero exposure to PM from residential roof spaces (ceiling voids) alters critical foetal neurodevelopmental processes.
Pregnant C57BL/6 mice were intranasally exposed to 100μg of roof space particles (~5mgkg
) in 50μl of saline, or saline alone under light methoxyflurane anaesthesia, throughout mid-to-late gestation. At 2 weeks post-natal age, pups were sacrificed and assessed for body and brain growth. The brain tissue was collected and examined for a range of neurodevelopmental markers for synaptogenesis, synaptic plasticity, gliogenic events and myelination by immunohistochemistry.
Gestational exposure to roof space PM reduced post-natal body and brain weights. There was no significant effect of roof space PM exposure on synaptogenesis, synaptic plasticity or astrocyte density. However, PM exposure caused increased myelin load in the white matter and elevated microglial density which was dependent on the PM sample. These effects were found to be consistent between male and female mice.

Our data suggest that exposure to residential roof space PM during pregnancy impairs somatic growth and causes neuropathological changes in the developing brain.It is well known that viscosity reduces the intake rates in nectar-feeding insects, such as nectivorous ants, though it remains unclear whether viscosity imposes a higher energy investment in these insects, and how this affects their feeding motivation. To address this issue, we studied feeding behavior, metabolism, and pharyngeal pump activity in the carpenter ant Camponotus mus during ingestion of ad libitum sucrose solutions. In some solutions tylose was added to modify viscosity without changing its sucrose concentration, in a way that allowed comparing (1) two solutions with the same viscosity and different sucrose concentration (10 T and 50), and (2) two solutions with different viscosity and the same sucrose concentration (50 and 50 T). The viscosity increase was detrimental to the metabolic rate and energy balance. Ants feeding on a solution with high sucrose concentration and increased viscosity (50 T) spent extra-time until reaching a crop load similar to that reached by ingesting the solution without tylose (50). For all solutions offered, ants started feeding with the same pharyngeal pump frequencies, reflecting a similar motivation. Interesting, when ants fed on a low sucrose concentration and increased viscosity solution (10 T), their pump frequencies dropped rapidly respect to the pure-sucrose solution (50). On the contrary, pump frequencies for 50 and 50 T remained similar until the end of the intake. Since the pump frequency is strongly modulated by the ant motivation, an increase in viscosity with low sucrose content, demotivates the ants rapidly, suggesting a rapid integration of different kinds of information about the food value. Our results helped to understand how nectivorous ants could modulate their foraging decision-making.Sucrose is a commonly used stabilizing excipient in protein formulations. However, recent studies have indicated the presence of nanoparticulate impurities (NPIs) in the size range of 100-200 nm in pharmaceutical-grade sucrose. Furthermore, isolated NPIs have been shown to induce protein aggregation when added to monoclonal antibody formulations. Moreover, nanoparticles are popular vaccine delivery systems used to increase the immunogenicity of antigens. Therefore, we hypothesized that NPIs may have immunostimulatory properties. In this study, we evaluated the immunomodulatory effects of NPIs in presence and absence of trastuzumab in vitro with monocyte-derived dendritic cells (moDCs). Exposure of trastuzumab, the model IgG used in this study, to NPIs led to an increase in concentration of proteinaceous particles in the sub-micron range. When added to moDCs, the NPIs alone or in presence of trastuzumab did not affect cell viability or cytotoxicity. Moreover, no significant effect on the expression of surface markers, and cytokine and chemokine production was observed. Our findings showed, surprisingly, no evidence of any immunomodulatory activity of NPIs. As this study was limited to a single IgG formulation and to in vitro immunological read-outs, further work is required to fully understand the immunogenic potential of NPIs.
