Intraperitoneal ceftriaxone administration in healthy horses results in high and prolonged peritoneal concentrations. Recent findings suggest that intraperitoneal ceftriaxone might increase survival rates in horses affected by peritonitis. The present study aimed to evaluate plasma and peritoneal concentrations of ceftriaxone after intraperitoneal administration in horses with septic peritonitis. Twenty-six horses presenting clinical, laboratorial, and sonographic findings compatible with the disease were included. All horses received daily intraperitoneal ceftriaxone (25 mg/kg bwt) in addition or not with other antibiotics and support therapies. High-performance liquid chromatography was used to determine plasma and peritoneal ceftriaxone concentrations before and after 12 and 24 hours of ceftriaxone administration. Mean plasma concentrations 12 and 24 hours after administration were, respectively, 1.84 ± 0.43 and 0.37 ± 0.07 μg/mL, and mean peritoneal concentrations were 5.7 ± 2.84 and 0.42 ± 0.13 μg/mL. Ceftriaxone concentration was lower in comparison with previous studies in healthy horses and presented under the minimal inhibitory concentration for enterobacteria (≤1 μg/mL) and for gram-positive isolates (≤0.5 μg/mL) at 24 hours. The variation of the results obtained between healthy horses and with septic peritonitis demonstrated that pharmacokinetics/dynamics are different between these patients and suggests the use of an interval of dose of 12 hours.A significant portion of equine lameness is localized to the stifle joint. Effective cartilage repair strategies are largely lacking, however, recent advances in surgical techniques, biomaterials, and cellular therapeutics have broadened the clinical strategies of cartilage repair. To date, no studies have been performed directly comparing neonatal and adult articular cartilage from the stifle across multiple sites. An understanding of the differences in properties between the therapeutic target cartilage (i.e., adult cartilage) as well as potential donor cartilage (i.e., neonatal cartilage) could aid in selection of optimal harvest sites within a donor joint as well as evaluation of the success of the grafted cells or tissues within the host. Given the dearth of characterization studies of the equine stifle joint, and in particular neonatal stifle cartilage, the goal of this study was to measure properties of both potential source tissue and host tissue. Articular cartilage of the distal femur and patella (Pagen per wet weight was found to be significantly higher in adult cartilage than neonatal when averaged across all levels. In terms of biomechanical properties, aggregate modulus varied significantly across the condyles of adult cartilage but not the neonate. Neonatal cartilage was significantly less permeable, and the Young's modulus of neonatal cartilage was significantly higher than the adult. The tensile strength did not vary in a statistically significant manner between age groups. An understanding of morphological, histological, biochemical, and biomechanical properties enhances the understanding of cartilage tissue physiology and structure-function relationships. This study revealed important differences in biomechanical and biochemical properties among the 17 sites and among the six joint regions, as well as age-related differences between neonatal and adult cartilage. These location and age-related variations are informative toward determining the donor tissue harvest site.The objective of the present study was to evaluate the probiotic properties, security and antibacterial ability in vivo of isolated strains from healthy equine. In the present study, two Pediococcus acidilactici (P1 and P2) and two Lactobacillus equi (L1 and L2) were isolated. All isolates were died when exposed to pH 2.0 for 3 hours but survived at pH 3.0 and pH 4.0 with differential survival rate, and there is a higher survival rate at pH 4.0. Similarly, the isolates showed different tolerance to bile. The viable bacteria count was sustained at high levels in a tolerance test with artificial gastrointestinal fluid. The isolates survived and grew at temperatures between 37 and 55°C but died at 65°C. Four strains exhibited inhibitory activity against pathogens, including Salmonella typhimurium (CVCC542), Escherichia coli (C83902), Staphylococcus aureus (BNCC186335), and Pasteurella multocida (clinical isolate). These isolates exhibited differential antibiotic susceptibility. In safety trials, all isolates were γ-hemolytic, and the oral toxicity of strains P1 (gavaged with 1 × 109 CFU/day) and L1 (gavaged with 1 × 109 CFU/day) were analyzed in mice. There were no effects on the overall health status of mice. There were no prominent differences in the incidence of bacteria translocation to blood, liver, and spleen. Mice gavaged with Pediococcus acidilactici P1 (1 × 108 CFU/day) or Lactobacillus equi L1 (1 × 108 CFU/day) as prevention showed lower rates of diarrhea and mortality after being challenged with Salmonella typhimurium (4 × 106 CFU signal dose, 0.1 mL by intragastric gavage). The results indicate that the isolated strains could act as potential probiotics, providing a new way to reduce salmonella infection, which merit future application studies.Argentine Polo (AP) is a young horse breed with a fast expansion because of an open registry policy and the application of assisted reproduction technologies. However, these practices could have affected the genetic variability of the breed. To monitor these potential changes, a parentage panel of 11-15 microsatellites was investigated for changes in allele frequencies, heterozygosity, and exclusion probability over a 16 year period. Frequency of 36 alleles in 13 markers showed significant departures from expected changes because of random genetic drift. click here Six markers showed both allelic frequency increase and expected heterozygosity (He) reduction, suggesting the influence of selective breeding or hitchhiking effects.
