BACKGROUND Metabolically healthy obese patients accounts for a large part of obese population, but its clinical significance and cardiac dysfunction are often underestimated. The microRNA profiles of metabolically healthy obese patients were investigated in the study, and the selected microRNA (miRNA) based on our microarray assay will be further verified in a relatively large metabolically healthy obese population. METHODS microRNA microarray was performed from six metabolically healthy obese and 6 health control blood samples. Based on the bioinformatics analysis, we further measured RT-PCR, fibrosis markers, echocardiograms, and TGF-β1/Smad signaling pathway in 600 metabolically healthy obese population. RESULTS We found that miRNAs expression characteristics in metabolically healthy obese groups were markedly different from healthy control group. MiRNA-21 was significantly increased in the samples of metabolically healthy obese patients. Besides, miRNA-21 levels were associated with cardiac fibrosis marker. Meanwhile, higher miRNA-21 levels were related to elevated E/E'. Besides, patients with the highest miRNA-21 quartile showed the lowest ratio of E/A. These associations between miRNA-21 and diastolic function parameters were independent of obesity and other confounding variables. Of note, TGF-β1and Smad 3 were significantly upregulated while Smad 7 was downregulated according to the miRNA-21 quartiles in metabolically healthy obese group. CONCLUSIONS We demonstrated the profiles of circulating microRNAs in metabolically healthy obese patients. Increased plasma miRNA-21 levels were related to impaired diastolic function independent of other relevant confounding variables. MiRNA-21 could be one of the mechanistic links between obesity and diastolic dysfunction through regulating cardiac fibrosis via TGF-β1/Smad signaling pathway in obese hearts, which may serve as a novel target of disease intervention. © 2020 The Authors. However, there is limited information about long-term outcome of HSCT and its effect to protect against cancer development in DOCK8-deficient patients. In this study, we retrospectively evaluated clinical and immunologic characteristics of 20 DOCK8-deficient patients and outcome of 11 patients who underwent HSCT. This research aims to investigate the role of SES on the dietary energy density (ED) in Australian women of preconception age. METHODS A secondary analysis of the Australian National Nutrition and Physical Activity Survey 2011-12 for females aged 18-39 years (n = 1617) was conducted. find more Dietary intake was assessed by 24-hr recalls and dietary ED by dietary energy per weight (kJ.g-1 ). ED was further categorised as ED of foods and beverages separately. SES was assessed by three variables Socio-Economic Indexes for Areas (SEIFA), developed by the Australian Bureau of Statistics; income decile; and level of education. Linear mixed model regressions were used to identify associations between ED and SES. RESULTS The median ED for food, beverages and combined food and beverages was 9.38 kJ g-1 , 1.02 kJ g-1 and 7.11 kJ g-1 , respectively. No significant variation was explained by SES variables when analysing combined ED in the adjusted model or ED from foods. Income decile reduced ED of beverages, although with little effect (coefficient -0.04, P = 0.002). Significant confounders included inactivity, which increased ED in both combined ED and ED foods (coefficient 0.51, P = 0.001 and coefficient 0.78, P less then 0.001). CONCLUSIONS SES explained little variation in dietary ED in women of childbearing age. A large proportion of women had high energy-dense diets regardless of their SES. These findings suggest that a large proportion of women, who may become pregnant, have diets that exceed the international recommendations for dietary energy density. © 2020 The British Dietetic Association Ltd.Most obese patients develop hyperleptinaemia. Leptin, mainly produced by adipocytes, demonstrates a promotional role in liver fibrosis. Hepatic stellate cell (HSC) activation, a key step in liver fibrogenesis, requires global reprogramming of gene expression. The remodeling of DNA methylation is a mechanism of the epigenetic regulation of gene expression. The biosynthesis of S-adenosylmethionine, a principle biological methyl donor, is catalyzed by methionine adenosyltransferase (MAT) such as MATⅡ which has been shown to promote HSC activation in vitro. This study was mainly aimed to determine the effect of leptin on MAT2A expression (the catalytic subunit of MATⅡ) in HSCs. Results showed that MAT2A knockdown reduced leptin-induced HSC activation and liver fibrosis in the leptin-deficient mouse model. Leptin promoted MAT2A expression in HSCs and increased MAT2A promoter activity. The axis of the β-catenin pathway/E2F-4 mediated the effect of leptin on MAT2A expression. Leptin-induced β-catenin signaling reduced E2F-4 expression and thus abated E2F-4 binding to MAT2A promoter at a site around -2779 bp, leading to an increase in the MAT2A promoter activity. These data might shed more light on the mechanisms responsible for liver fibrogenesis in obese patients with hyperleptinaemia. © 2020 Federation of American Societies for Experimental Biology.
