We retrospectively reviewed the records of 75 collegiate athletes from a single institution who had undergone unilateral or bilateral ACL reconstruction between 2001 and 2013 and participated in an extensive supervised rehabilitation program. Prospectively collected athlete data as well as data about preinjury exposure, associated lesions, surgical technique, time lost to injury, number of games missed, time to return to full sport activity or retire, and subsequent surgical procedures were extracted fr that reported in other studies in the literature. A total of 71 patients (75 knees) were retrospectively evaluated after MSC implantation with concomitant HTO. Clinical and radiological outcomes were evaluated, and magnetic resonance imaging (MRI) was used to assess cartilage regeneration. Statistical analyses were performed to determine the effect of different factors on clinical, radiographic, and MRI outcomes.

The aim of this study is to design, fabricate and determine the cytotoxic effects of dual loaded paclitaxel and 17-AAG in stealth polymeric nanoparticles. The nanoparticles were fabricated by dispersion polymerization.

Two breast cancer cell lines (MCF-7 and SKBR-3) were cultured and treated with media only, blank nanoparticles, paclitaxel (as a free drug), 17-AAG (free drug), paclitaxel + 17-AAG combination (as free drugs), and paclitaxel + 17-AAG combination loaded in poly-ε-caprolactone stealth nanoparticles. Each drug in the combination was half the concentration of the single free drug.

The cytotoxic effects of the paclitaxel treatment and that of the combination (free drug) were found to be similar in both SKBR3 and MCF7 cell lines. Similar cytotoxic effects were observed for the drug combination both in the drug loaded nanoparticles formulation and in free drug form for both cell lines.

Both paclitaxel and 17-AAG were effectively loaded and released from the polymeric nanoparticles. Paclitaxel (free drug), paclitaxel-17AAG combination (free drug), and dual drug-loaded nanoparticles had similar cytotoxic effects on both cell lines. Paclitaxel and 17-AAG combination resulted in synergistic effect paclitaxel in the combination with 17-AAG was half its original concentration and yielded similar cytotoxic effect. The cytotoxic effect of CAPE on MCF-7 cell line was found with an XTT analysis. Total RNA was isolated from the cells exposed to IC50 dose and untreated control cells. Expressions of genes related to cell cycle control (CCND2, RB1, ATM, CDC34, CDK5RAP1) were evaluated by qRT-PCR by the LightCycler 480 System (Roche). GAPDH and ACTB housekeeping genes were used for the normalization of gene expressions. IC50 value of CAPE in MCF-7 cells was calculated as 75µM. It was shown that IC50 dose of CAPE induced significant upregulation in expressions of cell cycle control genes, compared to control cells. CAPE increases the expression of genes that are important in cell cycle control, suggesting that this component can be used as an effective chemopreventive agent in breast cancer cells.
