People with type 1 diabetes have an increased risk of developing microvascular complications, which have a negative impact on the quality of life and reduce life expectancy. Numerous studies in animals with experimental diabetes show that c-peptide supplementation exerts beneficial effects on diabetes-induced damage in peripheral nerves and kidneys. There is substantial evidence that c-peptide counteracts the detrimental changes caused by hyperglycemia at the cellular level, such as decreased activation of endothelial nitric oxide synthase and sodium potassium ATPase, and increase in formation of pro-inflammatory molecules mediated by nuclear factor kappa-light-chain-enhancer of activated B cells cytokines, chemokines, cell adhesion molecules, vascular endothelial growth factor, and transforming growth factor beta. However, despite positive results from cell and animal studies, no successful c-peptide replacement therapies have been developed so far. Therefore, it is important to improve our understanding of the impact of c-peptide on the pathophysiology of microvascular complications to develop novel c-peptide-based treatments. This article aims to review current knowledge on the impact of c-peptide on diabetic neuro- and nephropathy and to evaluate its potential therapeutic role.Coxiella burnetii, a cause of infection in humans and several animal species, is transmitted through inhalations and oral route but also tick bites. Its spreading in ruminants has been largely investigated, whereas data about the occurrence of this infection in canine population are scanty. In this retrospective study, blood serum samples of 516 dogs were tested by indirect immunofluorescence assay to detect antibodies against C. burnetii; 42 (8.13%) were positive with titers ranging from 164 to 1512. The highest seroprevalences were detected in dogs aged > 5 years, employed in hunting activity and living in a peri-urban/rural environment. Diagnosis for C. burnetii infection should be always carried out in bitches with reproductive disorders. Moreover, in view of the zoonotic impact of this infection, asymptomatic dogs exposed to ticks' bites and/or to contact with infected farm animals should be checked, too.Due to their need for living cells, viruses have developed adaptive evolutionary strategies to survive and perpetuate in reservoir hosts that play a crucial role in the ecology of emerging pathogens. Pathogenic and potentially pandemic betacoronaviruses arose in humans in 2002 (SARS-CoV, disappeared in July 2003), 2012 (MERS-CoV, still circulating in Middle East areas), and 2019 (SARS-CoV-2, causing the current global pandemic). As universally recognized, bats host ancestors of the above-mentioned zoonotic viruses. However, hedgehogs have been recently identified in Europe and Asia as possible reservoirs of MERS-CoV-like strains classified as Erinaceus coronavirus (EriCoV). To elucidate the evolution and genetics of EriCoVs, NGS (next generation sequencing) and Sanger sequencing were used to examine fecal samples collected in Northern Italy in 2018/2019 from 12 hedgehogs previously found EriCoV-positive by RT-PCR. Interestingly, eight viral strains presented an additional ORF encoding for the CD200 ortholog located between the genes encoding for the Spike and the ORF3a proteins. Dorper and SC lambs (4-5 months of age) were subjected to cyclic heat stress (HS) (28-40 °C). The temperature was increased to 38-40 °C between 800 and 1700 h daily and maintained at 28 °C for the remainder of the day (30-60% relative humidity (RH)) in climatic chambers for 2 weeks (n = 12/group), with controls maintained in a thermoneutral (TN) (18-21 °C, 40-50% RH) environment (n = 12/group). Basal respiration rate (RR), rectal temperature (RT) and skin temperature (ST) were higher (p less then 0.01) in SC lambs than in Dorpers. HS increased RR, RT and ST (p less then 0.01) in both genotypes, but the levels reached during HS were lower (p less then 0.01) in Dorpers. HS increased (p less then 0.01) water intake to a greater extent in SC lambs, while feed intake was reduced (p less then 0.05) by HS in SC lambs but not in Dorpers. HS increased (p less then 0.01) blood urea nitrogen and creatinine in SC lambs only. Plasma non-esterified fatty acid concentrations were reduced (p less then 0.05) by HS in SC lambs but increased (p less then 0.05) in Dorpers. There was no effect of HS on pO2, cHCO3- and cSO2, but higher (p less then 0.01) blood pH and lower (p less then 0.01) pCO2 were recorded under HS in both genotypes. Blood electrolytes and base excess were reduced (p less then 0.01) under HS, while a genotype difference (p less then 0.05) was only observed in blood K+ and hemoglobin concentrations. Basal plasma prolactin concentrations were lower (p less then 0.01) in Dorpers but were elevated at a similar level during HS (p less then 0.01) in both genotypes. Dorper lambs are more resilient to HS than SC lambs. The aim of the present study was to compare the effectiveness of the addition of aerobic exercises performed in an unloaded or loaded position to a conventional exercise program in overweight subjects with knee osteoarthritis. Twenty-four subjects were randomly allocated to receive 36 sessions of 30-min duration of either sitting aerobic exercises (experimental group) or standing aerobic exercises (control group). Pain intensity, knee disability, and quality-of-life data were collected at baseline and at 12, 24, and 36 sessions. Generalized linear mixed models (GLMMs) were constructed for the analysis of the differences. Significant differences were found in the experimental group for self-reported pain and knee pain and disability at 24 and 36 sessions (p less then 0.05). Significant between-group differences were observed in change in self-reported knee pain and disability and quality of life from baseline to 24th- and 36th-session measurements in favor of the experimental group.
