Left SFGdor nodal efficiency was correlated with Repetitive Behavior and Communication, two core symptoms of autism. Numerous studies have long appreciated the diversity of these patterns, characterizing their differences in spatial and temporal dynamics. In the murine somatosensory cortex, neuronal co-activation is thought to guide the formation of sensory maps and prepare the cortex for sensory processing after birth. While pioneering studies deftly utilized slice electrophysiology and unit recordings to characterize correlated activity, a detailed understanding of the underlying circuits remains poorly understood. More recently, advances in in vivo calcium imaging in awake mouse pups and increasing genetic tractability of neuronal types have allowed unprecedented manipulation of circuit components at select developmental timepoints. These novel approaches have proven fundamental in uncovering the identity of neurons engaged in correlated activity during development. In particular, recent studies have highlighted interneurons as key in refining the spatial extent and temporal progression of patterned activity. Here, we discuss how emergent synchronous activity across the first postnatal weeks is shaped by underlying gamma aminobutyric acid (GABA)ergic contributors in the somatosensory cortex. Further, the importance of participation in specific activity patterns per se for neuronal maturation and perdurance will be of particular highlight in this survey of recent literature. Finally, we underscore how aberrant neuronal synchrony and disrupted inhibitory interneuron activity underlie sensory perturbations in neurodevelopmental disorders, particularly Autism Spectrum Disorders (ASDs), emphasizing the importance of future investigative approaches that incorporate the spatiotemporal features of patterned activity alongside the cellular components to probe disordered circuit assembly.Evidence suggests that γ-aminobutyric acid (GABA) receptors are involved in the development of drug dependence. Considering its exclusively extrasynaptic localization, GABA receptor delta subunit (GABRD) is likely involved in heroin addiction. The purpose of this study was to explore the association between the single nucleotide polymorphisms (SNPs) of GABRD and heroin addiction. Genotyping of five SNPs (rs13303344, rs4481796, rs2376805, rs2229110, and rs41307846) in GABRD gene was performed by using TaqMan SNP assay. The association between heroin addiction and these SNPs was assessed in 446 heroin dependent patients and 400 normal control subjects of male Han Chinese origin. Only the genotype and allele frequencies at rs13303344 differed significantly between the cases and controls (nominal P values were 0.028 and 0.019, respectively). selleck chemicals llc The C allele of rs13303344 was associated with an increased risk of heroin addiction (OR = 1.281, 95 % CI 1.042-1.575). After Bonferroni correction, the association lost significance. The frequencies of the haplotype C-C-A and A-C-A at GARBD (rs13303344-rs4481796- rs2376805) differed significantly between the cases and controls. The heroin craving score was significantly higher in patients with CC/AC genotypes at rs13303344 than in those with the AA genotype (nominal P = 0.017). The results suggest that GABRD rs13303344 may contribute to the susceptibility to heroin addiction and is associated with the drug cravings of heroin dependent patients.Corticospinal excitability in humans can be facilitated during imagination and/or observation of upper-limb motor tasks. However, it remains unclear to what extent facilitation levels may differ from those elicited during execution of the same tasks. Twelve able-bodied individuals were recruited in this study. Motor evoked potentials (MEPs) in extensor carpi radialis (ECR) and flexor carpi radialis (FCR) muscles were elicited through transcranial magnetic stimulation of the primary motor cortex during (i) rest; (ii) wrist extension; and (iii) wrist flexion. Responses were compared between (1) motor imagery combined with virtual action observation (MI + AO; first-person virtual wrist movements shown on a computer display, while participants remained at rest and imagined these movements); and (2) motor execution (ME; participants extended or flexed their wrist). During MI + AO, ECR MEPs were facilitated during the extension phase but not the flexion phase, while FCR MEPs were facilitated during the flexion phase but not extension phase, compared to rest. During the ME condition, same, but greater, modulations were shown as those during MI + AO, while background muscle activities were similar in the rest phase as during extension and flexion phase in the MI + AO condition. Our results demonstrated that kinesthetic MI that included imagination and observation of virtual hands can elicit phase-dependent muscles-specific corticospinal facilitation of wrist muscles, consistent to those during actual hand extension and flexion. Moreover, we showed that MI + AO can contribute considerably to the overall corticospinal facilitation (∼20 % of ME) even without muscle contractions. These findings support utility of computer graphics-based motor imagery, which may have implications for rehabilitation and development of brain-computer interfaces.
Cognitive frailty (CF) is associated with dementia and disability. It was initially proposed in 2013 by the International Institute of Nutrition and Aging and the International Geriatrics Association. Over the years, there have been many emerging definitions e.g., Motoric Cognitive Risk Syndrome (MCR), Physio-cognitive Decline Syndrome (PCDS), reversible CF and potentially reversible CF.
Our objective was to determine the demographics including physical, functional, and psychosocial factors for the four CF definitions amongst community-dwelling older adults. In addition, the effect of dual-task exercise on the reversibility of different definitions of CF was also studied.
Participants were community-dwelling older adults aged ≥60years recruited between August 2017 and December 2019. Older adults screened to be frail, prefrail or have cognitive impairment were invited to participate in dual-task exercise program called HAPPY (Healthy Ageing Promotion Program for You). Improvement in cognition, frailty, and physical performance after a 3-month dual-task exercise intervention program was compared with controls.
