However, the underlying pathological mechanism for AWE still needs to be further studied.Aim To compare free and nanoparticle (NP)-encapsulated epigallocatechin-3-gallate (EGCG) for the treatment of Huntington's disease (HD)-like symptoms in mice. Materials & methods EGCG was incorporated into PEGylated poly(lactic-co-glycolic) acid NPs with ascorbic acid (AA). HD-like striatal lesions and motor deficit were induced in mice by 3-nitropropionic acid-intoxication. EGCG and EGCG/AA NPs were co-administered and behavioral motor assessments and striatal histology performed after 5 days. Results EGCG/AA NPs were significantly more effective than free EGCG in reducing motor disturbances and depression-like behavior associated with 3-nitropropionic acid toxicity. EGCG/AA NPs treatment also mitigated neuroinflammation and prevented neuronal loss. Conclusion NP encapsulation enhances therapeutic robustness of EGCG in this model of HD symptomatology. Together with our previous findings, this highlights the potential of EGCG/AA NPs in the symptomatic treatment of neurodegenerative diseases.Photoionization mass spectrometry (PI-MS) is a powerful and highly sensitive analytical technique for online monitoring of volatile organic compounds (VOCs). However, due to the large difference of PI cross sections for different compounds and the limitation of photon energy, the ability of lamp-based PI-MS for detection of compounds with low PI cross sections and high ionization energies (IEs) is insufficient. Although the ion production rate can be improved by elevating the ion source pressure, the problem of generating plenty of cluster ions, such as [MH]+·(H2O) n (n = 1 and 2) and [M2]+, needs be solved. In this work, we developed a new nonuniform electric field high-pressure photoionization/photoionization-induced chemical ionization (NEF-HPPI/PICI) source with the abilities of both HPPI and PICI, which was accomplished through ion-molecule reactions with high-intensity H3O+ reactant ions generated by photoelectron ionization (PEI) of water molecules. By establishing a nonuniform electric field in a three-zone ionization region to enhance in-source declustering and using 99.999% helium as the carrier gas, not only the formation of cluster ions was significantly diminished, but the ion transmission efficiency was also improved. Consequently, the main characteristic ion for each analyte both in HPPI and PICI occupied more than 80%, especially [HCOOH·H]+ with a yield ratio of 99.2% for formic acid. The analytical capacity of this system was demonstrated by operando monitoring the hydrocarbons and oxygenated VOC products during the methanol-to-olefins and methane conversion catalytic reaction processes, exhibiting wide potential applications in process monitoring, reaction mechanism research, and online quality control.The cluster-in-molecule (CIM) local correlation approach with an accurate distant pair correlation energy correction is presented. For large systems, the inclusion of distant pair correlation energies is essential for the accurate prediction of absolute correlation energies and relative energies. Here, we propose a simple and efficient scheme for evaluating the distant pair correlation energy correction for the CIM approaches. The corrections can be readily extracted from electron correlation calculations of clusters with almost no additional effort. Benchmark calculations show that the improved CIM approach can recover more than 99.94% of the correlation energy calculated by the parent method. By combining the CIM approach with the domain-based local pair natural orbital (DLPNO) local correlation approach, we have provided accurate binding energies at the CIM-DLPNO-CCSD(T) level for a test set consisting of eight weakly bound complexes ranging in size from 200 to 1027 atoms. With these results as the reference data, the accuracy and applicability of other electron correlation methods and a few density functional methods for large systems have been assessed.α-Asarone and β-asarone are reported as bioactive constituents of Acorus calamus. Phase I metabolism of asarone isomers results in a multiple spectrum of genotoxic metabolites. Thus, the question arises whether structural analogues of the known phase I metabolites also naturally occur in A. calamus-based food products. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for three product classes, herbal infusions, alcoholic beverages, and food supplements. High asarone contents were detected in herbal infusions (total mean 9.13 mg/kg, n = 8) and food supplements (total mean 14.52 mg/kg, n = 6); hence, these food products can highly contribute to human exposure to genotoxic asarone derivatives. Also, the occurrence of asarone oxidation products found in food and food supplements has to be taken under consideration because data on toxicity is limited so far.Emerging evidence suggests that chronic exposure to aristolochic acids (AAs) is one of the etiological pathways leading to chronic kidney disease (CKD). Due to the traditional practice of herbal medicine and AA-containing plants being used extensively as medicinal herbs, over 100 million East Asians are estimated to be at risk of AA poisoning. Given that the chronic nephrotoxicity of AAs only manifests itself after decades of exposure, early diagnosis of AA exposure could allow for timely intervention and disease risk reduction. However, an early detection method is not yet available, and diagnosis can only be established at the end stage of CKD. GNE049 The goal of this study was to develop a highly sensitive and selective method to quantitate protein adducts of aristolochic acid I (AAI) as a biomarker of AA exposure. The method entails the release of protein-bound aristolactam I (ALI) by heat-assisted alkaline hydrolysis, extraction of ALI, addition of internal standard, and quantitation by liquid chromatography-tandem mass spectrometric analysis.
