Progesterone receptor (PgR) inhibits cell proliferation in pancreatic neuroendocrine neoplasms (PanNEN). In non-neoplastic pancreas, loss of PgR induces β-cell proliferation and insulin production. However, detailed association between PgR and insulin producing PanNENs is poorly understood. Insulin, proinsulin, and PgR were immunolocalized in 82 PanNENs (54 non-functioning PanNENs NF-PanNENs and 28 insulinomas). The status of immunoreactivity was compared to the clinicopathological factors of the patients. Immunoreactivity was also confirmed by employing the double-immunohistochemistry. These results were also compared with those in non-neoplastic Langerhans islets. PgR immunoreactivity was significantly higher in insulinomas than that in NF-PanNENs (p less then 0.001). Insulin and proinsulin immunoreactivity was also detected in 20 (37 %) of (single cell) insulin positive NFs (Inspos-NF-PanNEN), in which PgR expression was higher than in insulin negative NF-PanNENs (Insneg-NF-PanNEN, p = 0.03). The ratio of PgR-insulin double positive cells to overall insulin positive cells, as well as PgR-proinsulin double positive cells to proinsulin positive cells, was detected to the same degree in insulinoma (PgR-insulin 70 %, PgR-proinsulin 66 %), Inspos-NF-PanNENs (PgR-insulin 65 %, PgR-proinsulin 68 %) and normal islet (PgR-insulin 80 %, PgR-proinsulin 72 %). PgR and insulin expressing cells colocalize in tumor cells of the PanNENs regardless of the hormone-related symptoms of the patients. Inhibitory effect of PgR on tumor cells might be associated with the favourable clinical outcome of insulinoma patients.Adrenal corticosterone plays crucial roles in energy metabolism and immuno-reactivity throughout the body. As we have previously shown that corticosterone biosynthesis in C2C12 myoblasts, we study about corticosterone biosynthesis in rat skeletal muscles. It was found that enzymatic activities producing corticosterone and testosterone except the activity of P450scc in rat skeletal muscle as like as C2C12 cells. The CYP11B mRNA encoding cytochrome P45011β that mediates 11-deoxycorticosterone hydroxylase activity, producing corticosterone was expressed in skeletal muscles. In immunoblotting analysis, cytochrome P45011β protein was expressed in rat muscles and whole organs especially higher levels in adrenal and brain. The localizations of corticosterone content and enzymatic activities involved in the production of corticosterone were preferentially observed in gastrocnemius fibers rather than in soleus fibers. The immunohistochemical analysis showed that the fast-twitch or type II muscle fibers positive to antibody against fast myosin heavy chain were preferentially stained with anti-cytochrome P45011β antibody in the gastrocnemius fiber. In addition, we detected corticosterone biosynthesis from pregnenolone sulfate conjugates in perfusion of the rat hindquarter. Corticosterone is synthesized in rat skeletal muscles and the biosynthesis was localized in the fast-twitch or type II muscle fibers. We speculated that the local synthesized corticosterone might be involved in glucocorticoid-induced muscle atrophy that preferentially occurs in fast muscle fibers, and the initial substrate of the local CORT biosynthesis were supported to be performed from the conjugates such as pregnenolone sulfate circulating in the blood flow.Objectives It has been reported that green tea exerts antibacterial, anti-inflammatory, and antioxidant effects. The purpose of the present study was to evaluate the effects of drinking green tea on bone resorption in ligature-induced periodontitis in mice. Methods Sixty C57BL/6 eight-week-old male mice were used. To induce periodontitis, a ligature was placed for 7 days around the upper left second maxillary molar. After ligature removal, the animals were administered different concentrations of green tea (1.5 g/60 mL, 3 g/60 mL, or 6 g/60 mL) or distilled water. At 1 and 2 weeks of administration, the animals were sacrificed and micro-CT images of the maxillae were taken. Next, the depth and area of alveolar bone loss in the buccal and palatal sides were measured. The number of inflammatory cells and osteoclasts in histological sections were counted. Results The result showed ligature-induced alveolar bone loss. Green tea inhibited ligature-induced bone loss in the buccal side in a dose-dependent manner. Histologically, ligature increased the number of inflammatory cells and osteoclasts, but this effect was alleviated by green tea. Conclusions Evidence from this animal experiment suggested that drinking green tea would be potentially beneficial to reduce alveolar bone loss in ligature-induced periodontitis.Gambel's Quail, Callipepla gambelii, are gregarious birds commonly found in the southwestern deserts of the United States and Northwestern Mexico. With expanding urbanization, these birds are often found in exurban and suburban areas where they have access to food sources that may differ from those used by birds living in rural habitats and, as a result, also differ morphologically and physiologically. Cenicriviroc cost To investigate this hypothesis, we compared the morphology and nutritional physiology of quail sampled at sites varying with respect to land use and cover. We hypothesized that quail living in more developed areas have access to a greater variety of and to more stable food resources, and predicted that morphology and nutritional physiology would be associated with degree of urbanization. We sampled adult birds at locations in the greater Phoenix metropolitan, Arizona (USA) area that vary with respect to land use and cover types. At the time of capture, birds were weighed and chest circumference was recorded. We also collected a blood sample from the jugular vein of each individual for analysis of plasma glucose, total proteins, triglycerides, and free glycerol. Consistent with the hypothesis, birds living in more developed environments had larger chest circumferences and higher circulating lipid concentrations than birds living in less developed areas, suggesting greater access to lipid-rich foods. In addition, the areal proportion of grass and lakes was negatively correlated to plasma free glycerol (r = -0.46, p = .031), and positively, but not significantly, correlated to plasma protein concentrations (r = 0.388, p = .073). These results suggest that quail living in areas with more grass have access to less dietary fats than urban birds. The findings are the first to indicate an association between urbanization and the morphology and nutritional physiology of Gambel's Quail, but further study using more and larger samples is needed before these findings can be generalized.
