Planning PC with families which encourages family-centred and individualised approach is also discussed.The test statistics underpinning several methods for combining p-values are special cases of generalized mean p-value (GMP), including the minimum (Bonferroni procedure), harmonic mean and geometric mean. A key assumption influencing the practical performance of such methods concerns the dependence between p-values. Approaches that do not require specific knowledge of the dependence structure are practically convenient. Vovk and Wang derived significance thresholds for GMPs under the worst-case scenario of arbitrary dependence using results from Robust Risk Analysis (RRA). Here I calculate significance thresholds and closed testing procedures using Generalized Central Limit Theorem (GCLT). GCLT formally assumes independence, but enjoys a degree of robustness to dependence. The GCLT thresholds are less stringent than RRA thresholds, with the disparity increasing as the exponent of the GMP ( r) increases. I motivate a model of p-value dependence based on a Wishart-Multivariate-Gamma distribution for the underlying log-likelihood ratios. In simulations under this model, the RRA thresholds produced tests that were usually less powerful than Bonferroni, while the GCLT thresholds produced tests more powerful than Bonferroni, for all r> - ∞. Above r> - 1, the GCLT thresholds suffered pronounced false positive rates. Above r> - 1/2, standard central limit theorem applied and the GCLT thresholds no longer possessed any useful robustness to dependence. I consider the implications of these results in the context of various interpretations of GMPs, and conclude that the GCLT-based harmonic mean p-value procedure and Simes' (1986) test represent good compromises in power-robustness trade-off for combining dependent tests.
Trauma and sepsis are individually two of the leading causes of death worldwide. When combined, the mortality is greater than 50%. Thus, it is imperative to have a reproducible and reliable animal model to study the effects of polytrauma and sepsis and test novel treatment options. Porcine models are more translatable to humans than rodent models due to the similarities in anatomy and physiological response. We embarked on a study to develop a reproducible model of lethal polytrauma and intra-abdominal sepsis, which was lethal, though potentially salvageable with treatment.

Our laboratory has a well-established porcine model that was used as the foundation. Animals were subjected to a rectus crush injury, long bone fracture, liver and spleen laceration, traumatic brain injury and hemorrhage that was used as a foundation. We tested various colon injuries to create intra-abdominal sepsis. All animals underwent injuries followed by a period of shock, then subsequent resuscitation.

All animals had blood culture-proven sepsis. Attempts at long-term survival of animals after injury were ceased because of poor appetite and energy. We shifted to an 8-hour endpoint. The polytrauma injury pattern remained constant and the colon injury pattern changed with the intention of creating a model that was ultimately lethal but potentially salvageable with a therapeutic drug. An uncontrolled cecal injury (n=4) group resulted in very early deaths. A controlled cecal injury (CCI; n=4) group had prolonged time prior to mortality with one surviving to the endpoint. OTX015 The sigmoid injury (n=5) produced a similar survival curve to CCI but no animals surviving to the endpoint.

We have described a porcine model of polytrauma and sepsis that is reproducible and may be used to investigate novel treatments for trauma and sepsis.

This study will provide useful molecular resources for clarifying evolutionary and phylogenetic confusion within Calappidea.Polygala japonica Houtt. (Polygalaceae) is a perennial herbaceous plant widely distributed in south China. Here, we assembled and characterized the complete chloroplast (cp) genome of P. japonica using sequencing data. The complete cp genome of P. japonica was 165,439 bp in length, with a GC content of 36.7%. The complete cp genome shows a typical quadripartite structure with a pair of inverted repeats (IRs) of 36,786 bp, separated by a large single copy region (LSC) of 83,722 bp, and a small single copy region (SSC) of 8,145 bp. A total of 135 genes were annotated in the cp genome of P. japonica, consisting of 89 protein-coding genes, 8 rRNA genes, and 38 tRNA genes. The phylogenetic analysis showed P. japonica had a closer relationship with P. tenuifolia.
