This paper addresses the prevalence of DNA of drivers, passengers, and unknown individuals in vehicles. Five vehicles with a regular driver only, and five vehicles with a regular driver and regular passenger have each been sampled at twenty locations. Based on the findings, we propose a sampling strategy for investigative purposes as well as for evaluative purposes when evaluating the findings given scenarios that propose the person-of-interest as either the driver or passenger in a vehicle.Conflict casualties refer to those individuals who are lost due to military conflict or war. The involvement of forensic archaeologists and anthropologists in the legal search, recovery, documentation, identification, and repatriation/reburial of conflict casualties is well known. Internationally, there are a number of professional organisations who ethically recover and identify these individuals. However, at the same time, some organisations and individuals have raised significant concerns about working in other countries, understanding specific laws and protocols, and how the whole recovery and identification process should be undertaken. Through this special issue, Forensic Science International is interested in promoting these investigative good-practice procedures, illustrated with case studies, and ethical and legal considerations when undertaking and disseminating these humanitarian missions to the wider forensic community. This Special Issue focuses primarily on the First and Second World Wars, yet other conflicts are covered, and includes the views and perspectives from different organisations within specific countries in the format of original papers, commentaries, and case reports. Specifically, these papers include the legislative regulations, information regarding the authorities to be consulted on and who deal with human remains, the organisations and professionals who are involved with the recovery and analysis of human remains, the process of identification, and how this information is disseminated to the public.Perceptual adaptation is an active cognitive process where listeners re-analyse speech categories based on new contexts/situations/talkers. It involves top-down influences from higher cortical levels on lower-level auditory processes. Individuals with congenital amusia have impaired pitch processing with reduced connectivity between frontal and temporal regions. This study examined whether deficits in amusia would lead to impaired perceptual adaptation in lexical tone perception. Thirteen Mandarin-speaking amusics and 13 controls identified the category of target tones on an 8-step continuum ranging from rising to high-level, either in isolation or in a high-/low-pitched context. For tones with no context, amusics exhibited reduced categorical perception than controls. While controls' lexical tone categorization demonstrated a significant context effect due to perceptual adaptation, amusics showed similar categorization patterns across both contexts. These findings suggest that congenital amusia impacts the extraction of context-dependent tonal categories in speech perception, indicating that perceptual adaptation may depend on listeners' perceptual acuity.HLA-G, a non-classical HLA molecule expressed by extravillous trophoblasts, plays a role in the maternal immune tolerance towards fetal cells. HLA-G expression is regulated by genetic polymorphisms in the 3' untranslated region (3'UTR). Low levels of HLA-G in the maternal circulation and placental tissue are linked to preeclampsia. Our objective was to investigate whether variants of the 3'UTR of the HLA-G gene in mother and fetus are associated with acute atherosis, a pregnancy specific arterial lesion of the decidua basalis that is prevalent in preeclampsia. Paired maternal and fetal DNA samples from 83 normotensive and 83 preeclamptic pregnancies were analyzed. We sequenced the part of the HLA-G 3'UTR containing a 14-bp insertion/deletion region and seven single nucleotide polymorphisms (SNPs). SJ6986 Associations with acute atherosis were tested by logistic regression. The frequency of heterozygosity for the 14-bp polymorphism (Ins/Del) and the +3142 SNP (C/G) variant in the fetus are associated with acute atherosis in preeclampsia (66.7 % vs. 39.6 %, p = 0.039, and 69.0 % vs. 43.4 %, p = 0.024). Furthermore, the fetal UTR-3 haplotype, which encompasses the 14-bp deletion and the +3142G variant, is associated with acute atherosis in preeclampsia (15 % vs. 3.8 %, p = 0.016). In conclusion, HLA-G polymorphisms in the fetus are associated with acute atherosis. We hypothesize that these polymorphisms lead to altered HLA-G expression in the decidua basalis, affecting local feto-maternal immune tolerance and development of acute atherosis.This study evaluated the in vitro modulatory effect of progesterone (PG) and vitamin D (VD) on NLRP1/NLRP3 inflammasomes and TLR4/NF-κB pathway in monocytes from pregnant women with preeclampsia (PE). Monocytes from 20 preeclamptic and 20 normotensive (NT) pregnant women, and THP-1 cells were cultured with/without hyaluronan (HA), PG, or VD to determine gene and protein expression of TLR4 receptor, phosphorylated NF-κB, IκBα, TLR4, MYD88, NF-κB, NLRP1, NLRP3, caspase-1, IL-1β, IL-18, TNF-α, and IL-10. Higher endogenous activation of inflammatory genes and higher protein expression of TLR4 and NF-κB was detected in monocytes of PE group and decreased after PG or VD treatment. Monocyte from PE stimulated with HA increased while treatment with PG or VD decreased the expression of genes and proteins related to the inflammasomes. THP-1 cells showed a similar immune response profile as monocytes from PE. These results demonstrate that PG and VD play an immunomodulatory role in monocyte activation.
