The effectiveness of hypochlorites (NaClO and Ca(ClO)2) on the reduction of antibiotic resistance genes (ARGs) during waste activated sludge (WAS) fermentation was determined by the quantitative PCR. NaClO and Ca(ClO)2 exhibited distinct effects on ARGs fates. Ca(ClO)2 was effective in removing all investigated ARGs, and the efficiency was highly dose-dependent. Unexpectedly, the NaClO treatment attenuated ARGs with lower efficiency and even caused the propagation of certain ARGs (i.e., aadA1 and tetQ) at higher doses. The extracellular polymeric substances dissolution and membrane integrity suggested that unstable NaClO had acute effects on bacteria initially, while it was ineffective to further attenuate ARGs released from hosts due to the rapid consumption of oxidative ClO-. Without lasting and strong oxidative stress, the microbial activities of tolerant ARGs hosts will partially recover and then contribute to the ARGs dissemination across genera. In contrast, solid-state Ca(ClO)2 was slowly released and exhibited prolonged effects on bacteria by disrupting cell membranes and removing the susceptible ARGs released from hosts. Furthermore, bacterial taxa-ARG network analysis indicated that Ca(ClO)2 reduced the abundance of potential hosts, and the metabolic pathway and gene expression related to ARGs propagation were significantly downregulated by Ca(ClO)2, which contributed to efficient ARGs attenuation.Here, straight upward Au nanowires (NWs) were successfully grown onto Fe3O4@TiO2 matrix through a seed-mediated strategy to intensively improve its photocatalysis and SERS performances, facilitating a peculiar recyclable surface-enhanced Raman spectroscopy (SERS)-based immunoassay of CA19-9 in liquid form based on visible light irradiation. Such immunoassay was also supported by a smart heterobifunctional cross-linking agent-mediated probe of anti-CA19-9/4-MBA without metal nanoparticles. A low limit of detection of 5.65 × 10-4 IUmL-1 and a wide linear range from 1000 to 0.001 IUmL-1 were demonstrated through repeated constructing the sandwich immunostructure with only one batch of nanocomposites. Moreover, the actual levels of CA19-9 for colorectal cancer patients were readily measured by the recyclable immunoassay, the results of which are principally consistent with the conventional CLIA detection. Thus, such a green strategy of visible light-induced recyclable immunoassay could be expected to have a potential practicability in the clinical diagnoses of cancer.The occurrence, dissemination and assembly processes of antibiotic resistance genes (ARGs) in urban water ecosystems are far from being understood. Here, we examined the diversity and abundance of ARGs in urban water ecosystems including landscape ponds, drinking water reservoirs, influents (IFs) and effluents (EFs) of wastewater treatment plants of a coastal city, China through high-throughput quantitative PCR. A total of 237 ARGs were identified, where multidrug, aminoglycoside and beta-lactamase resistance genes were the most abundant. Urban ponds had a comparatively high diversity and large numbers of shared ARGs with IFs and EFs. The average absolute abundance of ARGs (1.38 × 107 copies/mL) and mobile genetic elements (MGEs) (4.19 × 106 copies/mL) in ponds were only one order of magnitude lower than those of IFs, but higher than those of EFs and reservoirs. Stochastic processes dominated the ARG community assembly in IFs and ponds due to the random horizontal gene transfer caused by MGEs. These results imply that urban ponds are hotspots of ARGs. We further identified 25, 3, and 11 indicator ARGs for tracing the ARG contamination from IFs, EFs and ponds, respectively. Our study represents the first to highlight the role of urban ponds in the dissemination of ARGs.The growing threat of terrorism has triggered an urgent need to find effective ways to improve the analysis of explosives. This paper reports the identification of nitro organic explosives such as pentaerythritol tetranitrate (PETN), trinitrotoluene (TNT) and 2,4,6-trinitrophenylmethylnitramine (tetryl) in post-blast debris by 400 MHz nuclear magnetic resonance spectrometer.Radioactive contamination resulting from major nuclear accidents presents harsh environmental conditions. Inside the Chernobyl exclusion zone, even more than 30 years after the accident, the resulting contamination levels still does not allow land-use or human dwellings. To study the potential of basidiomycete fungi to survive the conditions, a field trial was set up 5 km south-south-west of the destroyed reactor unit. A model basidiomycete, the lignicolous fungus Schizophyllum commune, was inoculated and survival in the soil could be verified. Indeed, one year after inoculation, the fungus was still observed using DNA-dependent techniques. Growth led to spread at a high rate, with approximately 8 mm per day. This shows that also white-rot basidiomycetes can survive the harsh conditions in soil inside the Chernobyl exclusion zone. The unadapted fungal strain showed the ability to grow and thrive in the contaminated soil where both stress from radiation and heavy metals were present.Diclofenac (DCF) is one of the most commonly utilized non-steroidal anti-inflammatory drugs (NSAIDs), which is known to pose an ecotoxicological threat. In this study, from activated sludge and contaminated soil, we isolated four new bacterial strains able to degrade DCF under mono-substrate and co-metabolic conditions with glucose supplementation. We found that the effectiveness of DCF removal is strictly strain-specific and the addition of the primary substrate is not always beneficial. To assess the multidirectional influence of DCF on bacterial cells we evaluated the alterations of increasing concentrations of this drug on membrane structure. A significant increase was observed in the content of 170 cyclo fatty acid, which is responsible for reduced fluidity and profound changes in membrane rigidity. The cell injury and oxidative stress were assessed with biomarkers used as endpoints of toxicity, i.e. catalase (CAT), superoxide dismutase (SOD), lipids peroxidation (LPX), and both intra- and extracellular alkaline and acid phosphatase activity.
