Here, we report the effect of dielectric constant on electrical outputs of piezoelectric nanogenerator using ZnO/PDMS composites with varied ZnO coverages. The dielectric constant of piezoelectric layers was adjusted from 3.37 to 6.75. The electrical output voltage of 9 mV was achieved in the nanogenerator containing the ZnO/PDMS composite with the dielectric constant of 3.46, which is an 11.3-fold enhancement compared to the value of the nanogenerator featuring the composite with high dielectric constants. Significantly, lowering the dielectric constant of the piezoelectric layer improves the electrical output performance of piezoelectric nanogenerators.Vision-based 3D human pose estimation approaches are typically evaluated on datasets that are limited in diversity regarding many factors, e.g., subjects, poses, cameras, and lighting. However, for real-life applications, it would be desirable to create systems that work under arbitrary conditions ("in-the-wild"). To advance towards this goal, we investigated the commonly used datasets HumanEva-I, Human3.6M, and Panoptic Studio, discussed their biases (that is, their limitations in diversity), and illustrated them in cross-database experiments (for which we used a surrogate for roughly estimating in-the-wild performance). For this purpose, we first harmonized the differing skeleton joint definitions of the datasets, reducing the biases and systematic test errors in cross-database experiments. We further proposed a scale normalization method that significantly improved generalization across camera viewpoints, subjects, and datasets. In additional experiments, we investigated the effect of using more or less cameras, training with multiple datasets, applying a proposed anatomy-based pose validation step, and using OpenPose as the basis for the 3D pose estimation. The experimental results showed the usefulness of the joint harmonization, of the scale normalization, and of augmenting virtual cameras to significantly improve cross-database and in-database generalization. At the same time, the experiments showed that there were dataset biases that could not be compensated and call for new datasets covering more diversity. We discussed our results and promising directions for future work.Staphylococcus aureus (S. aureus) is a major human pathogen that requires new antibiotics with unique mechanism. A new pleuromutilin derivative, 14-O-[(4,6-Diamino-pyrimidine-2-yl) thioacetyl] mutilin (DPTM), has been synthesized and proved as a potent antibacterial agent using in vitro and in vivo assays. In the present study, DPTM was further in vitro evaluated against methicillin-resistant Staphylococcus aureus (MRSA) isolated from dairy farms and outperformed tiamulin fumarate, a pleuromutilin drug used for veterinary. Moreover, a murine skin wound model caused by MRSA infection was established, and the healing effect of DPTM was investigated. The results showed that DPTM could promote the healing of MRSA skin infection, reduce the bacterial burden of infected skin MRSA and decrease the secretion of IL-6 and TNF-α inflammatory cytokines in plasma. These results provided the basis for further in-depth drug targeted studies of DPTM as a novel antibacterial agent.Cisplatin is a chemotherapy drug that kills cancer cells by damaging their DNA. In human cells, this damage is repaired primarily by nucleotide excision repair. While cisplatin is generally effective, many cancers exhibit initial or acquired resistance to it. Here, we studied cisplatin resistance in a defined cell line system. We conducted a comprehensive genomic characterization of the cisplatin-sensitive A2780 ovarian cancer cell line compared to A2780cis, its resistant derivative. The resistant cells acquired less damage, but had similar repair kinetics. Genome-wide mapping of nucleotide excision repair showed a shift in the resistant cells from global genome towards transcription-coupled repair. By mapping gene expression changes following cisplatin treatment, we identified 56 upregulated genes that have higher basal expression in the resistant cell line, suggesting they are primed for a cisplatin response. More than half of these genes are novel to cisplatin- or damage-response. Six out of seven primed genes tested were upregulated in response to cisplatin in additional cell lines, making them attractive candidates for future investigation. As a reserve agent, it will and should be used primarily in critically ill patients in the upcoming years. Due to the novelty of the substance little data on the pharmacokinetics in critically ill patients with septic shock and renal failure (including continuous renal replacement therapy and cytokine adsorber therapy) is available. We performed therapeutic drug monitoring in a cohort of five patients treated with cefiderocol, to improve the knowledge on pharmacokinetics in this vulnerable patient population. As expected for a cephalosporin with predominantly renal elimination the maintenance dose could be reduced in patients with renal impairment or on continuous renal replacement therapy. The manufacturer's dosing instructions were sufficient to achieve a drug level well above the MIC. However, the addition of a cytokine adsorber might reduce serum levels substantially, so that in this context therapeutic drug monitoring and dose adjustment are recommended.
