29; P = 0.0154) or sub-optimal endometrial build-up (OR 4.46 to 5.31; P less then 0.0001). Transfer of slow-developing blastocysts (expressed by expansion) significantly increased the risk of ectopic pregnancy (OR 2.59; P = 0.0102). Conclusions Unfavourable uterine environment, including uterine pathologies, uterine or tubal surgery and suboptimal endometrial build-up were related to ectopic pregnancy. Low expansion grade of blastocysts was identified as an additional putative risk factor for ectopic pregnancy, indicating the importance of proper embryonal-maternal synchronization. The overall ectopic pregnancy rate after blastocyst transfer was low, comparable with reported ectopic pregnancy rates in spontaneous conceptions. Proper evaluation of tubal and uterine pathologies, optimizing endometrial preparation and the transfer of expanded blastocysts in a frozen embryo transfer cycle, might be beneficial.Antimicrobial peptides are proving to be promising lead compounds for therapeutics. The major disadvantage of antimicrobial peptides is their proteolytic instability in the body with half-lives averaging less than an hour. Peptoids, or N-substituted glycines, have emerged as a promising field of peptidomimetics by retaining the beneficial properties of antimicrobial peptides while improving on their stability. Herein we evaluate peptoid derivatives of ultra-short lipophilic antimicrobial peptides, comparing their potency side-by-side against the most prevalent multidrug resistant bacteria (ESKAPE) and yeast pathogens (Candida albicans and Cryptococcus neoformans). Both peptide and peptoid counterparts were most effective against Gram-positive bacteria with minimum inhibitory concentration (MIC) values as low as 1.6 and 6.3 µg/mL, respectively. MYCMI-6 molecular weight In general, peptides retained better antimicrobial activity than their peptoid counterparts, however, certain peptoids proved to be more effective than peptides against Gram-negative bacteria. For example, peptoid MG10 displayed an MIC of 6.3 µg/mL against Pseudomonas aeruginosa compared to the peptide counterpart with an MIC of 100 µg/mL. All compounds tested were more potent against C. neoformans compared to C. albicans. Cytotoxicity analysis indicated that in general, peptoids were slightly less toxic than their peptide counterparts. Additionally, trypsin rapidly degraded one of the peptides evaluated while having no effect on comparable peptoids, demonstrating the proteolytic stability of peptoids.Adenovirus (HAdV) infection may have an important clinical impact in the immunosuppressed population with high morbidity and mortality rates. The lack of a specific, safe and effective antiviral treatment against HAdV makes necessary the search for new therapeutic alternatives. The aim of this study was to evaluate the in vitro activity of ganciclovir (GCV) against HAdV in co-infection by cytomegalovirus (HCMV) and HAdV in cellular cultures. Quantitative real-time PCR (qPCR) was used to measure HAdV and HCMV DNA replication efficiency in monocultures and in coinfection situations in the presence of both cidofovir and ganciclovir. The effects of GCV and cidofovir (CDV) were also evaluated in a burst assay for both viruses, alone and in combination. GCV decreased in 1-log the HAdV DNA replication efficiency in co-infection with HCMV compared with its activity in HAdV monoculture. Also, when the burst assay was used to measure the production of virus particles, in the presence of GCV the reductions in virus yield were higher for HCMV and co-infection than for HAdV in monoculture (145.2±35.5 vs. 116.4±27.3 vs. 23.0±10.0-folds, respectively, p less then 0.05). The improved anti-HAdV activity of GCV during co-infection suggests that this might be due to the more efficient phosphorylation of GCV by the HCMV protein kinase UL97. Although further evaluations will be required to confirm these results, here we suggest that patients treated with GCV as pre-emptive therapy for HCMV infection may be considered as low-risk patients to develop HAdV infections. The COVID-19 pandemic, caused by the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), is a current global threat for which there is an urgent need to search for an effective therapy. The transmembrane spike (S) glycoprotein of SARS-CoV-2 directly binds to the host angiotensin-converting enzyme 2 (ACE2) and mediates viral entrance, which is therefore considered as a promising drug target. Considering that new drug development is a time-consuming process, drug repositioning may facilitate rapid drug discovery dealing with sudden infectious diseases. Here, we compared the differences between the virtual structural proteins of SARS-CoV-2 and SARS-CoV, and selected a pocket mainly localizing in the fusion cores of S2 domain for drug screening. A virtual drug design algorithm screened the Food and Drug Administration-approved drug library of 1234 compounds, and 13 top scored compounds were obtained through manual screening. Through in vitro molecular interaction experiments, eltrombopag was further verified to possess a high binding affinity to S protein plus human ACE2 and could potentially affect the stability of the ACE2-S protein complex. Hence, it is worth further exploring eltrombopag as a potential drug for the treatment of SARS-CoV-2 infection. Background "Demonstrate insight and understanding into emotion" is a competency amenable to simulation-based assessment. The Jefferson Scale of Patient Perceptions of Physician Empathy (JSPPPE) has validity evidence for patients to assess provider empathy. A version adapted for a 3rd-party observers does not exist. Objective Modify the JSPPPE and use recorded standardized encounters to obtain validity evidence. Methods This cross-sectional study used video and data collected from two pediatric residencies. In 2018, 4 raters reviewed 24 videos of 12 interns communicating with standardized patients (SP) in two encounters and completed a modified JSPPE for observers (JSEO). Reliability between raters was established using Intraclass Correlations (ICC). JSEO mean scores were correlated to Essential Elements of Communication (EEC), JSPPPE, and faculty composite interpersonal communication (IC) scores using Spearman Rank. Results The mean ICC for all 4 raters was 0.573 (0.376-0.755). When ICC was calculated for pairs of raters, Rater 1 was an outlier.
