This study showed that the snake venom metalloprotease (SVMP) BaP1 activates FLSs to produce PGE2 by a mechanism dependent on COX-2, mPGES-1 and iPLA2s. BaP1 also induces IL-1β release, which up-regulates the production of PGE2 at a late stage of the stimulation. Expression of COX-2 and mPGES-1 are induced by BaP1 via activation of NF-κB pathway. While NF-κB p50 and p65 subunits are involved in up-regulation of COX-2 expression, only p65 is involved in BaP1-induced mPGES-1 expression. In addition, BaP1 up-regulates EP4 receptor expression. Engagement of this receptor by PGE2 triggers a positive feedback loop for its production by up-regulating expression of key components of the PGE2 biosynthetic cascade (COX-2, mPGES-1 and the EP4 receptor), thus contributing to amplification of BaP1-induced effects in FLSs. These data highlight the importance of FLS as a target for metalloproteases in joint inflammation and provide new insights into the roles of MMPs in inflammatory joint diseases. Moreover, our results may give insights into the importance of the catalytic domain, of MMPs for the inflammatory activity of these enzymes.In this work, two copper-based biometamaterials were engineered using leaves of water cabbage (Pistia stratiotes) and purple bauhinia (Phanera purpurea) as templates. The copper sputtering was implemented to produce a thin copper film on the surface of leaves. The scanning electron microscopy (SEM) images exhibited the root hair-like nanostructure of water cabbage leaf and single comb-like nanostructure of purple bauhinia leaf. In spite of copper coating, the leaf surfaces of water cabbage and purple bauhinia were black and exhibited excellent light absorption at visible and near infrarrred wavelengths. It was estimated that these two types of leaves could absorb roughly 90% of light. Finite-difference time-domain (FDTD) calculations predicted the low reflectance stemming from the leaf nanostructures and copper coating layer. Because of the low cost of copper as a coating metal and simple procedure, this can be a promising method for quick fabrication of a thin copper film on the leaf nanostructure for application in blackbody or as the light absorbers.Functionalizing the surfaces of transition metal dichalcogenide (TMD) nanosheets with noble metals is important for electrically contacting them to devices, as well as improving their catalytic and sensing capabilities. Solution-phase deposition provides a scalable approach to the creation of metal-TMD hybrid systems, but controlling such processes remains challenging. Here we elucidate the different pathways by which gold and silver deposit at room temperature onto colloidal 1T-WS2, 2H-WS2, 2H-MoSe2, 2H-WSe2, 1T'-MoTe2 and Td-WTe2 few-layer nanostructures to produce several distinct classes of 0D-2D and 2D-2D metal-TMD hybrids. Uniform gold nanoparticles form on all of the TMDs. By contrast, silver deposits as nanoparticles with a bimodal size distribution on the disulfides and diselenides, and as atomically thin layers on the ditellurides. The various sizes and morphologies of these surface-bound metal species arise from the relative strengths of the interfacial metal-chalcogen bonds during the reduction of Au3+ or Ag+ by the TMDs.A new approach to controlling the flow of a plasmatic electron packet at the interface between metallic and dielectric layers is described. The proposed metamaterial structure operates in the optical frequency range and can be used as a digital processing filter. It exhibits two double negative resonances and one special passband region, while the existence of a metal-dielectric nano-tunnel enhances electromagnetic wave-metal interactions. The structural arrangement of this metamaterial coupled with the tunnel layer can effectively control the electric field and allows digital encoding of electron packets.Blood-brain barrier (BBB) dysfunction occurs in cerebrovascular diseases and neurodegenerative disorders such as stroke. Opening of the BBB during a stroke has a negative impact on acute outcomes. We have recently demonstrated that miR-34a regulates the BBB by targeting cytochrome c (CYC) in vitro. To investigate the role of miR-34a in a stroke, we purified primary cerebrovascular endothelial cells (pCECs) from mouse brains following 1 h transient middle cerebral artery occlusion (tMCAO) and measured real-time PCR to detect miR-34a levels. We demonstrate that the miR-34a levels are elevated in pCECs from tMCAO mice at the time point of BBB opening following 1 h tMCAO and reperfusion. Interestingly, knockout of miR-34a significantly reduces BBB permeability, alleviates disruption of tight junctions, and improves stroke outcomes compared to wild-type (WT) controls. CYC is decreased in the ischemic hemispheres and pCECs from WT but not in miR-34a-/- mice following stroke reperfusion. We further confirmed CYC is a target of miR-34a by a dural luciferase reporter gene assay in vitro. Our study provides the first description of miR-34a affecting stroke outcomes and may lead to discovery of new mechanisms and treatments for cerebrovascular and neurodegenerative diseases such as stroke.An ultrasensitive indirect competitive enzyme-linked immunosorbent assay (ic ELISA) using monoclonal antibodies (mAbs) was developed for the specific detection of diethylstilbestrol (DES) residues. To establish an ELISA based on mAbs, hapten diethylstilbestrol mono-carboxypropyl-ether (DES-MCPE) was chemically synthetized and then conjugated to bovine serum albumin (BSA) for immunization in mice.
