This study examined the influence of preanalytical variables-staining protocol design, reagent quality, section attributes, and instrumentation-on the performance of automated DIA software. Our hypotheses were that (1) staining intensity is impacted by subtle differences in protocol design, reagent quality, and section composition and that (2) identically programmed and loaded stainers will produce equivalent immunohistochemical (IHC) staining. We tested these propositions by using 1 hematoxylin and eosin stainer to process 13 formalin-fixed, paraffin-embedded (FFPE) mouse tissues and by using 3 identically programmed and loaded immunostainers to process 5 FFPE mouse tissues for 4 cell biomarkers. Digital images of stained sections acquired with a commercial whole slide scanner were analyzed by customizable algorithms incorporated into commercially available DIA software. Staining intensity as viewed qualitatively by an observer and/or quantitatively by DIA was affected by staining conditions and tissue attributes. Intrarun and inter-run IHC staining intensities were equivalent for each tissue when processed on a given stainer but varied measurably across stainers. Our data indicate that staining quality must be monitored for each method and stainer to ensure that preanalytical factors do not impact digital pathology data quality.Drug-resistant Salmonella enteric serovar Typhi (S. Terephthalic Typhi) poses a vital public health issue. To overcome drug resistance issues, the development of effective drugs with novel mechanism(s) of action is required. In this regard, drug repurposing is a viable alternative approach to find novel drugs to overcome drug resistance. Therefore, a FDA-approved-drug-library containing 1930 drugs was analyzed against the dihydrofolate reductase (DHFR) of S. Typhi using deep learning regression algorithms. Initially, a total of 500 compounds were screened, followed by rescreening by molecular docking. Further, from screened compounds by molecular docking, the top eight compounds were subjected to molecular dynamics (MD) simulation. Analysis of MD simulation resulted in four potential compounds, namely; Duvelisib, Amenamevir, Lifitegrast and Nilotinib against the DHFR enzyme. During the MD simulation, these four drugs achieved good stability during the 100 ns trajectory period at 300 K. Further to know the insights of the complex's stability, we calculated RMSF, RG, SASA and interaction energy for the last 60 ns trajectory period because all complexes showed the stability after 40 ns trajectory period. MM-PBSA analysis of the last 10 ns of MD trajectories showed the stability of the complexes. From our results, we conclude that these drugs can also be useful for treating typhoid fever and can inhibit S. Typhi by interfering with the function of the DHFR enzyme. Communicated by Ramaswamy H. Viral RNA-dependent RNA polymerase (RdRp) catalyzes the replication of RNA from RNA and the main protease (Mpro) has a role in the processing of polyproteins that are translated from the RNA of SARS-CoV-2, and thus these two enzymes are strong candidates for targeting by anti-viral drugs. Small molecules such as lopinavir and favipiravir significantly inhibit the activity of Mpro and RdRp in vitro. Studies have shown that structurally modified lopinavir, favipiravir, and other similar compounds can inhibit COVID-19 main protease (Mpro) and RNA-dependent RNA polymerase (RdRp). In this study, lopinavir and its structurally similar compounds were chosen to bind the main protease, and favipiravir was chosen to target RNA-dependent RNA polymerase. Sarma.Dual career collegiate athletes, who combine sporting endeavours with academic studies, are commonplace in high-performance sport. Sleep is an important aspect of physical and psychological recovery for athletes, plays a role in memory and learning and is associated with academic achievement. The aim of this study was to assess variations in sleep characteristics of collegiate swimmers to understand the incidence of poor sleep, and which stressors might contribute towards this. A total of 22 (male n = 13, female n = 9) high-performance student swimmers (mean ± SD; age 20 ± 2 years) participated in this study. Sleep/wake behaviour was assessed using the Pittsburgh Sleep Quality Index each month over a 12-month period. Additionally, academic and sporting commitments were quantified on a monthly basis and summarised as 4 stressors average weekly training hours, total number of competitive races, total number of academic assessments, and average weekly learning hours. On average across the 12-month period, 41.7% of athletes reported poor sleep. The latest bedtimes, wake times and longest sleep durations were found in the months where academic and training demands were lowest. A statistically significant positive association was identified between median sleep quality scores and mean number of academic assessments (ρ(12) = 0.71, p=0.005). Hierarchal linear modelling analysis determined that number of academic assessments best predicted sleep quality within this cohort. The present study highlights the need for those working with athletes who have competing academic demands to consider sleep quality, and its potential impact on performance and wellbeing.
