T-cell receptor (TCR) signaling encourages the particular assemblage of RanBP2/RanGAP1-SUMO1/Ubc9 nuclear pore subcomplex by means of PKC-θ-mediated phosphorylation involving RanGAP1.
The proportion of tulathromycin-treated ewes that developed vaginal bleeding or aborted during the 21 days after C jejuni inoculation (2/9) was significantly less than that for the sham-treated ewes (7/9). CONCLUSIONS AND CLINICAL RELEVANCE Results suggested that administration of tulathromycin to pregnant ewes following exposure to C jejuni was effective in decreasing the number of C jejuni-induced abortions. Because of concerns regarding the development of macrolide resistance among Campylobacter strains, prophylactic use of tulathromycin in sheep is not recommended.OBJECTIVE To compare a ventral and a left lateral endoscopic approach to coelioscopy in bearded dragons (Pogona vitticeps). ANIMALS 18 adult bearded dragons. PROCEDURES In a randomized crossover design involving 2 surgical approaches, anesthetized bearded dragons first underwent coelioscopy with a ventral approach (left lateral of midline next to the umbilicus; animal positioned in dorsal recumbency) or left lateral approach (intercostal; animal positioned in right lateral recumbency) and then with the alternate approach. A 2.7-mm × 18-cm, 30° oblique telescope with a 4.8-mm operating sheath and CO2 insufflation at 2 to 5 mm Hg were used. Ease of entry into the coelom and ease of visual examination of visceral structures were scored. RESULTS Both approaches were straightforward, with the left lateral approach requiring significantly more time than the ventral approach. Scores for ease of visual examination for the heart, lungs, liver, stomach, intestines, pancreas, gallbladder, left kidney, gonads, and fat body were good to excellent. Visual examination of the spleen and adrenal glands was difficult in most animals via either approach. The left kidney, testis, and vas deferens were easier to see with the left lateral approach, whereas the pancreas in females and gallbladder in both sexes were easier to see with the ventral approach. All bearded dragons recovered without complications from the procedures, except for one with nephritis, renal gout, and hepatic necrosis. CONCLUSIONS AND CLINICAL RELEVANCE Both coelioscopy approaches could be safely and effectively used in bearded dragons. Choice of approach should be based on the coelomic structures requiring evaluation.OBJECTIVE To quantify acute immunologic and metabolic responses of beef heifers following topical administration of transdermal flunixin meglumine (TDFM) at various times relative to bovine herpesvirus 1 (BHV1) and Mannheimia haemolytica challenges. ANIMALS 32 beef heifers (mean body weight, 170 kg). PROCEDURES Heifers were assigned to 1 of 4 groups. Heifers in the control group did not receive TDFM, whereas 1 dose of TDFM (3.3 mg/kg) was topically applied to heifers of groups A, V, and B at -144, -72, and 0 hours. https://www.selleckchem.com/products/trilaciclib.html All heifers were inoculated with 1 × 108 plaque-forming units of BHV1 in each nostril at -72 hours and with 1.18 × 106 CFUs of M haemolytica intratracheally at 0 hours. Vaginal temperature was recorded and blood samples were collected for quantification of select immunologic and metabolic biomarkers at predetermined times from -144 to 360 hours. RESULTS Mean vaginal temperature was similar between group A and the control group. Mean vaginal temperatures for groups V and B were generally lower than that for the control group following BHV1 and M haemolytica challenges, respectively. Mean neutrophil oxidative burst capacity and L-selectin expression at 0 hours were significantly decreased for group V relative to the other groups. Other biomarkers did not differ among the groups at any time. CONCLUSIONS AND CLINICAL RELEVANCE Results suggested that topical administration of TDFM to beef cattle effectively alleviated pyrexia without adverse effects on acute immunologic or metabolic responses when TDFM was administered at the same time as, but not before, respiratory pathogen challenge.OBJECTIVE To develop a 3-D kinematic model to measure truncal motion in dogs and assess changes in truncal motion in dogs when wearing each of 2 service vests. ANIMALS 5 adult mixed-breed dogs. PROCEDURES 27 reflective markers were placed on the pelvis, trunk, and scapula of each dog. Six infrared cameras were placed around a treadmill to track the location of the markers within a calibrated space. Dogs were recorded during walking and trotting on the treadmill. Local and global coordinate systems were established, and a segmental rigid-body model of the trunk was created. Dogs were then recorded while wearing a custom vest and an adjustable vest during walking and trotting on the treadmill. Range of motion of the trunk when dogs were and were not wearing vests was compared by repeated-measures ANOVA. RESULTS An anatomic coordinate system was established by use of markers located at T1, T13, and the xiphoid process. Range of motion of the trunk during a gait cycle did not differ significantly regardless of the day of the test for both walking and trotting gaits. Trunk motion of dogs when walking and trotting was significantly reduced when dogs were wearing a vest, compared with trunk motion when not wearing a vest. CONCLUSIONS AND CLINICAL RELEVANCE A 3-D kinematic model for measuring truncal rotation was developed. Results indicated measurable differences in the gait of dogs when wearing each of the 2 service vests, compared with the gait when not wearing a vest.OBJECTIVE To evaluate histologic changes and gene expression patterns in body and limb wounds in horses in response to bacterial inoculation. SAMPLE Wound biopsy specimens from 6 horses collected on days 7, 14, 21, and 27 after excisional wounds (20 wounds/horse) were created over the metacarpal and metatarsal region and lateral thoracic region (body) and then inoculated or not inoculated on day 4 with Staphylococcus aureus and Pseudomonas aeruginosa. PROCEDURES Specimens were histologically scored for the amount of inflammation, edema, angiogenesis, fibrosis organization, and epithelialization. Quantitative PCR assays were performed to quantify gene expression of 10 inflammatory, proteolytic, fibrotic, and hypoxia-related markers involved in wound healing. RESULTS Except for gene expression of interleukin-6 on day 27 and tumor necrosis factor-α on day 14, bacterial inoculation had no significant effect on histologic scores and gene expression. Gene expression of interleukin-1β and -6, serum amyloid A, and matrix metalloproteinase-9 was higher in limb wounds versus body wounds by day 27.
