In the gastric cancer cell line KATOIII, quantitative PCR analysis demonstrated significantly reduced amounts of OBP2B and ABO transcripts in mutant cells with biallelic deletions of the site created using the CRISPR/Cas9 system, relative to those in the wild-type cells, and Western blotting demonstrated a corresponding reduction of OBP2B protein in the mutant cells. Moreover, single-molecule fluorescence in situ hybridization assays indicated that the amounts of both transcripts were correlated in individual cells. These findings suggest that OBP2B could be co-regulated by the + 22.6-kb site of ABO.Wnt signaling plays a key role in neurodevelopment and neuronal maturation. However, phosphorylated eIF2α promotes the translation of some proteins with more than one open reading frame in its 5' untranslated region. One of these proteins targeted by Wnt-HRI-eIF2α mediated translation is the GluN2B subunit of the NMDA receptor. The identified increase in GluN2B expression correlated with increased NMDA receptor function. This study examined the effects of model polystyrene (PS)-based submicron particles on the detection of aqueous geno- and cytotoxicity by genetically modified bioluminescent (GMB) bacteria. The toxicities were tested in three treated wastewater (TWW) effluents before and after chlorination. The PS plastics included negatively charged sulfate-coated (S-PS) and pristine (P-PS) particles of different sizes (0.1, 0.5, and 1.0 µm) that were present at different concentrations. Chlorinated or not, the S-PS and P-PS particles per se were not toxic to the GMB bacteria. However, exposure of PS particles to TWW effluents can significantly reduce the measured geno- and cytotoxicity. The long-term cross-calibration results further indicate that due to the adjustment of HJ-1A CCD gain state in October 2009, an abrupt change occurred 405 days after launch. However, the conventional evaluation method by using permeability damage rate as a constraint restricts the mechanistic understanding of the strong stress sensitivity effect. In this study, morphology of stress sensitivity test curve, coupled with rate change of permeability and extent of irreversible damage in actual sample measurement through micro-CT in-situ scanning, is used to characterize differentially. The strong stress sensitivity effects of the studied intervals can be divided into three types (1) rapid change in permeability-weak irreversible damage, (2) moderate change in permeability-strong irreversible damage and (3) moderate change in permeability-moderate irreversible damage. The strong stress sensitivity is caused by the micro-pores and micro-fractures, which are widely developed in the studied reservoir.
