Viruses have developed direct cell-to-cell transfer strategies to enter target cells without being released to escape host immune responses and antiviral treatments. These strategies are more rapid and efficient than transmission through indirect mechanisms of viral infection between cells. Here, we demonstrate that an H5N1 influenza virus can spread via direct cell-to-cell transfer in Madin-Darby canine kidney (MDCK) cells. We compared cell-to-cell transmission of the H5N1 virus to that of a human influenza H1N1 virus. The H5N1 virus has been found to spread to recipient cells faster than the human influenza H1N1 virus. Additionally, we showed that plasma membrane exchange (trogocytosis) occurs between co-cultured infected donor cells and uninfected recipient cells early point, allowing the intercellular transfer of viral material to recipient cells. Notably, the H5N1 virus induced higher trogocytosis levels than the H1N1 virus, which could explain the faster cell-to-cell transmission rate of H5N1. Importantly, this phenomenon was also observed in A549 human lung epithelial cells, which are representative cells in the natural infection site. Altogether, our results provide evidence demonstrating that trogocytosis could be the additional mechanism utilized by the H5N1 virus for rapid and efficient cell-to-cell transmission.Microorganisms, usually bacteria and fungi, grow and spread in skin wounds, causing infections. The use of photodynamic therapy (PDT) to eradicate microorganisms has been regarded as a promising alternative to anti-infective therapies, such as those based on antibiotics, and more recently, is being considered for skin wound-healing, namely for infected wounds. Specimens were analyzed with the endpoints clustering and interactions between CTL, normoxic, and hypoxic tumor areas, both visually and using spatial statistical tools implemented in the R package Spatstat. (3) Results Visual assessment suggested clustering of CTL in all instances. The visual analysis also suggested an inhibitory effect between hypoxic tumor areas and CTL in a minority of the whole-slide scans (9 of 22, 41%). Conversely, the objective mathematical analysis in Spatstat demonstrated statistically significant inhibitory interactions between hypoxia and CTL accumulation in a substantially higher number of specimens (16 of 22, 73%). It showed a similar trend in all but one of the remaining samples. (4) Conclusion Our findings provide non-obvious but statistically rigorous evidence of inhibition of CTL infiltration into hypoxic tumor subregions of cancers of the head and neck. Importantly, these shielded sites may be the origin of tumor recurrences. We provide the methodology for the transfer of our statistical approach to similar questions. We discuss why versions of the Kcross and pcf.cross functions may be the methods of choice among the repertoire of statistical tests in Spatstat for this type of analysis.Ecological conservation red line (ECRL) is gaining increasing academic attention as delimiting the minimum space scope of ecological protection and the bottom line of ecological security. Taking Nanjing as a case study, we divided the territory into ecological and non-ecological redline areas (ERAs and NERAs, respectively). This paper highlights two key research issues based on the 2005, 2010, 2015 and 2018 annual remote sensing data (i) quantitative analysis of the Ecological Redline Policy (ERP) validity by conducting a horizontal comparison of the ERAs and NERAs; and (ii) exploration of the land-use transitions and spatial pattern changes affecting ecosystem service value (ESV). Results showed that delineating ECRL could effectively slow down the decline rate of ESV. The trend of eco-quality deterioration was greater than eco-quality improvement in Nanjing, presenting an ESV that declined slightly in the whole. According to our findings, we suggest that reasonably increasing eco-lands (woodland and water area) and decreasing construction land will enhance the regional ESV. Meanwhile, promoting the transition from production space to ecological space and depressing the encroachment of living space on other space types, will be instrumental in mitigating the ESV decline. The results of this study are expected to provide valuable implications for spatial planning and sustainable development in Nanjing.
