Pregnant women with sickle cell disease (SCD) are at high risk for sickle cell-related complications, obstetrical complications, and perinatal morbidity. Chronic inflammation and the proangiogenic environment associated with SCD have been associated with endothelial damage. It is unknown whether SCD complications could be associated with placental dysfunction or abnormal placental morphology. Moreover, circulating angiogenic factors in pregnant women with SCD are unexplored.

Clinical records, placental and blood samples were collected at term delivery for 21 pregnant patients with SCD and 19 HbAA pregnant controls with adapted to gestational age birth weight newborns. Histological and stereological analyses and scanning electron microscopy (SEM) of the placenta, and PlGF and sFlt1 measurements in blood were performed.

In the SCD group, the parenchyma-forming villi of placentas were thinner than in controls, and increased fibrinoid necrosis and an overabundance of syncytial knots were seen. SEM revealed elongated intermediate villous endings with a reduction in the number of terminal villi compared to controls, indicating a significant branching defect in SCD placentas. Finally, SCD patients had an imbalance in the angiogenic ratio of sFlt1/PlGF (p=0.008) with a drop of PlGF concentrations.

Although lower respiratory infections (LRI) are among the leading causes of mortality in the US, their association with underlying factors and geographic variation have not been adequately examined.

In this study, explanatory variables (n = 46) including climatic, topographic, socio-economic, and demographic factors were compiled at the county level across the continentalUS.Machine learning algorithms - logistic regression (LR), random forest (RF), gradient boosting decision trees (GBDT), k-nearest neighbors (KNN), and support vector machine (SVM) - were employed to predict the presence/absence of hotspots (P < 0.05) for elevated age-adjusted LRI mortality rates in a geographic information system framework.

Overall, there was a historical shift in hotspots away from the western US into the southeastern parts of the country and they were highly localized in a few counties. The two decision tree methods (RF and GBDT) outperformed the other algorithms (accuracies 0.92; F1-scores 0.85 and 0.84; area under the precision-recall curve 0.84 and 0.83, respectively). Moreover, the results of the RF and GBDT indicated that higher spring minimum temperature, increased winter precipitation, and higher annual median household income were among the most substantial factors in predicting the hotspots.

Evaluating Health Information Technologies (HITs) can be challenging, but studies are necessary so that the most beneficial interventions can be identified. Our objective was to systematically review the available recommendations for improving the methods used in HIT evaluations.

HIT evaluation frameworks were identified from database (MEDLINE, EMBASE, CINAHL) and grey literature searches. Outcome measures included framework recommendations and characteristics. Recommendations were coded and organised using thematic analysis methods. A scoring instrument was used to measure framework quality.

The search identified 23 frameworks and 272 recommendations. These were organised into five evaluation domains and 42 themes. The themes included recommendations for improving the evaluation of technical aspects of HITs (e.g. N-Ethylmaleimide nmr describing aspects of HIT functionality) and suggestions for improving the evaluation of complex factors that may influence the overall effects of HITs (e.g. careful reporting of whether the Hssfully plan, conduct and appraise HIT evaluations. The quality appraisal demonstrated that HIT evaluation research may be improved by using more systematic methods and the involvement of participants from a range of differing backgrounds.Fingerprint enhancement often includes either physical or chemical approaches, such as fingerprint powder or cyanoacrylate fuming, to improve the quality of a fingerprint for visualization and analysis. However, these methods become more complex when fingerprints are partial bloody, and these procedures may interfere with downstream DNA analysis. Columnar thin film (CTF) deposition is a type of nanotechnology that utilizes an evaporant material to enhance a fingerprint under low-pressure conditions. Short tandem repeat (STR) analysis is the traditional method employed in crime laboratories. When DNA is of poor quality and quantity, like that often obtained from fingerprints, little to no genetic information may be obtained. Single nucleotide polymorphisms (SNPs) may be used to glean additional information when STR analysis fails. In this pilot study, 100 partial bloody fingerprints were collected from two donors and deposited on five different crime scene substrates, in which half were enhanced with CTFs and were graded for quality by an IAI-certified latent fingerprint examiner. CTF-developed fingerprints, on average, had higher grades compared to non-developed partial bloody fingerprints. STR analysis using Fusion 6C was performed to assess inhibition from the evaporant materials, in which no inhibition was observed. Sequencing of SNPs using the Precision ID Identity Panel was also employed, in which genetic information that could not be obtained from STRs was acquired with SNPs. Various sample types (i.e. pristine, low quality, and contaminated) utilized in this project demonstrated the acceptable performance of the Precision ID Identity Panel.
