In this investigation we developed total syntheses of both inhibitors. ML-SI1 was only obtained as a racemic mixture of inseparable diastereomers and showed activator-dependent inhibitory activity. The more promising tool is ML-SI3, hence ML-SI1 was not further investigated. For ML-SI3 we confirmed by stereoselective synthesis that the trans-isomer is significantly more active than the cis-isomer. Separation of the enantiomers of trans-ML-SI3 further revealed that the (-)-isomer is a potent inhibitor of TRPML1 and TRPML2 (IC50 values 1.6 and 2.3 μM) and a weak inhibitor (IC50 12.5 μM) of TRPML3, whereas the (+)-enantiomer is an inhibitor on TRPML1 (IC50 5.9 μM), but an activator on TRPML 2 and 3. This renders the pure (-)-trans-ML-SI3 more suitable as a chemical tool for the investigation of TRPML1 and 2 than the racemate. The analysis of 12 analogues of ML-SI3 gave first insights into structure-activity relationships in this chemotype, and showed that a broad variety of modifications in both the N-arylpiperazine and the sulfonamide moiety is tolerated. An aromatic analogue of ML-SI3 showed an interesting alternative selectivity profile (strong inhibitor of TRPML1 and strong activator of TRPML2).Over the last ten years, we have published various manuscripts on the recovery of assisted voluntary walking in people with chronic spinal chord injuries (SCI), following laparoscopic implantation of stimulation electrodes on the pelvic somatic nerves - the LION procedure. Three different length nano-titanium have been studied using Drosophila, TEM, and different biological markers. Nano-titanium, regardless of its shape, was eroded and degraded just entering the gut lumen of the larvae. Results showed that the distinguished shape of nanowires helps to understand the interactions of NMs with the intestinal barrier. The peritrophic membrane, as the first defense line of the intestinal barrier, succeeded in the reservation of NMs, though the perpendicular particles of nanowires stabbing it, making pores, and permitting their translocation into intestinal cells. Results showed that the peak overpressure, maximum flame temperature, and average flame propagation speed of the stoichiometric CH4/air explosion reach 0.84 MPa, 2614 K and 3.5 m/s, respectively. The optimal concentration of cornstarch explosion is 750 g/m3, and its peak overpressure, maximum flame temperature and average flame propagation speed are 0.76 MPa, 2098 K and 1.77 m/s, respectively. For a three-components system, adding methane can significantly increase the explosive intensity and combustion performance of cornstarch. The explosive intensity parameters (peak overpressure, maximum flame temperature, average flame propagation speed) of a certain concentration of cornstarch first increase and then decrease with the increase of methane concentration. The maximum explosion intensity parameters of a three-components system with a certain concentration of lean-methane/air are higher than that of single-phase, but always lower than that of the stoichiometric methane/air. Moreover, the mutual coordination of dust and combustible gas in energy release and the mutual competition mechanism in oxygen consumption are described.
