To reduce the cost of lignocellulosic pretreatment, rice straw was ensiled with dilute formic acid (FA, 0, 0.2, 0.4, and 0.6%) for 3, 6, 9, 15 and 30 days, and evaluated its effects on fermentation dynamics, lignocellulosic degradation and enzymatic hydrolysis. The results showed that the application of FA, especially at 0.6% level, reduced total fermentation losses of the resulting silages, as evidenced by low dry matter loss, ammonia nitrogen and ethanol content. Meanwhile, the 0.6% FA application promoted hemicellulose removal (232.41 vs 187.52 g/kg DM) and xylose production (0.35 vs 2.80 g/kg DM). The glucose yield and cellulose convertibility of rice straw increased after 30 days of ensiling, and further enhanced by the 0.6% FA application. In conclusion, the 0.6% FA-assisted ensiling pretreatment improved both biomass preservation, hemicellulose removal and enzymatic hydrolysis of rice straw, which is beneficial to the subsequent biofuel production chain.Current work for animal manure processing is not up to the required standards and hence are not supposed to reflect the actual performance in antibiotic resistance control. As a result, this study carried out temperature-controlled aerobic composting, with sulfamethoxazole (SMX) as a typical antibiotic. The results of four different treatments demonstrated that temperature, water content, C/N ratio, EC, and pH showed no significant (p > 0.05) difference. Antibiotic resistance genes (ARGs) significantly decreased in the initial 10 days of the thermophilic phase, but the abundance of sul1 and sul2 increased greatly after 30 days. Moreover, ARGs were closely related with each other during the late stages of composting. A noteworthy effect of composting properties, especially temperature on bacterial community, which then had a positive effect on ARGs abundances. These findings provided evidence that the standard composting was still insufficient to control antibiotic resistance. The impacts of magnesium-ammonium-phosphate (MAP) precipitation on the performance and microbial dynamics in an anaerobic continuously stirred tank reactor (CSTR) coupled with sequencing batch reactor (SBR) for swine wastewater treatment were investigated. In CSTR-SBR systems, an overall higher removal efficiency for COD, NH4+ and PO43-as 98.6%, 98.7% and 97.9% was achieved with MAP precipitation, compared to CSTR-SBR without MAP pretreatment (i.e., 97.5, 74.3% and 19.9% for COD, NH4+ and PO43-, respectively). With MAP precipitation, the high C/N ratio of 6.6 after anaerobic CSTR was observed. The increase in the richness and diversity of microbial communities in CSTR with MAP was conducive to nitrogen and phosphorus removal, as well as biogas production. The core community was affiliated with bacterial phyla Firmicutes, Bacteroidetes, Proteobacteria, Cloacimonetes, and Spirochaetae. https://www.selleckchem.com/products/bi-2852.html The study provide a new insight into the potential application of MAP precipitation as pretreatment for dealing with nutrient recovery from high-strength swine wastewater.High mobility group box 1 (HMGB1) is a nuclear protein that is released on injury triggers inflammation. This study aims to elucidate the effects of salidroside on diabetes-induced liver inflammation. The levels of glucose, inflammatory cytokines and hepatic functional parameters in serum and liver of type 2 diabetic db/db mice were examined. Immunohistochemistry, immunofluorescence and western blot tests were performed to determine the mechanisms underlying the action. Palmitic acid (PA) or HMGB1-stimulated was adopted as an in vitro cell model. Salidroside treatment improved glucose tolerance, lipid profiles while decreased the production of inflammatory cytokines. It also reduced the levels of serum biochemical markers. In addition, salidroside inhibited HMGB1 signaling pathway in db/db mice. In the salidroside treatment significantly inhibited PA or HMGB1 induced inflammatory signaling pathway, too. HMGB1 inhibitors and HMGB1 knockdown both hindered PA-induced HMGB1 signaling pathway, showing the same effect as salidroside. Salidroside treatment significantly alleviates insulin resistance, hyperglycemia and hepatic inflammation in db/db mice, and also showed beneficial to PA-stimulated. Salidroside proves to control hyperglycemia and hepatic inflammation via inhibiting HMGB1/RAGE/NF-κB and HMGB1/TLR4/NLRP3 signaling pathways.
The present study aimed to investigate the possible effects of metformin on the olanzapine-induced insulin resistance in rats.

Rats were randomly divided into three groups the control (Control) group, the olanzapine (Ola) group and the olanzapine + metformin (Ola + Met) group. Rats in the Ola group received olanzapine (8 mg/kg/day) intraperitoneally while rats in the Ola + Met group received olanzapine (8 mg/kg/day) intraperitoneally and metformin (300 mg/kg/day) orally for 8 weeks. Rats in the Control group received vehicle accordingly. Body weight and fasting blood glucose were recorded routinely. Inflammatory cytokines TNF-α, IL-6 and IL-1β and IL-10 were measured by ELISA. The gene expression of macrophages markers was examined by qPCR. The epididymal white adipose tissue, liver and skeletal muscle were also isolated for immunohistochemical analysis.

Olanzapine significantly induced body weight gain and insulin resistance compared to the control, which was markedly alleviated by metformin. Pro-inflaolanzapine-induced insulin resistance possibly by suppressing the inflammatory responses mediated by macrophage infiltration and polarization in epididymal white adipose tissue.
Compound sophorae decoction (CSD), a Chinese Herbal decoction, is frequently clinically prescribed for patients suffered from ulcerative colitis (UC) characterized by bloody diarrhea. Yet, the underlying mechanism about how this formulae works is remain elusive.

In the present study, the experimental colitis in C57BL/6 J mice was induced by oral administration of standard diets containing 3% dextran sodium sulfate (DSS), and CSD was given orally for treatment at the same time. The clinical symptoms including stool and body weight were recorded each day, and colon length and its histopathological changes were observed. Apoptosis of colonic epithelium was studied by detecting protein expression of cleaved caspase-3, and cell proliferation by Ki-67 immunohistochemistry. Tight junction complex like ZO-1 and occludin were also determined by transmission electron microscope and immunofluorescence. The concentration of FITC-dextran 4000 was measured to evaluate intestinal barrier permeability and possible signaling pathway was investigated.
