Delayed tooth eruption is often seen in patients with palatally displaced canines (PDC), but there is controversy over whether tooth formation is also delayed. This study attempts to elucidate the answer by addressing methodological shortcomings in previous studies.

Archived records were searched, and 100 patients (aged 8-16years) with unilateral PDC were identified. Records of 50 women and 50 men were matched for age and sex with unaffected controls. The exclusion criteria included hypodontia. Dental age (DA) was calculated using the population-specific Maltese database and subtracted from the chronological age (CA). Nonparametric tests were used to compare CA-DA in patients with PDC and controls to determine the DA of end-of-series teeth.

No statistically significant difference with CA-DA between patients with PDC and controls was found. In addition, there were no particular teeth significantly delayed in development.

The objective of this research was to analyze the effect of orthodontic treatment with maxillary permanent first molar extraction on the occlusal stress distribution and displacement in the human skull.

A 3-dimensional finite element model was constructed on the basis of a computed tomography scan, and it wasused as the pretreatment model. The software used for geometric modeling was Solid Works (DassaultSystèmes, Paris, France). For the extraction model, the maxillary permanent first molar was removed, followed by a repositioning of the anterior and posterior segments to create a space closure model. Stress distribution was evaluated under the simulation of 1000N for occlusal forces and 400N for masseter muscle force.

The highest von Mises stress was observed at the zygomatic process of the temporal bone across all 3 models (25MPa), whereas stress at the pterygomaxillary suture area was almost 50% less. However, the stress in the pterygomaxillary suture area was lowest in the extraction model (18%) and space closure (30%). Stress in the zygomatic process of the frontal bone and frontal process of the maxilla increased from pretreatment to extraction model followed by space closure model.

Emerging evidence suggests the significance of microRNA-155 (miR-155) in fibrogenesis and oxidative stress accumulation, but its function in oral submucous fibrosis (OSF) has not been investigated. In this study, we assessed the expression of miR-155 and its effects on the maintenance of myofibroblast activation.

qRT-PCR was conducted to assess the expression of miR-155 in OSF tissues and fibrotic buccal mucosal fibroblasts (fBMFs) derived from OSF samples. Collagen gel contraction, migration, and invasion capabilities were examined in fBMFs. DCFDA ROS assay was used to examine ROS generation. A luciferase reporter assay was carried out to verify the relationship between miR-155 and its potential target RPTOR.

We showed that the expression of miR-155 was aberrantly upregulated in OSF specimens and fBMFs. selleckchem Inhibition of miR-155 ameliorated various myofibroblast activities, including collagen gel contraction, migration, and invasion abilities as well as ROS production. Results from the luciferase reporter assay demonstrated that miR-155 directly interacted with its target RPTOR.

Taken together, these findings indicate that miR-155 is implicated in the pathogenesis of oxidative stress-associated OSF, possibly through the regulation of RPTOR.
