We sought to validate a recently introduced scoring algorithm for dystonia, predicting the diagnostic utility of whole-exome sequencing (WES) based on individual phenotypic aspects (age-at-onset, body distribution, presenting comorbidity).
We prospectively enrolled a set of 209 dystonia-affected families and obtained summary scores (0-5 points) according to the algorithm. Singleton (N = 146), duo (N = 11), and trio (N = 52) WES data were generated to identify genetic diagnoses.
Diagnostic yield was highest (51%) among individuals with a summary score of 5, corresponding to a manifestation of early-onset segmental or generalized dystonia with coexisting non-movement disorder-related neurological symptoms. Sensitivity and specificity at the previously suggested threshold for implementation of WES (3 points) was 96% and 52%, with area under the curve of 0.81.

The objective of this study was to report the identification of antibodies against the glutamate kainate receptor subunit 2 (GluK2-abs) in patients with autoimmune encephalitis, and describe the clinical-immunological features and antibody effects.
Two sera from 8 patients with similar rat brain immunostaining were used to precipitate the antigen from neuronal cultures. A cell-based assay (CBA) with GluK2-expressing HEK293 cells was used to assess 596 patients with different neurological disorders, and 23 healthy controls. GluK2-ab effects were determined by confocal microscopy in cultured neurons and electrophysiology in GluK2-expressing HEK293 cells.
Patients' antibodies precipitated GluK2. GluK2 antibody-specificity was confirmed by CBA, immunoprecipitation, GluK2-immunoabsorption, and GluK2 knockout brain immunohistochemistry. In 2 of 8 samples, antibodies reacted with additional GluK2 epitopes present in GluK1 or GluK3; in both, the reactivity was abrogated after GluK2 immuno-absorption. While there is evidence for an association of child abuse with suicidality in the course of life, the underlying mechanisms remain unclear. The Interpersonal Psychological Theory of Suicide (IPTS) provides a theoretical framework to investigate this relationship. The present study examines how different subtypes of child abuse are related to suicidal ideation and to attempts in the context of the IPTS.
146 psychiatric inpatients (M=37.9years, 62% female) with an acute suicidal crisis (n=71) or a recent suicide attempt (n=74) were examined at baseline (T0) and six (T1) months later. We measured emotional, physical, and sexual abuse, the constructs of the IPTS (thwarted belongingness, perceived burdensomeness, and capability for suicide) and suicidal ideation as well as suicide attempts. Using the statistics program R, a network analysis of all named constructs was conducted. Centrality measures were computed.
Emotional abuse was the most central kind of abuse in the network and had a direct relationship s. Sexual abuse was the only kind of abuse with a direct relation to suicidal ideation. Capability for suicide had just a marginal position in the network analysis. Early interventions addressing the effects of child abuse are recommended. Replications in larger samples and with more relevant variables are needed.Existing studies have identified crucial roles for the hippocampus and a distributed set of cortical regions (e.g., the inferior parietal cortex) in learning novel words. Nevertheless, researchers have not clearly determined how the hippocampus and cortical regions dynamically interact during novel word learning, especially during form-meaning associative learning. As a method to address this question, we used an online learning paradigm and representational similarity analysis to explore the contributions of the hippocampus and neocortex to form-meaning associative learning. Twenty-nine native Chinese college students were recruited to learn 30 form-meaning pairs, which were repeated 7 times during fMRI scan. Form-meaning associative learning elicited activations in a wide neural network including regions required for word processing (i.e., the bilateral inferior frontal gyrus and the occipitotemporal cortex), regions required for encoding (i.e., the bilateral parahippocampus and hippocampus), and regions required for cognitive control (i.e., the anterior cingulate cortex and dorsolateral prefrontal cortex). More importantly, our study revealed the differential roles of the left hippocampus and bilateral inferior parietal lobule (IPL) in form-meaning associative learning. Specifically, higher pattern similarity in the bilateral IPL in the early learning phase (repetitions 1 to 3) was related to better learning performance, while higher pattern similarity in the left hippocampus in the late learning phase (repetitions 5 to 7) was associated with better learning performance. These findings indicate that the hippocampus and cortical regions (e.g., the IPL) contribute to form-meaning learning in different stages.
