Seawater-derived brines within both sea ice (sea-ice brine) and ancient layers of permafrost (cryopeg brine) support diverse microbes adapted to subzero temperatures and high salinities, yet little is known about viruses in these extreme environments, which, if analogous to other systems, could play important evolutionary and ecosystem roles. Here, we characterized viral communities and their functions in samples of cryopeg brine, sea-ice brine, and melted sea ice. Viral abundance was high in cryopeg brine (1.2 × 108 ml-1) and much lower in sea-ice brine (1.3 × 105 to 2.1 × 105 ml-1), which roughly paralleled the differences in cell concentrations in these samples. Five low-input, quantitative viral metagenomes were sequenced to yield 476 viral populations (i.e., specieing foundational data sets for these climate-threatened habitats, we found evidence that the viruses had habitat specificity, infected dominant microbial hosts, encoded host-derived metabolic genes, and mediated horizontal gene transfer among hosts. These results advance our understanding of the virosphere and how viruses influence extreme ecosystems. More broadly, the evidence that virally mediated gene transfers may be limited by host range in these extreme habitats contributes to a mechanistic understanding of genetic exchange among microbes under stressful conditions in other systems.By virtue of complex ecologies, the behavior of mutualisms is challenging to study and nearly impossible to predict. However, laboratory engineered mutualistic systems facilitate a better understanding of their bare essentials. On the basis of an abstract theoretical model and a modifiable experimental yeast system, we explore the environmental limits of self-organized cooperation based on the production and use of specific metabolites. We develop and test the assumptions and stability of the theoretical model by leveraging the simplicity of an artificial yeast system as a simple model of mutualism. We examine how one-off, recurring, and permanent changes to an ecological niche affect a cooperative interaction and change the population composition of an engineered mutualistic system. Moreover, we explore how the cellular burden of cooperating influences the stability of mutualism and how environmental changes shape this stability. Our results highlight the fragility of mutualisms and suggest interventions, including those that rely on the use of synthetic biology.IMPORTANCE The power of synthetic biology is immense. Will it, however, be able to withstand the environmental pressures once released in the wild. As new technologies aim to do precisely the same, we use a much simpler model to test mathematically the effect of a changing environment on a synthetic biological system. We assume that the system is successful if it maintains proportions close to what we observe in the laboratory. Extreme deviations from the expected equilibrium are possible as the environment changes. While cirrhosis is the main risk factor for HCC, the factors influencing progression from cirrhosis to HCC remain largely unknown. Gut microbiota plays a key role in liver diseases; however, its association with HCC remains elusive. This study aimed to elucidate microbial differences between patients with HCC-associated cirrhosis (HCC-cirrhosis) and cirrhotic patients without HCC and healthy volunteers and to explore the associations between diet, lifestyle, and the microbiome of these patients. Fecal samples and food frequency questionnaires were collected from 95 individuals (30 HCC-cirrhosis patients, 38 cirrhotic patients without HCC, and 27 age- and body mass index [BMI]-matched healthy volunteers). 16S rRNA gene sequencing was performed. Bacterial richness in cirrhosis and HCC-cirrhosis patients was significantly lower than in healthy controls. The HCC-cirrhosis group was successfully classified with an areocellular carcinoma, independently of cirrhosis severity and dietary habits.Diversification can generate genomic and phenotypic strain-level diversity within microbial species. This microdiversity is widely recognized in populations, but the community-level consequences of microbial strain-level diversity are poorly characterized. Using the cheese rind model system, we tested whether strain diversity across microbiomes from distinct geographic regions impacts assembly dynamics and functional outputs. We first isolated the same three bacterial species (Staphylococcus equorum, Brevibacterium auranticum, and Brachybacterium alimentarium) from nine cheeses produced in different regions of the United States and Europe to construct nine synthetic microbial communities consisting of distinct strains of the same three bacterial species. Comparative genomics identified distinct phylogenetic clusters and significant variation in genome content across the nine synthetic communities. When we assembled each synthetic community with initially identical compositions, community structure diverged ovrsity has the potential to drive variability in the aesthetics and quality of surface-ripened cheeses.Introduction Increases in tobacco price are known to reduce smoking prevalence, but these correlations may be blunted by the availability of budget cigarettes, promoted by the tobacco industry to maintain profits. Objective To investigate the effect of budget cigarettes on cigarette consumption using data from Europe 2004-2014. Methods Data on the annual population-weighted cigarette consumption per adult come from the International Cigarette Consumption Database. Data on the annual tobacco price come from Euromonitor International for 23 European countries. Median prices and price differentials (operationalised as percentages obtained by dividing the difference between median and minimum prices by the median price) were examined. A linear random-effects model was used to assess associations between median prices and price differentials with cigarette consumption within 1 year and with a 1-year time lag.
