We aimed to identify the microbiota alterations in the airway and gut related to mite-specific IgE responses in young children with airway allergies. This study enrolled 60 children, including 38 mite-sensitized children (20 rhinitis and 18 asthma), and 22 non-mite-sensitized healthy controls. Microbiome composition analysis of the throat swab and stool samples was performed using bacterial 16S rRNA sequencing. An integrative analysis of the airway and stool microbial profiling associated with IgE reactions in childhood allergic rhinitis and asthma was examined. The Chao1 and Shannon indices in the airway were significantly lower than those in the stool. Additionally, an inverse association of the airway microbial diversity with house dust mite (HDM) sensitization and allergic airway diseases was noted. Fecal IgE levels were positively correlated with the serum Dermatophagoides pteronyssinus- and Dermatophagoides farinae-specific IgE levels. Airway Leptotrichia spp. related to asthma were strongly correlated with fecal Dorea and Ruminococcus spp., which were inversely associated with fecal IgE levels and risk of allergic rhinitis. Dibutyryl-cAMP Moreover, four airway genera, Campylobacter, Selenomonas, Tannerella, and Atopobium, were negatively correlated with both serum mite-specific and fecal IgE levels. Among them, the airway Selenomonas and Atopobium spp. were positively correlated with stool Blautia and Dorea spp. related to asthma and allergic rhinitis, respectively. In conclusion, airway microbial dysbiosis in response to HDM and its cross-talk with the gut microbial community is related to allergic airway diseases in early childhood.Considering the elasticity of the real networks, the components in the network have a redundant capacity against the load, such as power grids, traffic networks and so on. Moreover, the interaction strength between nodes is often different. This paper proposes a novel nonlinear model of cascade failure in weighted complex networks considering overloaded edges to describe the redundant capacity for edges and capture the interaction strength of nodes. We fill this gap by studying a nonlinear weighted model of cascade failure with overloaded edges over synthetic and real weighted networks. The cascading failure model is constructed for the first time according to the overload coefficient, capacity parameter, weight coefficient, and distribution coefficient. Then through theoretical analysis, the conditions for stopping failure cascades are obtained, and the analysis shows the superiority of the constructed model. Finally, the cascading invulnerability is simulated in several typical network models and the US power grid. The results show that the model is a feasible and reasonable change of weight parameters, capacity coefficient, distribution coefficient, and overload coefficient can significantly improve the destructiveness of complex networks against cascade failure. Our methodology provides an efficacious reference for the control and prevention of cascading failures in many real networks.Fascioliasis is a neglected zoonotic disease that infects humans and ruminant species worldwide. In the absence of vaccines, control of fascioliasis is primarily via anthelminthic treatment with triclabendazole (TCBZ). Parasitic flatworms, including Fasciola hepatica, are active secretors of extracellular vesicles (EVs), but research has not been undertaken investigating EV anthelmintic sequestration. Adult F. hepatica were cultured in lethal and sub-lethal doses of TCBZ and its active metabolites, in order to collect EVs and evaluate their morphological characteristics, production and anthelmintic metabolite content. Transmission electron microscopy demonstrated that F. hepatica exposed to TCBZ and its metabolites produced EVs of similar morphology, compared to non-TCBZ exposed controls, even though TCBZ dose and/or TCBZ metabolite led to measurable structural changes in the treated F. hepatica tegument. qNano particle analysis revealed that F. hepatica exposed to TCBZ and its metabolites produced at least five times greater EV concentrations than non-TCBZ controls. A combined mass spectrometry and qNano particle analysis confirmed the presence of TCBZ and the TCBZ-sulphoxide metabolite in anthelmintic exposed EVs, but limited TCBZ sulphone was detectable. This data suggests that EVs released from adult F. hepatica have a biological role in the sequestration of TCBZ and additional toxic xenobiotic metabolites.Tumours with an immunosuppressive microenvironment respond poorly to therapy. Activation of the stimulator of interferon genes (STING) pathway can enhance intratumoural immune activation, but STING agonists are associated with high toxicity and degrade prematurely, which limits their effectiveness. Here, we show that the extended intratumoural release of the STING agonist cyclic di-AMP transforms the tumour microenvironment from immunosuppressive to immunostimulatory, increasing the efficacy of antitumour therapies. The STING agonist was electrostatically complexed with nanotubes comprising a peptide-drug conjugate (a peptide that binds to the protein neuropilin-1, which is highly expressed in tumours, and the chemotherapeutic agent camptothecin) that self-assemble in situ into a supramolecular hydrogel. In multiple mouse models of murine tumours, a single low dose of the STING agonist led to tumour regression and increased animal survival, and to long-term immunological memory and systemic immune surveillance, which protected the mice against tumour recurrence and the formation of metastases. Locally delivered STING agonists could help to reduce tumour immunosuppression and enhance the efficacy of a wide range of cancer therapies.The "geochemical effect of lanthanides" is a new concept proposed by the authors during the past decade. This concept reflects lanthanide shrinkage in elemental geochemistry, and it is statistically quantifiable. However, the geological significance of the various parameters of the equations obtained following quantization is not entirely clear. Cooperation and discussions from scholars in related research fields of rare earth elements in geology are required. In the present paper, from the perspective of coal geochemistry, the metamorphic coal seam of C2 series in Fengfeng Mine of Handan Coalfield in Hebei Province, China was used as an example. The geochemical significance of parameters in geochemical effect regression equation for lanthanides was evaluated, and two new formulas (regression equations) that characterized the geochemical behaviors of lanthanides were proposed. On this basis, concepts related to the geochemical effect of lanthanides, such as "individual parameters", "parameters in common", "two-sided parameters", and the "deviation value" of lanthanides, were proposed.
