Bis-cyclometallated Ir(iii) complexes containing 2-(1H-pyrazol-3-yl)pyridine ligands have been synthesised. Their absorption is almost unchanged with changes in pH however the emission intensities vary by a factor of up to three and the complexes have emission pKas in the range 8.0 to 10.0. Substituents on the pyrazole have only a minor effect on the emission pKa. Surprisingly the complexes with phenylpyrazole cyclometallated ligands 3aL1-3 showed an intensity decrease with increasing pH (switch off) whilst the corresponding phenylpyridine ones 3cL1-3 showed an increase in emission intensity with increasing pH. Putting electron-withdrawing CF3 substituents on the cyclometallating phenyls reduced the pKa of the complexes to 6.8-7.8, thereby extending the useful pKa range; however, in general it tended to reduce the magnitude of the change in emission intensity. Surprisingly the CF3-substituted complexes also showed a complete reversal in the direction of the intensity change when compared to their respective unsubstituted congeners.Selenium (Se) is closely associated with kidney disease, and renal injury often occurs together with hyposelenemia. This study was designed to reveal the mechanism underlying renal injury induced by Se deficiency in pigs. Twenty-four castrated male Yorkshire pigs were divided into two groups fed either a Se-deficient diet (0.007 mg Se per kg) or a Se-adequate diet (0.3 mg Se per kg). Serum and kidney samples were collected at the 16th week of the trial, processed, and analyzed for serum biochemistry, Se concentration, kidney index markers, histology, selenoprotein mRNA expression, redox status, and inflammatory cytokines. Dietary Se deficiency induced kidney injury, decreased (P less then 0.05) Se concentrations, and increased (P less then 0.05) kidney index and serum blood urea nitrogen, creatinine, and carbon dioxide values. Histological analysis indicated that Se deficiency induced inflammatory lesions and renal tubular atrophy in the renal medulla. Se deficiency downregulated (P less then 0.05) nine selenoprotein genes (GPX1, SELENOW, SELENOH, SELENOP, GPX3, TXNRD2, SELENOI, SELENON, and SELENOM) and upregulated (P less then 0.05) SEPHS2 in the kidneys. Se deficiency decreased (P less then 0.05) the activity of glutathione peroxidase, thioredoxin reductase, and catalase, as well as the hydroxyl radical inhibition capacity, and increased (P less then 0.05) the content of malondialdehyde and nitric oxide. Se deficiency increased (P less then 0.05) the expression of the transcription factors NF-κB and HIF-1α, and regulated inflammatory cytokines. Se deficiency increased (P less then 0.05) the expression of IL-6, IL-8, IL-12, IL-17, and cyclooxygenase-2, and decreased (P less then 0.05) the expression of IL-10, IL-13, and TGF-β. These results indicated that Se deficiency induces kidney injury through the regulation of selenoproteins, oxidative stress, and inflammation.The use of engineered nanoscale magnetic materials in healthcare and biomedical technologies is rapidly growing. Two examples which have recently attracted significant attention are magnetic particle imaging (MPI) for biological monitoring, and magnetic field hyperthermia (MFH) for cancer therapy. Here for the first time, the capability of a Lissajous scanning MPI device to act as a standalone platform to support the application of MFH cancer treatment is presented. The platform is shown to offer functionalities for nanoparticle localization, focused hyperthermia therapy application, and non-invasive tissue thermometry in one device. Combined, these capabilities have the potential to significantly enhance the accuracy, effectiveness and safety of MFH therapy. Measurements of nanoparticle hyperthermia during protracted exposure to the MPI scanner's 3D imaging field sequence revealed spatially focused heating, with a maximum that is significantly enhanced compared with a simple 1-dimensional sinusoidal excitation. The observed spatial heating behavior is qualitatively described based on a phenomenological model considering torques exerted in the Brownian regime. In vitro cell studies using a human acute monocytic leukemia cell line (THP-1) demonstrated strong suppression of both structural integrity and metabolic activity within 24 h following a 40 min MFH treatment actuated within the Lissajous MPI scanner. Furthermore, reconstructed MPI images of the nanoparticles distributed among the cells, and the temperature-sensitivity of the MPI imaging signal obtained during treatment are demonstrated. In summary, combined Lissajous MPI and MFH technologies are presented; demonstrating for the first time their potential for cancer treatment with maximum effectiveness, and minimal collateral damage to surrounding tissues.Self-assembly of cyclodextrin (CD) with guest polymers has attracted much attention owing to its biocompatibility and accessibility. In this study, we investigate the composition effect of poly(ethylene oxide)m-b-poly(propylene oxide)n-b-poly(ethylene oxide)m (EOmPOnEOm) triblock copolymers on lamellar or plate structures formed by complexation with β-CD. EO5PO29EO5, EO14PO29EO14, and EO75PO29EO75 show periodic lamellar morphology consisting of single-crystalline pseudo-polyrotaxane (PPR) nanosheets with a thickness equal to the central PO length. This is because β-CDs selectively cover the PO component and cause the microphase separation between β-CD and EO layers. The thickness of the EO layers increases linearly with increasing number of EO units, which suggests that the EO chains are constrained into virtual cylinders with the diameter of the β-CD. Tacrolimus This means that we can precisely control the thickness of both the crystal (β-CD and PO) and the amorphous (EO) layers in the lamellar structure. In contrast, EO2PO29EO2 forms a thin plate structure, where not only PO but also EO chains are covered with β-CD. Furthermore, the length of the central PO component is necessary to form the lamellar structure with the phase separation between the β-CD and EO layers. These findings provide a more fundamental understanding to enhance the variety and applicability of CD-based self-assembled materials.
