The preponderance of a specific d- or l-chirality in fats, sugars, amino acids, nucleic acids, and so on is ubiquitous in nature, yet the biological origin of such chiral dominance (i.e., with one enantiomer overwhelmingly present) remains an open question. One plausible proposal for the predominance of l-chirality in amino acids could be through evolutionary templating of chiral RNA-folding via chaperone activity. To help evaluate this possibility, single molecule fluorescence experiments have been performed that measure the chiral dependence of chaperone folding dynamics for the simple tetraloop-tetraloop receptor (TL-TLR) tertiary binding motif in the presence of a series of chiral amino acids. Specifically, d- vs l-arginine is found to accelerate the unfolding of this RNA motif in a chirally selective fashion, with temperature-dependent studies of the kinetics performed to extract free energy, enthalpy, and entropy landscapes for the underlying thermodynamics. Furthermore, all-atom molecular dynamics (MD) simulations are pursued to provide additional physical insight into this chiral sensitivity, which reveal enantiomer-specific sampling of nucleic acid surfaces by d- vs l-arginine and support a putative mechanism for chirally specific denaturation of RNA tertiary structure by arginine but not other amino acids.Fentanyl and its related synthetic analogs have recently become more readily available as a growing threat to public safety, such as pain relief and anesthetics. Sources of fentanyl are more likely to be illicitly manufactured than pharmaceutically manufactured and are often laced with other opioids, which ultimately increases the potency of fentanyl and results in an increased number of overdose deaths. The methods used to detect these compounds safely and quickly are of high interest due to their extreme potency. This study investigates the use of paper spray mass spectrometry (PS-MS), which is a simple atmospheric ionization process that can be used as a rapid study (60 s) with limited sample preparation and sample handling. PS-MS can be utilized with a synthetic microporous polyolefin silica matrix substrate, known as Teslin, which is manufactured by PPG Industries. The main characteristic of paper spray ionization with the Teslin substrate is the hydrophobicity, which is useful for a fast and direct analysis requiring only 1 μg of the sample. The application of this novel synthetic substrate to PS-MS has been illustrated with a fentanyl analog screening kit (FAS Kit), which was designed by the Centers for Disease Control (CDC) for the screening of 212 evolving synthetic opioids, including more than 190 fentanyl analogs. The comparable fragmentation with precursor molecule mass data from this study can be useful in improving the accurate detection and structural characterization of complex samples with a minimum interference of the isobaric components.We investigate the spin-state energetics and the role of ionic charges in the electronic configuration of binuclear complexes of the form [(NH3)5Co(py)-X-(py)Ru(NH3)5]q+. In these compounds with q = 4-6, py = pyridine, and X = C≡C and C60, the Co-Ru distance varies from ∼1.4 to ∼2.1 nm. We carry out a systematic electronic structure calculation using different exchange-correlation (xc) approaches within spin-density functional theory, which are largely employed to investigate the properties of a variety of coordination complexes. To evaluate the effects of spin states and type of spacer in the bridging ligand on the valence tautomerism between Co2+/3+ and Ru2+/3+, we examine in more detail the case of Creutz-Taube-type ions [(NH3)5Co(py)-X-(py)Ru(NH3)5]5+. Our analysis shows that the stabilization of low- and high-spin states critically depends on the total charge of the complex, type of X-bridged ligand, and employed xc approach to calculate the electron spin density. Importantly, the C60-bridged group may result in a blockage of the valence tautomerism of the Creutz-Taube complex, inducing bistable charge configurations. Overall, our results also show that an adiabatic description in terms of the frontier molecular spin-orbitals for analyzing the distinct spin-charge states of these complexes may dramatically depend on the density-functional description.The sensitive surfaces of many unvarnished 20th century oil paintings are of great concern for conservators and collection keepers. They may show degradation problems such as paint delamination, dripping, and soft and sticky paint and pose challenges for cleaning due to solvent sensitivity. We report for the first time the use of an innovative ambient ionization technique, surface acoustic wave nebulization-mass spectrometry (SAWN-MS), for the identification and characterization of fatty acids, dicarboxylic species and glycerides in water-sensitive modern oil paints. The composition of 10 relevant Winsor and Newton 1964-1965 paint swatches that present different degrees of water sensitivity and two paint samples from a painting by the British artist Francis Bacon were studied. Principal component analysis was used for SAWN-MS data classification. Electrospray ionization (ESI)-MS was used as control method, specifically to compare the obtained ratios of markers of interest by the two ionization techniques. U0126 cost The results obtained by both ESI-MS and SAWN-MS are correlated and discussed in a broader context including the information on the oil media obtained by gas chromatography (GC-MS) and also on the inorganic materials and salts characterized using a combination of methods in previous reports on samples from the same manufacturer. SAWN-MS was found to be a suitable tool for the determination of soluble organic constituents present in the paints. The method provides an indication of the level of oxidation and hydrolysis of the paint film by monitoring specific markers such as free palmitic and stearic acids, azelaic acid, monoacylglycerols, and diacylglycerols. The data showed that a higher level of water sensitivity coupled with a high level of oxidation and hydrolysis is linked to higher dicarboxylic acid, diacyl- and triacylglyceride content and lower levels of short chain fatty acids. The data obtained by SAWN-MS provided a good correlation between the monitored species and the degree of water sensitivity.
