Background MicroRNAs (miRNAs) have been shown to contribute to the initiation and progression of human cancer, including retinoblastoma. However, expression levels and potential roles of miRNAs in retinoblastoma remain largely unknown. In this study, we aimed to identify dysregulated miRNAs and explore their functional roles in the development of retinoblastoma. Material and Methods First, miRNA expression profiling in retinoblastoma tissues was performed via microarray analysis. To evaluate the involvement of miR-214-3p in multi-drug resistance, gain-of-function experiments were employed in vitro and in vivo. Bioinformatics analysis, luciferase reporter assay, qRT-PCR and Western blot were used to investigate the underlying mechanisms. Results Here, we identified 57 up-regulated and 34 down-regulated miRNAs. Among them, miR-214-3p was the most significantly decreased. We found that miR-214-3p level was positively correlated with clinical outcome and chemotherapy response. Overexpression of miR-214-3p significantly sensitized retinoblastoma cells to multiple chemodrugs and promoted cell apoptosis in vitro and in vivo. Further investigations revealed that miR-214-3p directly regulated ABCB1 and XIAP expression through interacting with the 3' untranslated regions (3'UTRs). Pearson correlation analysis showed that miR-214-3p expression in retinoblastoma tissues was negatively correlated with ABCB1 and XIAP expression. We also observed that overexpression of ABCB1 or XIAP partly reversed the chemoresistance inhibition-induced by miR-214-3p overexpression. Conclusion Our data demonstrate that miR-214-3p functions as a tumor suppressor to inhibit the chemoresistance in retinoblastoma, suggesting that miR-214-3p might be potential diagnostic and therapeutic targets for retinoblastoma treatment. © 2020 Yang et al.Background Lymphoepithelioma-like carcinomas (LELCs), especially colorectal cancers (CRCs), are uncommon pathological phenotypes generally associated with poor aggressiveness and a preferable prognosis. However, PD-L1 overexpression in CRCs is associated with poor outcomes. We report a case of moderately differentiated adenocarcinoma with PD-L1 overexpression, an LELC component, and Crohn's-like lymphoid reaction (CLR) presenting with extreme locoregional aggression and complete remission with chemotherapy after noncurative excision. Case presentation A 69-year-old man was referred to our hospital for abdominal fullness and pain. Computed tomography (CT) showed a circumferential tumor in the ascending colon, accompanied by bulky swollen lymph nodes. Under the preoperative diagnosis of T4N2M1 (lymph nodes) ascending colon cancer, we performed a right hemicolectomy; however, paracaval and parailiac vein lymph nodes were abandoned. Pathological findings showed moderate to poorly differentiated adenocarcinoma invcomplete remission was finally achieved with chemotherapy. This type of LELC with CLR could be associated with a good response to chemotherapy and a good prognosis in CRC patients. © 2020 Nagano et al.Objectives Pregnant women are at increased risk of complications from influenza, and infants are at increased risk of pertussis. Our objective was to estimate vaccination coverage with influenza and Tdap vaccines during pregnancy among women with a recent live birth. Methods An opt-in Internet panel survey was conducted from March 28 to April 10, 2018 among pregnant and recently pregnant women. Respondents with a live birth from August 1, 2017 through the date in which the participant completed the survey were included in the analysis. Receipt of influenza vaccination since July 1, 2017 and Tdap vaccination during pregnancy were assessed by sociodemographic characteristics, receipt of a health care provider (HCP) recommendation and/or offer of vaccination, and vaccination-related knowledge, attitudes, and beliefs. Results Less than a third (30.3%) of women with a live birth were unvaccinated during their pregnancy with both Tdap and influenza vaccines. Almost a third (32.8%) of the women reported being vaccinated with both vaccines. The majority (73.0%) of women reported receiving an HCP recommendation for both vaccines, and 54.2% of women were offered both vaccines by an HCP. Reasons for nonvaccination included negative attitudes toward influenza vaccine and lack of awareness about Tdap vaccination during pregnancy. Conclusions Maternal Tdap and influenza vaccinations can prevent morbidity and mortality among infants and their mothers, yet many pregnant women are unvaccinated with either Tdap or influenza vaccines. Clinic-based education, along with interventions, such as standing orders and provider reminders, are strategies to increase maternal vaccination. © The Author(s) 2020.Background Public health concerns over the addictive potential of electronic cigarettes (e-cigs) have heightened in recent years. Brain function during e-cig use could provide an objective measure of the addictive potential of new vaping products to facilitate research; however, there are limited methods for delivering e-cig aerosols during functional magnetic resonance imaging (fMRI). The current study describes the development and feasibility testing of a prototype to deliver up to four different e-cig aerosols during fMRI. Methods Standardized methods were used to test the devices' air flow variability, nicotine yield, and free radical production. MRI scans were run with and without the device present to assess its safety and effects on MRI data quality. Five daily smokers were recruited to assess plasma nicotine absorption from e-liquids containing nicotine concentrations of 8, 11, 16, 24, and 36 mg/ml. Feedback was collected from participants through a semi-structured interview and computerized questionnns, and other e-cig characteristics. © The Author(s) 2020.Hyperuricemia is usually associated with hypertension, diabetes mellitus, metabolic syndrome and chronic kidney disease. Accumulating data from epidemiological studies indicate an association of increased uric acid (UA) with cardiovascular diseases. Possible pathogenic mechanisms include enhancement of oxidative stress and systemic inflammation caused by hyperuricemia. Arterial stiffness may be one of the possible pathways between hyperuricemia and cardiovascular disease, but a clear relationship between increased UA and vascular alterations has not been confirmed. The review summarizes the epidemiological studies investigating the relationship between UA and arterial stiffness and highlights the results of interventional studies evaluating arterial stiffness parameters in patients treated with UA-lowering drugs. © 2020 Albu et al.
