AIM To compare the efficacy of FVIII/VWF complex (Fanhdi) and five different VWF-free FVIII preparations in restoring thrombin generation and activation of thrombin-activatable fibrinolysis inhibitor (TAFI) in haemophilic plasma, with and without inhibitor, and in cell-based models. METHODS Experiments were performed in haemophilic plasma supplemented with inhibitory IgG or in plasma samples obtained from haemophilia A patients without (n = 11) and with inhibitor (n = 12). Thrombin generation was evaluated by calibrated automated thrombography (CAT) under standard conditions, in the presence of activated protein C (APC) or thrombomodulin (TM), and in cell-based models including endothelial cells, either alone or in combination with platelets or tissue factor-expressing blood mononuclear cells. The kinetics of TAFI activation was determined by a two-stage functional assay in the absence and in the presence of APC. RESULTS In haemophilic plasma without inhibitor, Fanhdi enhanced thrombin generation and TAFI activation as well as recombinant (2nd-4th generation) and plasma-derived FVIII preparations devoid of VWF. On the contrary, in plasma with inhibitor, Fanhdi displayed a greater ability to restore thrombin generation and TAFI activation under all tested conditions. find more Notably, in cell-based models including endothelial cells, Fanhdi proved more efficient than all other preparations in improving thrombin generation even in the absence of inhibitor. However when selecting, applying, and evaluating an instrument with a focus on OHRQoL assessment for adults, several methodological considerations are essential to derive valid and meaningful results. METHODS In this review article, criteria for selecting the appropriate OHRQoL instrument, aspects of administering the instrument, and how to evaluate resulting scores of single and multiple assessments are presented and critically assessed. RESULTS Oral disease-generic and dimension-generic instruments capturing the entire construct OHRQoL allow for best comparability of findings across different diseases, settings, and populations, with the Oral Health Impact Profile (OHIP) being the most often used and methodologically best investigated one. It is available in several versions with the 5-item version being the one with the lowest burden for the patient. Responses are given on a 5-point ordinal rating scale, the recommended response scale for dPROMs. A 7-day recall period allows for assessment of short-term effects. Clinically relevant effects of item or instrument order or of administration method on OHIP scores do not seem to be likely. OHIP summary and dimension scores can be compared to norms from general population or different patient populations. Change scores should be set into context with the minimal important difference of the instrument. CONCLUSION OHIP-5 has greatest potential to be used across all settings for assessment and evaluation of OHRQoL in adults. Due to the inherent ability of magnets to provide forces at a distance, the technology of CTCs isolation based on the magnetophoresis mechanism has become a routine methodology. This historical review aims to introduce two principles of magnetic isolation and recent techniques, facilitating research in this field and providing alternatives for researchers in their study of magnetic isolation. Researchers intend to promote effective CTC isolation and analysis as well as active development of next-generation cancer treatment. The first part of this review summarizes the primary principles based on positive and negative magnetophoretic isolation and describes the metrics for isolation performance. The second part presents a detailed overview of the factors that affect the performance of CTC magnetic isolation, including the magnetic field sources, functionalized magnetic nanoparticles, magnetic fluids, and magnetically driven microfluidic systems. © 2020 The Authors. Neben teilweise bereits umgesetzten Lösungen wie Isolationsbereiche oder Clusterquarantäne sollten auch Pläne zur Evakuierung dicht belegter Unterkünfte erwogen werden. Dem Öffentlichen Gesundheitsdienst (ÖGD) kommt im Rahmen der Covid-19-Pandemie eine zentrale Rolle zu. Um einerseits Routineaufgaben des Infektionsschutzes wahrnehmen zu können, andererseits ein effizientes gesundheitliches Management von Flüchtlingsunterkünften zu unterstützen, sollte der ÖGD dringend personell wie materiell gestärkt werden. Wege für einen schnellen, Bundesländer übergreifenden Austausch von Best-Practice-Beispielen zu Covid-19-Maßnahmen, sowie Kapazitäten und kollaborative Strukturen innerhalb des ÖGD für operative Begleitforschung wären wichtig.
