Chronic lymphocytic leukemia (CLL) is an extremely heterogeneous disease. With the advent of oral targeted agents (Tas) the treatment of CLL has undergone a revolution, which has been accompanied by an improvement in patient's survival and quality of life. This paradigm shift also affects the value of prognostic and predictive biomarkers and prognostic models, most of them inherited from the chemoimmunotherapy era but with a different behavior with Tas. This review discusses (i) the role of the most relevant prognostic and predictive biomarkers in the setting of Tas; and (ii) the validity of classic and new scoring systems in the context of Tas. In addition, a critical point of view about predictive biomarkers with special emphasis on 11q deletion, novel resistance mutations, TP53 abnormalities, IGHV mutational status, complex karyotype and NOTCH1 mutations is stated. We also go over prognostic models in early stage CLL such as IPS-E. Finally, we provide an overview of the applicability of the CLL-IPI for patients treated with Tas, as well as the emergence of new models, generated with data from patients treated with Tas.Inflammasome targeting and controlling dysbiosis are promising therapeutic approaches to control ulcerative colitis. This report is the first to investigate the mechanisms underlying the coloprotective effects of rosuvastatin and Lactobacillus and their combined therapy on dextran sodium sulfate (DSS)-induced colitis in high-fat diet (HFD)-fed rats. Our results demonstrate the aggravation of intestinal inflammation as a consequence of an HFD following DSS administration. An association between dyslipidemia, LDL oxidation, CD36 expression, ROS generation, thioredoxin-interacting protein (TXNIP) upregulation, and NLRP3 inflammasome activation was demonstrated by DSS exposure in HFD-fed rats. We demonstrated that rosuvastatin/Lactobacillus significantly suppressed the DSS/HFD-induced increase in colon weight/length ratio, DAI, MDI, and myeloperoxidase, as well as corrected dysbiosis and improved histological characteristics. Additionally, caspase-1 activity and IL-1β-driven pyroptotic activity was significantly reduced. Rosuvastatin/Lactobacillus showed prominent anti-inflammatory effects as revealed by the IL-10/IL-12 ratio and the levels of TNF-α and IL-6. These latter effects may be attributed to the inhibition of phosphorylation-induced activation of NF-κB and a concomitant reduction in the expression of NLRP3, pro-IL-1β, and pro-IL-18. Furthermore, rosuvastatin/Lactobacillus reduced Ox-LDL-induced TXNIP and attenuated the inflammatory response by inhibiting NLRP3 inflammasome assembly. To conclude, rosuvastatin/Lactobacillus offers a safe and effective strategy for the management of ulcerative colitis.Edge computing is intended to process events that occur at the endpoint of the Internet of Things (IoT) network quickly and intelligently. Edge regions must be organized effectively to facilitate cooperation so that the intention of edge computing can be realized. However, inevitably, many human and material resources are required in the process of arranging things in the edge area to confirm the appropriateness of the thing operation. To address this problem, we proposed a simulator that created a virtual space for edge computing and provided an interactive role and effective organization for edge things. The proposed simulator was aimed at Raspberry Pi as the physical hardware target. To prove the accuracy of the proposed simulator, the similarity between the proposed simulator and the physical target Raspberry Pi was evaluated based on three metrics while executing several applications. In the experiment, several edge-computing service applications were performed in various cluster architecture types formed by the proposed simulator. To support effective resource usage and fast real-time response for edge computing, the proposed simulator identified a suitable number of things in forming the edge cluster.Pancreatic cancer remains as one of the most aggressive cancer types. In the absence of reliable biomarkers for its early detection and more effective therapeutic interventions, pancreatic cancer is projected to become the second leading cause of cancer death in the Western world in the next decade. Therefore, it is essential to discover novel therapeutic targets and to develop more effective and pancreatic cancer-specific therapeutic agents. To date, 45 monoclonal antibodies (mAbs) have been approved for the treatment of patients with a wide range of cancers; however, none has yet been approved for pancreatic cancer. In this comprehensive review, we discuss the FDA approved anticancer mAb-based drugs, the results of preclinical studies and clinical trials with mAbs in pancreatic cancer and the factors contributing to the poor response to antibody therapy (e.g. tumour heterogeneity, desmoplastic stroma). MAb technology is an excellent tool for studying the complex biology of pancreatic cancer, to discover novel therapeutic targets and to develop various forms of antibody-based therapeutic agents and companion diagnostic tests for the selection of patients who are more likely to benefit from such therapy. These should result in the approval and routine use of antibody-based agents for the treatment of pancreatic cancer patients in the future.Laser thermal forming is an application of laser heating without any intentional use of external forces. Force-assisted laser bending and laser-assisted bending are hybrid techniques, which combine the use of external forces and local heating to increase the effectiveness of forming. p38 MAPK activity A quantitative description of bending deformation induced by concurrent laser heating and mechanical loading is proposed in this study. Mechanical loading is expressed by the bending moment while the curvature is used to describe the resulting deformation. The contribution of a relatively less known mechanism of laser thermal bending in the hybrid process is identified. The mechanism is able to produce the so-called convex deformation, i.e., bending away from the incident laser beam. Experimental and numerical analysis is performed with thin-walled beams made of Inconel 718 nickel-based superalloy in the factory-annealed state. The Johnson-Cook constitutive material model is used in numerical simulations validated by experimental results.
