The food industry requires rapid and simple detection methods for preventing harm from pathogenic bacteria. Until now, various technologies used to detect foodborne bacteria were time-consuming and laborious. Therefore, we have developed an automated immunomagnetic separation combined with a colorimetric assay for the rapid detection of E. coli O157H7 in food samples. The colorimetric detection method using enzymatic reaction is fascinating because of its simplicity and rapidity and does not need sophisticated devices. Moreover, the proposed procedures for the detection of bacteria in food take less than 3 h including pre-enrichment, separation and detection steps. First, target-specific immunomagnetic beads were introduced to contaminated milk in a pre-enrichment step. Second, the pre-enriched sample solution containing target bacteria bound on immunomagnetic beads was injected into an automated pretreatment system. Subsequently, the immunomagnetic beads along with target bacteria were separated and concentrated into a recovery tube. Finally, released β-galactosidase from E. coli O157H7 after lysis was reacted with chlorophenol red β-galactopyranoside (CPRG) used as a substrate and the colorimetric change of CPRG was determined by absorbance measuring or the naked eye. By the proposed approach in this study, we could detect 3 × 102 CFU/mL of E. coli O157H7 from a milk sample within 3 h.Based on the theory of planned behavior, this research examines the influence of different types of information on the behavioral intentions of college students in the context of perceived behavioral control (perceived self-efficacy and perceptual control) as mediating variables. The results showed that (1) Different types of information intervention factors have different effects on perceptual self-efficacy and perceptual control; the influence degree of economic cost has the strongest effect, followed by group pressure, while the influence degree of publicity and education has the weakest effect. However, policy intervention has no statistically significant effect on both of them (perceived self-efficacy and perceptual control). (2) Two variables, perceived self-efficacy and perceptual control, serve as mediators between information intervention factors and energy-saving behavior intention. (3) Individual characteristic factors have significant moderating effects on each path in the model of information intervention-perceived behavior control-intention. Finally, suggestions are made on how to encourage college students to more effectively save energy.An Ag/AgCl electrode used as a corrosion sensor in a reinforced concrete structure isconsidered as having good application prospect. However, its performance under complexconditions, such as dry-wet cycle condition, is not affirmed. In the current study, the performanceof Ag/AgCl as chloride selective electrode in mortar exposed to dry-wet cycle condition wasinvestigated. A simple Ag/AgCl electrode was prepared and fabricated by electrochemicalanodization. These Ag/AgCl electrodes were embedded into a mortar specimen with temperaturesensors, humidity sensors and anode ladder monitoring system (ALS). After 28 d curing time, theupper surface of mortar specimen was wetted (with 5% NaCl solution) and dried regularly. Theobtained results indicate that Ag/AgCl electrode responds to the ingress of chloride ion, sensitively.The chloride ion concentration variation can be reflected by the potential trend. Furthermore, thebalance potential of Ag/AgCl electrodes is influenced by dry-wet cycles. Compared with ALS, itdemonstrates that Ag/AgCl electrodes are more sensitive to chloride. The research provides the keyelement for the specific application of Ag/AgCl electrode for corrosion monitoring in the future.The objective of this study was to characterize oligofructose-induced acute rumen lactic acidosis and its consequences in zebu cattle. We used 29 Nellore heifers which were submitted to experimental induction of laminitis by oligofructose excess. During the induction period, the animals underwent clinical examination, including laminitis diagnosis (hoof pressure testing and locomotion score) and blood and ruminal fluid sampling every six hours (over the initial 24 h) and every 12 h (up to 72 h), after the highest dose. Almost half of the animals (48.1%) required treatment with bicarbonate and saline to correct metabolic acidosis and dehydration. Due to this treatment, the animals were analyzed in treated (n = 13) and non-treated (n = 14) groups. The induction model promoted marked reduction in rumen pH, rumen anaerobiosis, carbon dioxide pressure, and increase in rumen lactate, blood osmolarity, and cortisol concentration. The animals treated had lower values of rumen pH and marked dehydration, evidenced by the increase in globular volume and serum urea. The clinical condition caused by excess oligofructose is severe, with the differential of the appearance of ephemeral fever and respiratory compensation against systemic acidosis, in addition to the frequent appearance of laminitis.Gastrocnemius medialis (GM) architecture and ankle angle were compared between flexibility trained (n = 10) and not trained (n = 6) female athletes, aged 8-10 years. Ankle angle, fascicle length, pennation angle and muscle thickness were measured at the mid-belly and the distal part of GM, at rest and at the end of one min of static stretching. Flexibility trained (FT) and not trained athletes (FNT) had similar fascicle length at the medial (4.19 ± 0.37 vs. 4.24 ± 0.54 cm, respectively, p = 0.841) and the distal part of GM (4.25 ± 0.35 vs. 4.18 ± 0.65 cm, respectively, p = 0.780), similar pennation angles, and muscle thickness (p > 0.216), and larger ankle angle at rest (120.9 ± 4.2 vs. 110.9 ± 5.8°, respectively, p = 0.001). During stretching, FT displayed greater fascicle elongation compared to FNT at the medial (+1.67 ± 0.37 vs. +1.28 ± 0.22 cm, respectively, p = 0.048) and the distal part (+1.84 ± 0.67 vs. +0.97 ± 0.97 cm, respectively, p = 0.013), larger change in joint angle and muscle tendon junction displacement (MTJ) (p 0.053). Ankle dorsiflexion angle significantly correlated with fascicle elongation at the distal part of GM (r = -0.638, p less then 0.01) and MTJ displacement (r = -0.610, p less then 0.05). selleck compound Collectively, FT had greater fascicle elongation at the medial and distal part of GM and greater MTJ displacement during stretching than FNT of similar age.
