Glycoside hydrolases and the recently discovered lytic polysaccharide monooxygenase for their use in biomass degradation to obtain biofuels or valued chemical entities. Glycosyltransferases or engineered glycosidases and phosphorylases for the synthesis of carbohydrates and glycosylated products. Quantum mechanics-molecular mechanics (QM/MM) methods are highly contributing to establish their different chemical reaction mechanisms. Other computational methods are also used to study enzyme conformational changes, ligand pathways, and processivity, e.g. Fenretinide chemical structure for processive glycosidases like cellobiohydrolases. There is still a long road to travel to fully understand the role of conformational dynamics in enzyme activity and also to disclose the variety of reaction mechanisms these enzymes employ. Additionally, computational tools for enzyme engineering are beginning to be applied to evaluate substrate specificity or aid in the design of new biocatalysts with increased thermostability or tailored activity, a growing field where molecular modeling is finding its way.
Immune checkpoint inhibition (ICI) triggers immune-related adverse events (irAEs). The relevance of lipase elevation remains unclear.

Skin cancer patients with newly detected serum lipase elevation (at least twofold upper normal limit) or newly diagnosed type I diabetes mellitus upon ICI therapy were retrospectively collected at 14 German skin cancer centres.

We identified 68 patients with lipase elevation occurring after a median time of 19 (range 1-181) weeks on ICI, 15 (22%) thereof had symptoms consistent with pancreatitis. Forty-seven patients (73%) had other irAE, mainly colitis. Discontinuation (n=24, 35%) or interruption (n=26, 38%) of ICI resulted in decrease of lipaseafter reinduction of ICI lipase levels increased again in 12 of 24 patients. In 18 patients (27%), ICI was continued unchanged, and in 12 (67%) of them, lipase levels normalised. Twenty-two patients were identified with newly diagnosed type I diabetes mellitus related to ICI, and 12 (55%) thereof had also lipase elevation mainly shortly before or after the diagnosis of diabetes. Fourteen (64%) patients had other irAE, mainly thyroiditis. Irrespective of lipase elevation, patients frequently showed a rapid onset with ketoacidosis, decreased c-peptide, and strongly increased blood glucose levels.

Increased serum lipase during ICI is often not associated with pancreatitis but with other irAE as possible cause. Therefore, it might be sufficient to regularly monitor blood glucose levels and perform further workup only in case of signs or symptoms of pancreatitis and/or exocrine pancreas insufficiency.

A lignin-degrading bacterial consortium LDC, which can efficiently degrade lignin of reed, was screened in the early stage of our laboratory work. In present study, 7-day incubation of LDC can degrade rice straw lignin by 31.18% in mineral salt medium. The communities' structure of different incubation phases varied greatly, in which high abundance (44.78%) of Anaerocolumna was first found. The expression levels of lignin degradation enzyme class II peroxidase (AA2), vanillyl alcohol oxidase (AA4) and 1,4-benzoquinone reductase (AA6) during peak phase (48 h) were significantly up-regulated than initial phase (24 h), increasing by 112%, 165% and 67%, respectively, and 42.86% AA2 was from Thaurea; 100% AA4 was from Clostridium; 62.5% AA6 was from Pseudomonas. These provide microbial resources and data support for the industrialization of rice straw bio-pulping.A microalgal biofilm-attached-system is an alternative cultivation method, that offers potential advantages of improved biomass productivity, efficient harvesting, and water saving. These biofilm systems have been widely tested and utilized for microalgal biomass production and wastewater treatment. This research a microalgal growth model for the biofilm attached culture system has been developed and experimentally validated, both, in single and arrayed biofilm systems. It has been shown that the model has the capability to accurately describe microalgae growth. Moreover, via the model simulation, it was observed that system structural parameters, light dilution rate, and light intensity significantly affected the culture performance. The limitations, and improvement aspects of the model, are also discussed in this study. To our knowledge, this is the first time that a mathematical model for an arrayed-biofilm-attached-system has been developed and validated. This model will certainly be helpful in the design, improvement, optimization, and evaluation of the biofilm-attached-systems.With Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) and fluorescence spectroscopy, this study investigated the transformation of dissolved organic matter (DOM) and nitrogen (DON) during the widely-applied anaerobic/anoxic/oxic (A2O) processes to provide molecular insights into the removal, generation, and reduction of DOM/DON species in different biological treatment units. Results indicated that the anaerobic process decomposed the macromolecules of influent DOM/DON and decreased their mass. The anoxic process denitrified DON and generated DOM, as indicated by the decreased molecule number of CHON and CHONS and the increased CHO and CHOS species, as well as the increased overall DOM intensities. DOM mineralization and ammonia nitrogen-DON conversion occurred in the oxic process. Aromaticity and unsaturation degree increased slightly after the A2O processes, which was correlated with the relative abundance of Proteobacteria (positively) and Bacteroidetes (negatively). The results have strong implications to the understanding of DOM/DON dynamics in wastewater treatment plants.
