We previously reported that an EAV-HP inserted mutation (IM+) in the 5' flanking region of SLCO1B3 was the causative mutation of chicken blue eggs, and a further research showed that IM+ significantly reduced the expression of SLCO1B3 in liver. Herein, we confirmed a cholate response element (IR-1) played an important role in activating SLCO1B3 and in vitro experiments showed that the activation of IR-1 can be significantly reduced by the EAV-HP IM+ . We performed transcriptome and proteomic analysis using the same set of IM+ and IM- liver tissues from Yimeng hens (a Chinese indigenous breed) to study the effect of SLCO1B3 and OATP1B3 expression reduction on chicken liver function. The results showed that common differential expression pathways were screened out from both transcriptome and proteome, in which fatty acid metabolism and drug metabolism-cytochrome P450 were significantly enriched in the KEGG analysis. The lipid-related metabolism was weakened in IM+ group, which was validated by serum biochemical assay. We unexpectedly found that EAV-HP fragment was highly expressed in the liver of the IM+ chickens. We cloned the EAV-HP full-length transcript and obtained the complete open reading frame. It is worth noting that there was some immune related differential expressed genes, such as NFKBIZ, NFKBIA, and IL1RL1, which were higher expressed in the IM+ group, which may due to the high expression of EAV-HP. Our study showed that EAV-HP IM+ reduced the expression of SLCO1B3 in liver, resulting in the decrease of fatty metabolism and exogenous substance transport capacity. The mutation itself also expressed in the liver and may be involved in the immune process. The mechanism needs further study.This study deals with the determination of new natural fibers extracted from the Corypha taliera fruit (CTF) and its characteristics were reported for the potential alternative of harmful synthetic fiber. The physical, chemical, mechanical, thermal, and morphological characteristics were investigated for CTF fibers. X-ray diffraction and chemical composition characterization ensured a higher amount of cellulose (55.1 wt%) content and crystallinity (62.5%) in the CTF fiber. The FTIR analysis ensured the different functional groups of cellulose, hemicellulose, and lignin present in the fiber. The Scherrer's equation was used to determine crystallite size 1.45 nm. The mean diameter, specific density, and linear density of the CTF fiber were found (average) 131 μm, 0.86 g/cc, and 43 Tex, respectively. The maximum tensile strength was obtained 53.55 MPa for GL 20 mm and Young's modulus 572.21 MPa for GL 30 mm. The required energy at break was recorded during the tensile strength experiment from the tensile strength tester and the average values for GL 20 mm and GL 30 mm are 0.05381 J and 0.08968 J, respectively. The thermal analysis ensured the thermal sustainability of CTF fiber up to 230 °C. Entirely the aforementioned outcomes ensured that the new CTF fiber is the expected reinforcement to the fiber-reinforced composite materials.Samples in biobanks are generally preserved by formalin-fixation and paraffin-embedding (FFPE) and/or optimal cutting temperature compound (OCT)-embedding and subsequently frozen. Mass spectrometry (MS)-based analysis of these samples is now available via developed protocols, however, the differences in results with respect to preservation methods needs further investigation. Here we use bladder urothelial carcinoma tissue of two different tumor stages (Ta/T1-non-muscle invasive bladder cancer (NMIBC), and T2/T3-muscle invasive bladder cancer (MIBC)) which, upon sampling, were divided and preserved by FFPE and OCT. Samples were parallel processed from the two methods and proteins were analyzed with label-free quantitative MS. Over 700 and 1200 proteins were quantified in FFPE and OCT samples, respectively. Multivariate analysis indicates that the preservation method is the main source of variation, but also tumors of different stages could be differentiated. Proteins involved in mitochondrial function were overrepresented in OCT data but missing in the FFPE data, indicating that these proteins are not well preserved by FFPE.
