δ-opioid receptor (DOPr) agonists have analgesic efficacy in chronic pain models but development of tolerance limits their use for long-term pain management. Although agonist potential for inducing acute analgesic tolerance has been associated with distinct patterns of DOPr internalization, the association between trafficking and chronic tolerance remains ill-defined. Moreover, endosomal endothelin-converting enzyme 2 (ECE2) blocker 663444 prevented DOPr recycling by deltorphin II and TIPP and precipitated tolerance by these ligands. Journal of Cellular and Molecular Medicine published by Foundation for Cellular and Molecular Medicine and John Wiley & Sons Ltd.BACKGROUND & AIMS Whether nonalcoholic fatty liver disease (NAFLD) is associated with risk of incident atrial fibrillation (AF) independent of established cardiovascular risk factors remains controversial. We aimed to provide a quantitative estimate of the association between NAFLD and risk of AF after adjustment for cardiometabolic risk factors. METHODS In this study, we searched PubMed and Embase for studies published from database inception until January 31, 2020. Cohort studies reported adjusted relative risks (RRs) and 95% confidence intervals (CIs) for AF of NAFLD compared with non-NAFLD were included for analysis. RESULTS A total of 6 cohort studies were included, comprising 614,673 individuals for analysis. The median follow-up duration was 10.0 years with 7,271 cases of incident AF. Compared with non-NAFLD, minimally adjusted models without adjustment for cardiometabolic risk factors showed that NAFLD was associated with increased risk of AF (RR 1.65, 95% CI 1.23-2.20, I2 = 63.0%). After adjustment for multiple cardiometabolic risk factors, the association between NAFLD and risk of AF was still higher than that in non-NAFLD (RR 1.19, 95% CI 1.04-1·31, I2 = 54.0%). There was significant heterogeneity for the risk of AF between minimally and maximally adjusted models (I2 = 77.1%, P for heterogeneity = 0.04). Compared with non-NAFLD, the absolute risk increase in NAFLD for AF was 1.3 (95% CI 0.5-2.1) per 1000 person-years. CONCLUSIONS NAFLD is associated with increased risk of incident AF. However, the biological function and molecular mechanism of ASH2L in endometrial cancer (ECa) is largely unknown. Endometrial cancer is one of the common female malignant tumors and the incidence is on the rise. Estrogen-ERα signaling as an oncogenic pathway, plays a crucial role in endometrial carcinogenesis. Thus, further exploring the molecular mechanism around ERα-mediated gene transcription in ECa would be helpful to understand the tumor development and to find a new therapeutic target for ECa. Here, our study demonstrated that ASH2L was highly expressed in ECa samples, and the higher expression of ASH2L was positively correlated with the poor prognosis. Moreover, we identified that ASH2L associated with ERα and knockdown of ASH2L resulted in decreased expression of a subset of the estrogen-induced target genes, including paired box 2 (PAX2), an oncogenic gene in ECa. ASH2L was recruited to cis-regulatory elements at PAX2, thereby altering histone H3K4me3 and H3K27me3 levels, to enhance ERα-mediated transactivation. Finally, depletion of ASH2L suppressed endometrial cancer cell proliferation and migration. Our findings suggest that ASH2L participates in promotion of ECa progression, if not all, at least partially via upregulation of PAX2 transcription. This article is protected by copyright.
