Some clinical trials have shown that targetable driver gene mutations, such as EGFR, ALK or BRAF, have rarely been detected in PPC patients, but the incidence of MET exon 14 mutations is more frequent. For these patients with driver gene mutations, corresponding molecular targeted therapy may be valid. In addition, limited cases have suggested that immunotherapy may be effective for PPC without sensitising EGFR or ALK alterations, but the efficacy in patients with other driver mutations remains unclear. Herein, we reported two PPC patients with different targetable gene mutations who both responded dramatically to the PD-1 inhibitor camrelizumab combined with the oral anti-angiogenic drug anlotinib one harbouring a BRAF V600E mutation with positive PD-L1 expression, few tumour-infiltrating lymphocytes (TILs) and abundant tumour blood vessels; and the other exhibiting a MET exon 14 skipping mutation with PD-L1 overexpression, scattered TILs and abundant tumour blood vessels. Our findings suggest that PD-1 inhibitor combined with anlotinib may be a potential treatment for PPC patients, and abundant tumour vessels should be investigated as a possible therapeutic biomarker.
Monotherapy with pembrolizumab is the preferred first-line treatment for metastatic non-small cell lung cancer with programmed death-ligand 1 (PD-L1) expression ≥50 %, without targetable oncogenic drivers. Although targeted therapies are in development for patients with specific Kirsten rat sarcoma (KRAS) mutations, these are not available in daily care yet. It is not clear whether there is a difference in survival on first-line pembrolizumab for patients with a high PD-L1 status with or without a KRAS mutation. We aim to compare this survival based on real-world data.

This is a real-world retrospective population-based study using data from the Netherlands Cancer Registry. We selected patients with stage IV lung adenocarcinoma with PD-L1 expression ≥50 % diagnosed between January 2017 and December 2018, treated with first-line pembrolizumab. Patients with EGFR mutations, ALK translocations or ROS1 rearrangements were excluded. LY3039478 The primary outcome parameter was overall survival.

388 (57 %) of 595 patients had a KRAS mutation. KRAS was seen more frequently in women than in men (65 % versus 49 % respectively, p < 0.001). The median overall survival was 19.2 months versus 16.8 months for patients with and without KRAS mutation, respectively (p = 0.86). Multivariable analysis revealed WHO performance score, number of organs with metastases and PD-L1 percentage as independent prognostic factors. KRAS mutation status had no prognostic influence (hazard ratio = 1.03, 95 % CI 0.83-1.29).

A financing model, that is economically feasible and accepted by all stakeholders, has been a limitation for the implementation of pollution control measures. A case-study on a 279-km Portuguese Highway is presented as a basis for a co-financing model. Runoff pollution load was estimated for quality indicators (TSS, COD, Zn, Cu, Pb), and the total cost of infiltration trenches, sand filters, bioretention filters, wet basins, dry basins and constructed wetlands systems was computed for four catchment scenarios. The effect of the equivalent catchment size and system type on the total cost was evaluated. The users 'Willingness to Pay' (defined as pay-per-user and availability to participate actively and financially) was assessed through a survey (1192 responses). A proposed co-financing model suggests that citizens will participate up to 36.8% of the constructed wetlands cost. Amended BRDAs under natural conditions suffer from long-term erosion processes. Nevertheless, the effect of erosion on amendment efficacy is rarely assessed. In this study, by integrating the geochemical modelling of PHREEQC and column leaching experiments, the dissolution of alkaline solids in bauxite residue (BR) and gypsum amendment, as well as their environmental behaviors, were determined through a 1-year simulated rainfall leaching experiment. The PHREEQC simulation results demonstrated that Na+ ion strength, CO2 partial pressure and rainfall, all affected the saturation index (SI) of calcite significantly and accelerated its corrosion, leading to the dissolution of gypsum and calcite in a relatively stable state. However, Na+ ion strength and rainfall significantly acted on the SI of gypsum, which lead to loss of Ca2+ and reduction of alkaline stability. In addition to the effects of Na+ and Ca2+ on the saturation concentration of gypsum and calcite solution, Na+ and Ca2+ also exhibited significant effects on the equilibrium of chemical species reactions.
