Further work defining what number and which combination of risk variables works best for predicting risk of dementia in LMICs is needed. BACKGROUND The reduction by a third of premature non-communicable disease (NCD) mortality by 2030 is the ambitious target of Sustainable Development Goal (SDG) 3.4. However, the indicator is narrowly defined, including only four major NCDs (cardiovascular diseases, cancer, diabetes, and chronic respiratory diseases) and only for people aged 30-70 years. This study focuses on premature avertable mortality from NCDs-premature deaths caused by NCDs that could be prevented through effective public policies and health interventions or amenable to high-quality health care-to assess trends at global, regional, and national levels using estimates from the Global Burden of Disease, Injuries, and Risk Factors Study (GBD) 2017. Epacadostat METHODS We reviewed existing lists of NCD causes of death that are either preventable through public health policies and interventions or amenable to health care to create a list of avertable NCD causes of death, which was mapped to the GBD cause list. We estimated age-standardised years of life l and regional reduction in premature avertable mortality from NCDs, whereas premature mortality from substance use disorders, chronic kidney disease and acute glomerulonephritis, and diabetes have been increasing. INTERPRETATION Worldwide, there has been a substantial reduction in premature avertable mortality from NCDs, but progress has been uneven across populations. Countries vary substantially in current levels and trends and, hence, the extent to which they are on track to achieve SDG 3.4. By accounting for premature avertable mortality while avoiding arbitrary age cutoffs, premature avertable mortality from NCDs is a robust, comprehensive, and actionable indicator for quantifying and monitoring global and national progress towards NCD prevention and control. Mutations in Shank3 are strongly associated with autism spectrum disorders and neural circuit changes in several brain areas, but the cellular mechanisms that underlie these defects are not understood. Homeostatic forms of plasticity allow central circuits to maintain stable function during experience-dependent development, leading us to ask whether loss of Shank3 might impair homeostatic plasticity and circuit-level compensation to perturbations. We found that Shank3 loss in vitro abolished synaptic scaling and intrinsic homeostatic plasticity, deficits that could be rescued by treatment with lithium. Further, Shank3 knockout severely compromised the in vivo ability of visual cortical circuits to recover from perturbations to sensory drive. Finally, lithium treatment ameliorated a repetitive self-grooming phenotype in Shank3 knockout mice. These findings demonstrate that Shank3 loss severely impairs the ability of central circuits to harness homeostatic mechanisms to compensate for perturbations in drive, which, in turn, may render them more vulnerable to such perturbations. Evidence suggests interplay among the three major risk factors for Alzheimer's disease (AD) age, APOE genotype, and sex. Here, we present comprehensive datasets and analyses of brain transcriptomes and blood metabolomes from human apoE2-, apoE3-, and apoE4-targeted replacement mice across young, middle, and old ages with both sexes. We found that age had the greatest impact on brain transcriptomes highlighted by an immune module led by Trem2 and Tyrobp, whereas APOE4 was associated with upregulation of multiple Serpina3 genes. Importantly, these networks and gene expression changes were mostly conserved in human brains. Finally, we observed a significant interaction between age, APOE genotype, and sex on unfolded protein response pathway. In the periphery, APOE2 drove distinct blood metabolome profile highlighted by the upregulation of lipid metabolites. Our work identifies unique and interactive molecular pathways underlying AD risk factors providing valuable resources for discovery and validation research in model systems and humans. INTRODUCTION Mathematics anxiety (MA) is an important risk factor hindering the development of confidence and capability in mathematics and participation in the science, technology, engineering, and mathematics workforce. The aim of the present study is to further our understanding of these relations in adolescence by adopting a threefold approach. First, we adopted a longitudinal design to clarify the temporal order in the developmental relations between (a) MA and mathematics achievement and (b) MA and mathematics self-perceived ability. Second, we investigated whether the developmental relations between MA and mathematics achievement/self-perceived ability differed between boys and girls. Finally, we explored the domain-specificity of MA by examining its role in foreign language (L2) learning. METHODS Data were collected from 1043 Italian high school students. Students reported their anxiety, self-perceived ability, and school achievement in mathematics and L2 over two separate waves, one semester apart. RESULTS Using multi-group cross-lagged panel analyses, we found that (a) mathematics achievement predicted MA longitudinally, whereas MA did not predict subsequent mathematics achievement; (b) there was a negative reciprocal relation between MA and mathematics self-perceived ability in male, but not female students; and (c) there were longitudinal relations between MA and L2 achievement and self-perceived ability above and beyond L2 anxiety. CONCLUSIONS These findings support the deficit view of the developmental relation between MA and mathematics achievement, highlight high school male students as a vulnerable group evincing vicious transactions between high anxiety and low self-efficacy in mathematics, and reveal the importance of internal cross-domain comparison processes in MA development.
