The timing of germination has long been recognized as a key seedling survival strategy for plants in highly variable alpine environments. Seed dormancy and germination mechanisms are important factors that determining the timing of germination. To gain an understanding of how these mechanisms help to synchronize the germination event to the beginning of the growing season in two of the most popular Primula species (P. secundiflora and P. sikkimensis) in the Hengduan Mountains, Southwest China, we explored their seed dormancy and germination characteristics in the laboratory and their soil seed bank type in the field. Germination was first tested using fresh seeds at two alternating temperatures (15/5 and 25/15°C) and five constant temperatures (5, 10, 15, 20, and 25°C) in light and dark, and again after dry after-ripening at room temperature for 6 months. Germination tests were also conducted at a range of temperatures (5-30, 25/15, and 15/5°C) in light and dark for seeds dry cold stored at 4°C for 4 years, aurial, showing that the seeds of these two species could cycle back to dormancy if the conditions were unfavorable during spring. In this study, fresh seeds of the two Primula species exhibited type 3 non-deep physiological dormancy and required light for germination. After dormancy release, they had a low thermal requirement for germination control, as well as rapid seed germination in spring and at/near the soil surface from the soil seed bank. Such dormancy and germination mechanisms reflect a germination strategy of these two Primula species, adapted to the same alpine environments.[This corrects the article DOI 10.3389/fimmu.2021.639902.].DOCK8 deficiency is a combined immunodeficiency due to biallelic variants in dedicator of cytokinesis 8 (DOCK8) gene. The disease has a wide clinical spectrum encompassing recurrent infections (candidiasis, viral and bacterial infections), virally driven malignancies and immune dysregulatory features, including autoimmune (cytopenia and vasculitis) as well as allergic disorders (eczema, asthma, and food allergy). Hypomorphic function and somatic reversion of DOCK8 has been reported to result in incomplete phenotype without IgE overproduction. Here we describe a case of DOCK8 deficiency in a 8-year-old Caucasian girl. The patient's disease was initially classified as autoimmune thrombocytopenia, which then evolved toward a combined immunodeficiency phenotype with recurrent infections, persistent EBV infection and lymphoproliferation. Two novel variants (one deletion and one premature stop codon) were characterized, resulting in markedly reduced, but not absent, DOCK8 expression. Somatic reversion of the DOCK8 deletion was identified in T cells. Hypomorphic function and somatic reversion were associated with restricted T cell repertoire, decreased STAT5 phosphorylation and impaired immune synapse functioning in T cells. Although the patient presented with incomplete phenotype (absence of markedly increase IgE and eosinophil count), sclerosing cholangitis was incidentally detected, thus indicating that hypomorphic function and somatic reversion of DOCK8 may delay disease progression but do not necessarily prevent from severe complications.Self- and non-self ligand discrimination is a core principle underlying T cell-mediated immunity. A growing number of studies have highlighted the relationship between neoantigens and T cells' recognition of cancer cells. Vaccines developed against neoantigens are now being used in clinical trials in various solid tumors. In this review, we summarized the latest advances in the classification of immunotherapy and the process of classification, identification and synthesis of tumor-specific neoantigens, as well as their role in current cancer immunotherapy. Finally, the application prospects and existing problems of neoantigens were discussed.The tumor microenvironment (TME) plays a crucial role in cancer progression and recent evidence has clarified its clinical significance in predicting outcomes and efficacy. However, there are no studies on the systematic analysis of TME characteristics in bladder cancer. In this study, we comprehensively evaluated the TME invasion pattern of bladder cancer in 1,889 patients, defined three different TME phenotypes, and found that different subtypes were associated with the clinical prognosis and pathological characteristics of bladder cancer. We further explored the signaling pathways, cancer-immunity cycle, copy number, and somatic mutation differences among the different subtypes and used the principal component analysis algorithm to calculate the immune cell (IC) score, a tool for comprehensive evaluation of TME. Univariate and multivariate Cox regression analyses showed that ICscore is a reliable and independent prognostic biomarker. In addition, the use of anti-programmed death-ligand (PD-L1) treatment cohort, receiver operating characteristic (ROC) curve, Tumor Immune Dysfunction and Exclusion (TIDE), Subnetwork Mappings in Alignment of Pathways (SubMAP), and other algorithms confirmed that ICscore is a reliable prognostic biomarker for immune checkpoint inhibitor response.
